Abstract:
Two fusidane-type active compounds (6 and 7) and five new ones (1-5), along with other nine known compounds (8-16) were isolated from the metabolites of Schizophyllum commune MST7-3. Their structures were elucidated on the basis of spectroscopic analysis. The absolute configurations of compounds 2 and 3 were established by Mosher's method and optical rotation. Compounds 6 and 7 showed significant antibacterial activities against Stenotrophomonas maltophilia with MIC values of 4 μg/mL and 16 μg/mL, respectively.Keywords:

The leaves and stems of Hyptis atrorubens Poit. (Lamiaceae) were collected in Guadeloupe. The methanol extract was tested at six concentrations (10mg/mL, 5mg/mL, 2.5mg/mL, 1.2mg/mL, 0.6mg/mL and 0.3mg/mL) against a panel of pathogenic Gram-positive and Gram-negative bacteria in vitro. This methanolic extract demonstrated antibacterial activity against two Gram-positive bacteria (Staphylococcus epidermidis and Enterococcus faecalis), and two Gram-negative bacteria (Burkholderia cepacia and Stenotrophomonas maltophilia) at all of the concentrations. Bioautography enabled the obtention and identification of two antibacterial compounds from this plant: rosmarinic acid and methyl rosmarinate. This work supports the recognition of H. atrorubens, a traditional remedy in French West Indies, as a medicinal plant in France.

Aim. Study spectrum and expressiveness of antibacterial and antifungal properties oflichen extracts. Materials and methods. Antimicrobial activity of acetone extracts from Hypogymnia physodes, Xanthoria parietina, Evernia prunastri, Ramalina pollinaria, Cladonia arbuscula lichens was determined by micro-dilution methods in broth for 4 - 500 mcg/ml concentrations against 13 strains from ATCC collection and 6 clinical isolates. Results. High antibacterial activity of H. physodes and C. arbuscular extracts against staphylococci and enterococci was detected (MIC 31 - 62 mcg/ml). Antimicrobial activity against enterobacteria and Pseudomonas aeruginosa was absent for all the extracts. E.prunastri, H.physodes and C arbuscula extracts were active against Stenotrophomonas maltophilia strains (MIC 250 - 500 mcg/ml). Antifungal activity (MIC 500 mcg/ml for 4 Candida strains) was only detected for the E. prunastri extract. Conclusion. H.physodes and C. arbuscula lichens can be examined as a perspective source of antibacterial substances, effective against antibiotics resistant staphylococci, streptococci and S. maltophilia strains.

Stenotrophomonas sepilia strain SM16975 (= JCM 32102; = KCTC 62052) is a new species isolated from the blood culture of a hospitalized patient. The biochemical characterization, phenotypic criteria, phylogenomic reconstruction, and genomic analysis were carried out to differentiate it from its phylogenetic neighbours, establishing novel species status in the genus Stenotrophomonas and within Stenotrophomonas maltophilia complex (Smc).

This study reports the presence of the blaNDM-1 gene in an isolate of Stenotrophomonas maltophilia obtained from a Brazilian soil, inside an IncA/C plasmid with ~ 45 Kb. To the best of our knowledge, this is the second report in the world and the first in Brazil of NDM-producing bacterium isolated from soil.

