    Abstract:
    Objective To investigate the expression of pituitary tumor transforming gene(PTTG)and P53 in colorectal cancerpatients and the relationship between levels of PTTG and and P53 and progression and metastasis of colorectal cancer Methods Immunochemical staining was performed to detect the expression of PTTG and P53 in 42 colorectal cancer tissues and 42 normal control tissues.Results The positive rate of PTTG,and P53 were 88.1% and 95.2% in colorectal cancer tissues.The positive rate and expression grade in colorectal cancer tissues were significantly higher than that in normal control tissues(P0.01),No relationship between expression of PTTG and tumor typing or differentiation colorectal cancer(P0.05).but the expression of PTTG in metastasis group was much higher than the negative group(P0.05).No relationship between expression of P53 and tumor typing,metastasis or differentiattion of colorectal cancer was observed(P0.05).Conclusion The expression of PTTG,is associated with the progression and metastasis of colorectal cancer,and PTTG,may be used as molecular markers for judging the metastasis potential and the prognosis of colorectal cancer.

    ObjectiveTo explore the roles of pituitary tumor transforming gene(PTTG) in the occurrence and development of colorectal carcinoma.MethodRT-PCR and immunohistochemical method were used to examine the expression of PTTG mRNA and PTTG protein in 80 colorectal carcinoma tissues and 20 normal mucosa tissues,the correlation between which and clinical and pathologic parameters were analyzed.ResultsExpression of PTTG mRNA and protein in colorectal carcinoma tissues were both signifycantly higher than those in normal mucosa tissues(both P0.01);both the expression of PTTG mRNA and protein were positively correlated with depth of invasion,lymph node metastasis and Duke's stage.ConclusionsThe high expression of PTTG may improve the occurrence and development of colorectal cancer;It could be a predictive and prognostic factor of colorectal carcinoma.

    Objective:To evaluate the expressions of PTTG and COX2 in breast invasive ductal carcinoma and their clinical significance.Methods:The expressions of PTTG and COX2 were examined in 104 patients with breast invasive duct carcinoma using Evinson techniques.Results:The positive rates of PTTG and COX2 expressions were 91% and 67% respectively.There were significant associations between PTTG expression and histological grade and tumor size.There were nosignificant associations between PTTG expression and age,lymph node metastasis,ER,PR,HER2 state.The expressions of COX2 had correlation with ER state,tumor size,histological grade,lymph node metastasis.There were no significant associations between COX2 expressions and PTTG expression and age,PR,HER2 state.Conclusion:The expression of PTTG was possibly involved in the development of breast cancer,and had correlations with low differentiation degree and bad prognosis.COX2 might promote breast cancer proliferation of cells by stimulating estrogen combining.

    Objective To detect the expression and role of PTTG mRNA in human colorectal cancer.Methods The expression of PTTG mRNA was evaluated in 12 normal colorectal tissues,20 colorectal adenoma tissues and 44 colorectal cancer tissues by RT-PCR.Results The expression of PTTG mRNA in colorectal cancer was significantly higher than that in colorectal adenoma and normal colorectal tissues.The PTTG mRNA expression in the Dukes C,D colorectal cancer was higher than that in the Dukes A,B cancer(P0.05).The expression in the colorectal cancer with lymph node metastasis was higher than that in the cancer without lymph node metastasis(P0.05).Conclusion The expression of PTTG mRNA increases in colorectal cancer,and is related with cell differentiation and metastasis.The abnormal expression of PTTG probably participates in genesis and development of colorectal cancers.

