Studies of the Chitosan Microspheres Prepared as Affinity Chromatography Carrier and used for Purifying Thrombin from Bovine Blood Plasma

In the preparation of chitosan microspheres,the concentration of chitosan and connector are the important factors for the quality and function of chitosan microspheres.In the present study of the preparation of chitosan microspheres,the carrier of affinity chromatography of bovine thrombin,it is proved that 1.5% of the chitosan concentration and higher connector concentration is good for the output of chitosan microspheres.Actived by epichlorohydrin and connected with heparin,the microspheres of chitosan can be used to seperate and purify the bovine thrombin and the absorption reaches 5800 IU per gram dry product.After advanced purification,the recovery rate reaches 84%.It is also proved that chitosan unactivated and uncombined with heparin has no specific absorption with bovine thrombin.
