The objective of this work was to investigate the effects of dietary supplementation of purified lycopene (LYC) or a source of LYC as in tomato powder (TP) on live hen performance, egg yolk cholesterol concentrations as well as the gene expressions related to mechanism and regulation of cholesterol, namely Niemann-Pick C1-like 1 protein (NPC1L1), acyl-CoA: cholesterol acyltransferase 2 (ACAT2), microsomal triacylglycerol transport protein (MTP), ATP binding cassette transporters sub-family G member 5 and 8 (ABCG5/8), ATP citrate lyase (ACLY), liver X receptor alpha (LXRα) and sterol regulatory element-binding proteins (SREBP1c). A total of 150 20-week-old white laying hens were divided into one of 3 treatments, with 10 replicates of 5 hens per cage, totaling 50 hens per treatment. The birds were fed either a basal diet (Control) or basal diet supplemented with 20 mg purified lycopene/kg diet (LYC) or an equal amount of lycopene-containing tomato powder (TP) for 12 weeks. Feed intake, egg production and feed conversion ratio remained similar among treatments (P ≥ 0.27). However, egg weights from hens fed a diet supplemented with lycopene particularly the purified form were greater (P = 0.036). Supplementing lycopene either purified or as in TP increased the concentrations of serum and egg yolk lycopene but decreased serum and egg yolk cholesterol concentrations (P < 0.001). Supplementing LYC purified or as in TP decreased the gene expression of intestinal NPC1L1, MTP, ACAT2, hepatic SREBP1c, ACLY, and LXRα but increased hepatic ABCG5 and ABCG8 (P < 0.001). The gene expressions were more pronounced with TP supplements compared with that of the LYC supplements. The results of the present work revealed that egg yolk cholesterol metabolism is downregulated through decreased activities of intestinal NPC1L1, MTP, ACAT2, hepatic SREBP1c, ACLY, and LXRα but upregulated with increased hepatic activities of ABCG5 and ABCG8. RJX is an Intravenous (IV) formulation of known physiologically compatible compounds that is being developed for more effective supportive therapy of patients with sepsis, including COVID-19 patients with viral sepsis and Acute Respiratory Distress Syndrome (ARDS). The RJX formulation is a solution of buffered acid products, electrolyte components, and vitamins, including ascorbic acid, cyanocobalamin, thiamine hydrochloride, riboflavin 5' phosphate, niacinamide, pyridoxine hydrochloride, and calcium d-pantothenate, and magnesium sulfate heptahydrate, a mineral with a negative oxidation-reduction potential. The components of RJX exhibited promising activity in clinical studies involving ARDS patients and/or non-clinical studies in animal models of ARDS. The published data from these clinical and non-clinical studies provided the medical-scientific rationale for our clinical development strategy for RJX and a clinical study in COVID-19 patients.
Abstract The study tested new oral plant‐based formulations (F) on rats with monosodium iodoacetate (MIA)‐induced osteoarthritis, measuring inflammation, antioxidant levels, paw size, stride, and analyzing knee joint images. Fifty‐six female Sprague Dawley rats were allocated into 8 groups: (1) Control, (2) MIA (OA induced with MIA), (3) MIA + F1 [curcuminoids+gingerols+acetyl‐11‐keto‐β boswellic acid (AKBA)], (4) MIA + F2 (curcuminoids+Withania glycosides+AKBA), (5) MIA + F3 (curcuminoids+total withanolides+AKBA), (6) MIA + F4 (curcuminoids, AKBA), (7) MIA + UCII (type II collagen), and (8) MIA + GCHON (Glucosamine Chondroitin). Treatments F1 to F4 reduced right joint diameter and improved stride length and paw area in OA rats. Despite improvements with treatments F1 to F4, there was no significant difference between these groups ( p > .05). In OA animals, F1 to F4 treatments decreased MDA levels and increased antioxidant enzymes activities ( p < .001). This was done by reducing levels of inflammatory markers and enzymes like IL‐1β, IL‐6, MMP‐8, TNF‐α, CRP, COMP, and LOX‐5, while increasing the anti‐inflammatory cytokine IL‐10. In conclusion, these plant‐based treatments significantly reduced osteoarthritis severity, slowed disease progression by reducing inflammation, and protected joints from damage, showing a protective effect in rats with induced osteoarthritis, likely due to their anti‐inflammatory and antioxidant properties.
There has been an increasing interest in the usage of natural products for the prevention and treatment of chronic diseases such as cardiovascular disease (CVD). Regular consumption of fruits, vegetables and whole grains has been shown to be negatively correlated with the risk of CVDs. These foods provide a diversity of nutrients and different bioactive compounds including phytochemicals, vitamins, minerals and fibers which play critical roles in the sustainability of optimal cardiovascular health. Plant-based foods or a nutritional cure are gradually being integrated into medical practice for CVD management partly due to the supporting experimental studies, clinical trials and epidemiological studies. These products have anti-oxidant, anti-inflammatory, hypoglycemic, hypolipidemic, hypotensive, anti-atherosclerotic, anti-thrombotic and hypocholesterolemic effects, depending on the dosage in cell and tissue cultures, animal models as well as in humans. The present review considers some novel ideas on some major phytochemicals which have been suggested to have protective and therapeutic potential in CVD. The data presented in this work have been compiled from studies that have mostly been carried out in recent years. GirişRubella virus Togaviridae familyasının Rubivirus genusunun tek üyesi olan zarflı ve tek sarmallı ribonükleik asit taşıyan bir virüstür (1).Virus solunum yoluyla bulaşır ve nazofarinkste replike olduktan sonra servikal lenf nodlarında çoğalmaya devam eder.Kan dolaşımına karışan virus bu yolla vücuda yayılır.ÖZET Amaç: Rubella virus Togaviridae familyasının Rubivirus genusunun tek üyesidir.Zarflıdır ve tek sarmallı ribo nükleik asit genomuna sahiptir.Bu viruse ilk trimesterde maruz kalan annelerin doğmamış çocukları konjenital rubella sendromu açısından yüksek risk altındadırlar.Bu çalışmada hastanemiz gebe polikliniklerine başvuran gebelerde rubella antikorlarının sıklığının tespit edilmesi amaçlandı.Gereç ve Yöntem: Bu çalışmada hastanemiz gebe polikliniğine 31.07.2009ile 01.08.2014 tarihleri arasında başvuran 1037 gebenin rubella IgM ve 424 gebenin rubella IgG antikor değerleri retrospektif olarak incelendi.Bulgular: Rubella IgG ölçümüne göre olguların %1,9'unun (n=8) rubella IgG sonucu negatif, %4,2'sinin (n=18) ara değer ve %93,9'unun (n=398) rubella IgG sonucu pozitif olarak saptandı.Rubella IgM ölçümüne göre olguların %99,2'sinin (n=1029) rubella IgM sonucu negatif, %0,5'inin (n=5) ara değer ve %0,3'ünün (n=3) rubella IgM sonucu pozitif olarak saptandı.Sonuç: Bu çalışma ile yöremizdeki gebe kadınlarda 5 yıllık dönemde rubella virüs seroprevalansı tespit edildi.Bölgemizde rubella IgG seroprevalansı oldukça yüksektir ve bu sebeple gebelerde rutin olarak taranmasının gerekli olmadığını düşünüyoruz.Rubella IgM seropozitivitesi ise oldukça düşüktür ve bu sebeple de bu bölgede rutin rubella IgM taraması yapmadan önce obstetrisyenlerin maliyet etkinlik açısından değerlendirme yapmaları gerekir.
