    Objective The aim of this study was to evaluate the consistency and accuracy of all the parameters of the urine samples detected by two automated urine sediment analyzers from Sysmex Corporation. Methods Two automated analyzers and manual microscopy examined 1,059 urine samples. The sensitivity, specificity, positive predictive value, and negative predictive value were evaluated. The consistency of all the parameters was tested. The influencing factors of false positive and false negative samples were analyzed and compared. Results All the parameters had good specificity, negative predictive value, and coincidence rate (83.95%-99.61%). The RBC, WBC, and X'TAL analyzed by UF-5000 and UF-1000i exhibited good agreement (Kappa=0.597-0.784) with those by manual microscopy. The overall concordance rates of RBC and WBC were good (RBC: r=0.9842, CCC=0.9693; WBC: r=0.9955, CCC=0.9711). Among the influencing factors, mucus filament accounted for a large proportion, which mainly affected the detection of CAST. Concurrently, the false-positive factors of EC detection were reduced, and CAST did not affect the detection of EC. Conclusion The parameters of the two instruments tested have shown high accuracy, consistency, coincidence rate, and low negative predictive value for RBC and WBC, which has ensured that UF-5000 and UF-1000i meet the clinical requirements for urine tests for disease screening. For the samples with poor consistency and false-positive factors, a conventional microscopic examination should be applied to verify the accuracy of the instrument detection.

    Objective To study the clinical value of protein chip system in the detection of multi-tumor markers(C-12 system).Methods The serums of 114 malignant tumour patients,32 benign disease patients and 52 healthy subjects were detected by C-12 system.The sensitivity,specificity,accuracy,positive predictive value and negative predictive value of C-12 system were analyzed.The results of C-12 system were compared with those of chemiluminescence immunoassay,DELFIA and IRMA,and the correlation between C-12 system and the others was evaluated.Results Among the 114 malignant tumour patients,106 were positive,while 19 were positive among 84 controls.The sensitivity,specificity,accuracy,positive predictive value and negative predictive value of C-12 system were 92.9%,77.4%,86.4%,84.8%,89.0%,respectively.There was significant positive correlation between the results of C-12 system and the others(r=0.494~0.895,P 0.001).Conclusion C-12 system is appropriate for clinical diagnosis and outcome prediction of tumours,particularly for tumour screening of healthy group.

    Objective To explore the effectiveness of ThinPrep imaging system (TIS) in screening for cervical cancer.Methods The same specimens of cervical cells were preliminarily screened by manual screening (MS) and TIS.The final diagnosis for the screened specimen with different results was made by two senior physicians.The results diagnosed by MS and TIS were compared with the final diagnosis to calculate the detection rate of ASC+,false negative rate,false positive rate,positive predictive value,negative predictive value,sensitivity,and specificity.Results Of 925 screened specimens,75 (8.1%) were ASC+ positive by MS and 86 (9.3%) by TIS.The false negative rate was 23 (2.5%) in MS and 10 (1.1%) in TIS; the false positive rate was 15 (1.6%) and 13 (1.4%); the negative predictive value was 97.3% and 98.8%; the positive predictive value was 72.3% and 88.0%; the sensitivity was 72.3% and 88.0%; and the specificity was 98.2% and 98.5%,respectively.Conclusions The preliminary data suggest that the sensitivity for TIS was much better than that for MS.TIS significantly reduces the false negative rate but has no effect on the specificity.Doctors need to be properly trained to adapt the new screening method after application of new technology.     ABSTRACT The purpose of this study was to evaluate the diagnostic performance of LIAISON Treponema Screen (DiaSorin, Saluggia, Italy), a new automated chemiluminescence immunoassay (CLIA), in comparison with that of rapid plasma reagin (RPR) and the following currently used treponemal tests: hemagglutination test (TPHA), immunoenzymatic assay (EIA), and Western blot (WB). First, a retrospective study was performed with a panel of 2,494 blood donor sera, a panel of 131 clinical and serologically characterized syphilitic sera, and 96 samples obtained from subjects with potentially interfering diseases or conditions. A prospective study was also performed by testing 1,800 unselected samples submitted to the Microbiology Laboratory of the St. Orsola Hospital in Bologna, Italy, for routine screening for syphilis. As expected, RPR was the least specific method, especially when potentially cross-reacting sera were tested. On the contrary, all of the treponemal tests proved to be very specific (99.9%) and they performed with the following sensitivities: 100% (WB), 99.2% (CLIA), 95.4% (EIA), and 94.7% (TPHA).