    The purpose of the present study was to investigate the association between the expression of pituitary tumor-transforming 1 (PTTG1) and the expression of matrix metalloproteinase (MMP)-2 and MMP-9 in laryngeal carcinoma tissues, and to elucidate the association between PTTG1 expression and the prognosis of patients with laryngeal cancer. Immunohistochemical staining was used to detect PTTG1 expression in laryngeal cancer and normal tumor-adjacent laryngeal tissues. Western blotting was used to determine the levels of PTTG1 and MMP-2 and -9 in laryngeal carcinoma tissues and to assess their correlation. In addition, the associations between PTTG1 expression and the clinical parameters of laryngeal cancer and patient survival were determined. The immunohistochemistry results revealed that the positive expression rates of PTTG1, MMP-2 and MMP-9 in the laryngeal cancer tissues were significantly higher than those in the carcinoma-adjacent normal laryngeal tissues (all P<0.05). In addition, expression levels of PTTG1, MMP-2 and MMP-9 were significantly associated with lymph node metastasis, histological grade and clinical stage (P<0.05). Furthermore, the levels of PTTG1 were positively correlated with the levels of MMP-2 and MMP-9 in laryngeal cancer tissues (P<0.05). In summary, the expression levels of PTTG1, MMP-2 and MMP-9 are closely associated with the biological behaviors of laryngeal cancer tissues, showing that they serve important roles in the occurrence and development of laryngeal cancer, and may be useful as biological indicators of laryngeal tissue invasion, metastasis and patient prognosis.

    Objective To explore the role of PTTG expression in the occurrence and development of colorectal carcinoma.Methods RT-PCR technique was used to examine mRNA expression of PTTG and immunohistochemical staining was used to examine protein expression of PTTG in 20 cases of normal mucosa,20 cases of adenoma and 80 cases of colorectal carcinoma tissues.Results Expression of PTTG at mRNA and protein appeared an upward tendency in the order from normal mucosa and adenoma to colorectal carcinoma and the differences was significant between any two groups(P0.01).Expression of PTTG at mRNA and protein was also positively correlated with depth of invasion,lymph node metastasis(P0.05 and P0.01,respectively).Conclusion The high expression of PTTG may play an important role in the occurrence and development of colorectal cancer.

    Objective To determine the expression of PTTG and c-Myc in colorectal adenocarcinoma and their significance.Methods To detect the expressions of PTTG and c-Myc in 55 cases of colorectal adenocarcinoma and 15 cases of normal colorectal mucosa by immunohistochemistry SP method.Results The expressions of PTTG and c-Myc were located within the cytoplasm and nucleus.The positive expression rates were 75%(41/55) and 67%(37/55)and 13%(2/15) and 27%(4/15) the normal colorectal mucosa,respectively.Both of their expressions were not related with the age and sex of the patients(P0.05),but were related with lymph node metastasis,clinical stage and pathological staging(P0.05).The positive expressions of PTTG in colorectal adenocarcinoma were positively correlated with those of c-Myc(P0.05).[WTHZ]Conclusion PTTG and c-Myc in colorectal adenocarcinoma expression suggested that PTTG expression may be associated with invasion,development and metastasis of the colorectal adenocarcinoma.PTTG and c-Myc played an important role in the development of colorectal adenocarcinoma.

    Objective:To investigate expression of PTTG and its signification in prostatic tissues,especially in prostate cancer.Methods:The expression of PTTG was detected with two-step immunohistochemical staining technique in 8 cases of normal prostatic tissues,16 cases of BPH,47 cases of non-metastasis prostate cancer and 19 cases of metastasis prostate cancer.The PTTG expression situation of normal prostatic tissue,BPH and prostate cancer was analysed by Stata as well as the relationship between the PTTG expression in prostate cancer and PSA and Gleason grade.Results:PTTG expression was found in 42 of 66(63.7%)of prostate cancer tissues.no marked expression was found in normal and BPH tissues.There was significantly different expression between Benign and malignant prostate tissue(x~2=25.2487,P= 0.000).No significantly different expression between non-metastasis prostate cancer and metastasis prostate cancer.No association was found between PTTG expression and Gleason score or pretreatment PSA.Conclusions:These data suggested that there is significant PTTG overexpression in prostate cancer tissues,which may indicate PTTG playing a important role in the development of prostate cancer.
