    Abstract:
    The bioconversion of environmental wastes into energy is gaining much interest in most developing and developed countries. The current study is concerned with the proper exploitation of some industrial wastes. Cellulosic fiber waste was selected as a raw material for producing bioethanol as an alternative energy source. A combination of physical, chemical, and enzymatic hydrolysis treatments was applied to maximize the concentration of glucose that could be fermented with yeast into bioethanol. The results showed that the maximum production of 13.9 mg/mL of glucose was achieved when 5% cellulosic fiber waste was treated with 40% HCl, autoclaved, and followed with enzymatic hydrolysis. Using SEM and FTIR analysis, the instrumental characterization of the waste fiber treatment confirmed the effectiveness of the degradation by turning the long threads of the fibers into small pieces, in addition to the appearance of new functional groups and peak shifting. A potent yeast strain isolated from rotten grapes was identified as Starmerella bacillaris STDF-G4 (accession number OP872748), which was used to ferment the obtained glucose units into bioethanol under optimized conditions. The maximum production of 3.16 mg/mL of bioethanol was recorded when 7% of the yeast strain was anaerobically incubated at 30 °C in a broth culture with the pH adjusted to 5. The optimized conditions were scaled up from flasks to a fermentation bioreactor to maximize the bioethanol concentration. The obtained data showed the ability of the yeast strain to produce 4.13 mg/mL of bioethanol after the first 6 h of incubation and double the amount after 36 h of incubation to reach 8.6 mg/mL, indicating the efficiency of the bioreactor in reducing the time and significantly increasing the product.

    Filamentous fungi can secrete cellulases and related enzymes to decompose cellulosic substrates,and it is served as the primary producer of cellulase in the industry. This review gives an overview of the enzyme systems for cellulose hydrolysis and the new proposed mechanism of cellulose decomposition in filamentous fungi. The recent progress in cellulase exploration by the omics technologies is introduced,and the genetic strategies for improving cellulase efficiency and protein production are discussed.

    The plentiful renewable resources-cellulosic materials could be used to produce fuel alcohol through bioconversion. The research on bioconversion of cellulosic materials to produce fuel alcohol around the world mainly focused on the pretreatment of raw materials, the approaches to reduce cellulase cost, the optimization of fermentation course, appropriate use and transformation of microbial species, and the comprehensive use of waste distiller's materials etc. In this paper, the present status of fuel alcohol production by cellulosic materials and the industrilization trend in the near future in the United States, Japan, Canada, Sweden and China were introduced.

    Living organisms produce a wide variety of polymeric carbohydrates, of which cellulose is the most abundant. The main deterioration of cellulose is caused by microorganisms, and the biological degradation of cellulosic substances is an extremely important process for the maintenance of the carbon balance in nature. Since crystalline cellulose is an insoluble substance with a supramolecular structure, the enzymological approaches to this substrate are rather complicated and difficult . In most cellulolytic microorganisms, several cellulase components together constitute a "cellulase system" and cellulosic materials are converted to soluble sugars by their synergistic action . To understand completely the biodegradation mechanism of cellulose, therefore, the strict purification and characterization of each cellulase component is absolutely necessary . This review describes the cellulolytic enzymes and focuses on the mechanism of their action . It is divided into seven sections and includes the historical background of cellulase . Our recent findings concerning the enzyme mechanisms involved in native cellulose degradation are also described.