Endophytic bacteria are the source of novel bioactive compounds, used as therapeutic agent. Molecular docking is a computational technique use frequently, to find novel drugs targets and drugs-receptors interactions. The current study was designed to isolate and identify endophytic bacteria for the extraction of bioactive compounds. Further, to characterized extracts and to explore compounds interactions with bacterial cell wall and outer membrane synthesizing proteins. Endophytes were identified using 16s rRNA amplification technique. For bioactive compounds, solvent extraction method was followed and characterized further through GC-MS analysis. To find targets and drugs-receptors interactions, molecular docking studies and biological assays were performed. The isolated endophytes belong to five different genera namely Enterobacter , Bacillus , Erwinia , Stenotrophomonas and Pantoea . In case of antibacterial assay Stenotrophomonas maltophilia extract showed significant inhibitory zones (15.11±0.11mm and 11.3±0.16) against Staphylococcus caseolyticus and Acinetobacter baumanni , with MIC 33.3 and 50μg/mL respectively. Among the characterized fifty compounds, from endophytic bacteria “antibacterial compound” N-(5-benzyl-10b-hydroxy-2-methyl-3,6-dioxooctahydro-8H-oxazolo[3,2-α] pyrrolo[2,1c] pyrazin-2-yl)-7-methyl2,3,3a,3a 1 ,6,6a,7,8,9,10,10a,10b-dodecahydro-1H-4λ 2 -indolo[4,3- f g]quinoline-9-carboxamide of bacteria Stenotrophomonas maltophilia were an excellent binder with MurF ligase active site, with binding energy of -10.2 kcal/mol. Extracts of endophytic bacteria composed of various pharmacologically active ingredients such as antibacterial compounds. Molecular docking studies provide important information regarding drug-receptor interaction, thus can be used in novel drug discovery.

To assess the influence of Stenotrophomonas on plants, the interaction of 16 Stenotrophomonas strains from clinical and environmental sources with strawberry plant seedlings was analysed.In vitro, all Stenotrophomonas strains influenced plant growth when applied to seedlings. Whereas most of the Stenotrophomonas strains promoted root growth and hair development, a statistically significantly negative influence on the length of stem was found. Although strains from a clinical origin also showed statistically significant effects on plants, this was generally lower when compared with environmental strains. For three selected strains, a strong dose-dependent effect was observed for all parameters. In vitro, a correlation was found between plant growth promotion and production of a plant growth hormone, indole-3-acetic acid (IAA). Xanthomonas campestris, a phylogenetically very closely related species to Stenotrophomonas, was used as a phytopathogenic control. It too confirmed the reduction of plant growth in this in vitro system.Independent of their origin, Stenotrophomonas strains can produce IAA in vitro and subsequently, influence plant growth. The effect of Stenotrophomonas presence on plants was dose-dependent.The dose-dependent effect of Stenotrophomonas, a bacterium of both biotechnological and medical interest, is of great interest for biocontrol applications of plant-associated strains. This paper is the first report that clearly demonstrates the phytopathogenic capacity of Stenotrophomonas.

In vitro antibacterial efficacy of 14 commercially available disinfectants against the hospital pathogen Stenotrophomonas maltophilia was tested. The test disinfectants included 5 "pure" quartenary ammonium compounds (QACs) and 9 combination formulations with QACs as the major ingredient. Antibacterial efficacy was expressed as MIC, ED50 and inhibition of the rate of incorporation of radioactive precursors [14C] adenine and [14C] leucine. Based on their activity, the disinfectants were divided into four groups. Group I characterized by high inhibitory activity (MIC 0.045-0.09 microg/ml) comprised benzalconium chloride and Triquart. Group II included formulations with good antibacterial activity (MIC 0.19-0.78 microg/ml) while group III formulations showed MICs between 1.56 and 25 microg/ml. The less active Cetrimid (MIC 50-100 microg/ml) was classified into group IV. When effects on biosynthetic processes expressed as R values (IC50 Ade : IC50 Leu) were tested, R < 1 was only recorded for the following formulations: Hexaquart plus, Diesen forte, Sokrena, Forten, ID 212 and Cetrimid. Lower values of IC50 Ade and IC50 Leu are suggestive of effects on the synthesis of nucleic acids and proteins leading to inhibition of the two precursors. Endogenous respiration was almost 50% inhibited by Cetrimid and Microbac forte at a concentration of 0.78 microg/ml, and 100% inhibited by Almyrol, Diesen forte and Microbac forte at a concentration of 6.25 microg/ml and by FD312, Triquart, Hexaquart plus, Hexaquart S, ID 212, TPH 5225 at a concentration of 12.5 microg/ml.
