    Effect of Processing Methods and Storage Time on the Content of Bioactive Compounds in Blue Honeysuckle Berry Purees

    Abstract:
    In this study, we aimed to investigate the effect of processing methods and storage time on selected parameters of quality in the purees of blue honeysuckle berries. We investigated the content of bioactive compounds such as anthocyanins, L-ascorbic acid, and total polyphenols (TPs). We tested four processing methods and two varieties of blue honeysuckle berry (Lonicera caerulea L.var. Sineglaska and Volshebnica). The purees were analyzed immediately after production, and after two and four months of storage at 20 °C without exposure to light. According to our results, thermal treatment of the fruits for 5 min resulted in obtaining purees with a higher content of anthocyanins and TPs, but lower content of L-ascorbic acid. However, sieving fruits that were not thermally treated resulted in a higher content of L-ascorbic acid. Furthermore, purees prepared from the fruits of the Volshebnica cultivar were richer in bioactive compounds than that of purees prepared from the Sineglaska cultivar. In addition, the content of anthocyanins, L-ascorbic acid, and TPs decreased with storage time.

    The extraction of chlorogenic acid from spectacular honeysuckle with ultrasonic wave was studied.On the basis of mono factor and orthogonal experimental design,the influence of several parameters on the extraction rate of chlorogenic acid was investigated,including ultrasonic time,material/extraction solution ratio,ethanol concentration,extraction temperature.The optimal extraction process was determined.The result shows that the optimum extraction conditions extracting of chlorogenic acid from spectacular honeysuckle by ultrasound were as follows:ethanol concentration 60%,extraction temperature 70 ℃,ultrasonic time 40 min and ratio of liquor to material 20 mL/g.Under the above optimum conditions,the chlorogenic acid extracted from spectacular honeysuckle was yielded by 36.72 mg/g.

    Objective To identify the best picking-up time and dehydrating method of Xianglei Honeysuckle.Methods The content of chlorogenic acid in buds of Honeysuckle was assayed by HPLC.Results With growth of buds,the content of chlorogenic acid decreases,the content of chlorogenic acid reaches the maximum in those Honeysuckle picked in early morning,also,after different dehydrating methods,the content of chlorogenic acid in buds varies a lot,manually fried buds contain the largest amount of chlorogenic acid while directly fried ones contain the least.Conclusion The best time for picking-up Honeysuckle is about June 27th,especially in early morning,and the best dehydrating method is ShaQing Dehydrating.

    Objective Optimization of Lonicera chrysantha fruit of chlorogenic acid extraction process.Methods Using ultrasonic extraction to extract chlorogenic acid from Lonicera chrysantha fruit,UV-visible determination of chlorogenic acid in Lonicera chrysantha fruit.Results The Lonicera chrysantha fruit extraction of chlorogenic acid in the best extraction conditions for ultrasonic extraction.Conclusion Huanghua honeysuckle fruit contains chlorogenic acid and its content is higher,the flower is the comprehensive exploitation and utilization of fruit honeysuckle laid a foundation.

    Chlorogenic acid in aqueous solution produces a weak fluorescence,while the fluorescence intensity is greatly enhanced in 95% ethanol.Based on this fact,a new method for the determination of chlorogenic acid was established.At pH=1.72,chlorogenic acid fluorescence intensity and concentration showed good linear relationship with the correlation coefficient of 0.9963.The detection limit was 0.081 μg/mL,and the relative standard deviation was 0.54%.The method was sensitive,stable,and successfully used for the determination of chlorogenic acid in Honeysuckle and Eucommia Bark.The results also showed that chlorogenic acid in Honeysuckle and Eucommia Bark have good antioxidant activity.

    Objective To study the water extraction method of chlorogenic acid in Honeysuckle.Methods Orthogonal experiment design was used.Three factors were studied as indexes that might affect the yield rate of chlorogenic acid,including water quantity,extraction temperature and time required.Results The best extraction condition was:10 times amount of water,1 hour of extraction at 75 ℃.Conclusion This extraction method is convenient and feasible,and can improve the extraction of chlorogenic acid from honeysuckle.

    In order to improve the extraction rate of Chlorogenic Acid from Honeysuckle,response surface methodology( RSM) was applied to optimize the extraction technology of Chlorogenic Acid from Honeysuckle. Based on single factor experiment,the influence factors of alcohol concentration、solution and material ration、 extraction temperature were studied on the contents of Chlorogenic Acid from Honeysuckle extraction. According to Box-Behnken combination method( Design-expert 8. 0. 5. 0),the optimal conditions were determined as follows: alcohol concentration 62% 、solution and material ration 18. 7∶ 1( mL / g) 、extraction temperature 58℃ 、 extraction time 40min. Under the conditions,the relative error between the testing values and predicted value was 3. 5%. This optimal extraction process can be used for extracting the Chlorogenic Acid in Honeysuckle. It is simple,stable and reproducible.

    The chlorogenic acid contents in Luotian honeysuckle and Dadi honeysuckle from Luotian County were compared by using UV spectrophotometer.This study result shows that the contents of chlorogenic acid in two honeysuckles leaves luotian County are higher,and the chlorogenic acid content in leaf is almost 2.5 times as much as that in flower from Luotian honeysuckle.The result provides abasis for rational exploitation and comprehensive utilization of Luotian honeysuckle plant resource.

    OBJECTIVE To establish an analytical method of determinating Chlorogenic acid in Honeysuckle Stem by HPLC.METHODS Chlorogenic acid in Honeysuckle Stem was separated by HPLC on Thermo Hypersil BDS (4.6×250mm,5μm)with a acetonitrile-0.4%H_3PO_4 (10∶90)as mobile phase and detection wavelength was at 327nm.RESULTS There was a good linear relationship with a range of 0.1656~0.9936μg for chlorogenic acid,r=0.9999,the average recovery rate were 98.8%.CONCLUSION The method was accurate,rapid and simple for quantitative analysis of Honeysuckle Stem.

    The effect of four factors on the concentration of chlorogenic acid in honeysuckle's extraction with orthogonal test is examined.The four factors are the concentration of ethanol,volume of solvent,extraction hour and extraction times.This method does not need chlorogenic acid.The results show that the concentration of ethanol has an apparent effect on the concentration of chlorogenic acid.
