Abstract:
Objective To detect the expression and role of PTTG mRNA in human colorectal cancer.Methods The expression of PTTG mRNA was evaluated in 12 normal colorectal tissues,20 colorectal adenoma tissues and 44 colorectal cancer tissues by RT-PCR.Results The expression of PTTG mRNA in colorectal cancer was significantly higher than that in colorectal adenoma and normal colorectal tissues.The PTTG mRNA expression in the Dukes C,D colorectal cancer was higher than that in the Dukes A,B cancer(P0.05).The expression in the colorectal cancer with lymph node metastasis was higher than that in the cancer without lymph node metastasis(P0.05).Conclusion The expression of PTTG mRNA increases in colorectal cancer,and is related with cell differentiation and metastasis.The abnormal expression of PTTG probably participates in genesis and development of colorectal cancers.Keywords:

ObjectiveTo explore the roles of pituitary tumor transforming gene(PTTG) in the occurrence and development of colorectal carcinoma.MethodRT-PCR and immunohistochemical method were used to examine the expression of PTTG mRNA and PTTG protein in 80 colorectal carcinoma tissues and 20 normal mucosa tissues,the correlation between which and clinical and pathologic parameters were analyzed.ResultsExpression of PTTG mRNA and protein in colorectal carcinoma tissues were both signifycantly higher than those in normal mucosa tissues(both P0.01);both the expression of PTTG mRNA and protein were positively correlated with depth of invasion,lymph node metastasis and Duke's stage.ConclusionsThe high expression of PTTG may improve the occurrence and development of colorectal cancer;It could be a predictive and prognostic factor of colorectal carcinoma.

Objietive: To investigate the expression and significance of selenoprotein-P in colorectal cancer,metastatic lymph node and adjacentmucosa of colorectal cancer,and discuss the effect of selenoprotein-p on occurring and developing of colorectal cancer.Methods: The expression of selenopreotein-P in adjacentmucosa of colorectal cancer,and in the cancer、 metastatic lymph node were detected by means of immunohistochemistry.Results: The positive rate of selenorotein-P were 53.1%,55.7%,80.0% in tumor cell,metastatic lymph node and adjacentmucosa of colorectal cancer.The expression of adjacentmucosa was significantly higher than that two sites(χ2=4.87,P0.05).The positive of selenoprotein-P expression in the tissues of colorectal cancer was not related to other pathological factors.Conclusions: The expression of selenoprotein-P is low in colorectal cancer,and may play an important role in occurring and developing of colorectal cancer.

Objective To explore the role of PTTG expression in the occurrence and development of colorectal carcinoma.Methods RT-PCR technique was used to examine mRNA expression of PTTG and immunohistochemical staining was used to examine protein expression of PTTG in 20 cases of normal mucosa,20 cases of adenoma and 80 cases of colorectal carcinoma tissues.Results Expression of PTTG at mRNA and protein appeared an upward tendency in the order from normal mucosa and adenoma to colorectal carcinoma and the differences was significant between any two groups(P0.01).Expression of PTTG at mRNA and protein was also positively correlated with depth of invasion,lymph node metastasis(P0.05 and P0.01,respectively).Conclusion The high expression of PTTG may play an important role in the occurrence and development of colorectal cancer.

Objective To determine the expression of PTTG and c-Myc in colorectal adenocarcinoma and their significance.Methods To detect the expressions of PTTG and c-Myc in 55 cases of colorectal adenocarcinoma and 15 cases of normal colorectal mucosa by immunohistochemistry SP method.Results The expressions of PTTG and c-Myc were located within the cytoplasm and nucleus.The positive expression rates were 75%(41/55) and 67%(37/55)and 13%(2/15) and 27%(4/15) the normal colorectal mucosa,respectively.Both of their expressions were not related with the age and sex of the patients(P0.05),but were related with lymph node metastasis,clinical stage and pathological staging(P0.05).The positive expressions of PTTG in colorectal adenocarcinoma were positively correlated with those of c-Myc(P0.05).[WTHZ]Conclusion PTTG and c-Myc in colorectal adenocarcinoma expression suggested that PTTG expression may be associated with invasion,development and metastasis of the colorectal adenocarcinoma.PTTG and c-Myc played an important role in the development of colorectal adenocarcinoma.

Objective To study the expression of eukaryotic initiation factor 4E(eIF4E)in colorectal cancer and to explore the mechanism of eIF4E in the pathogenesis of colorectal cancer and its clinical significance.Methods The expression of eIF4E in 62 colorectal cancers,43 colorectal adenomas and 30 normal colorectal tissues were detected by S-P immunohistochemical method.Results The positive rate of expression of eIF4E in colorectal cancer was significantly higher than that of colorectal adenoma and normal colorectal tissue(P0.01).The eIF4E overexpression in colorectal cancers significantly correlated with depth of invasion,lymph node metastasis(P0.05).However,no significant correlation was noted between the expression of eIF4E and the locations or differentiated degrees of cancer.Conclusion The overexpression of eIF4E may play a critical role in the carcinogenesis of colorectal cancer and the overexpression of eIF4E may be used as a prognosis marker of colorectal cancer.

Objective To investigate the expression of pituitary tumor transforming gene(PTTG)and P53 in colorectal cancerpatients and the relationship between levels of PTTG and and P53 and progression and metastasis of colorectal cancer Methods Immunochemical staining was performed to detect the expression of PTTG and P53 in 42 colorectal cancer tissues and 42 normal control tissues.Results The positive rate of PTTG,and P53 were 88.1% and 95.2% in colorectal cancer tissues.The positive rate and expression grade in colorectal cancer tissues were significantly higher than that in normal control tissues(P0.01),No relationship between expression of PTTG and tumor typing or differentiation colorectal cancer(P0.05).but the expression of PTTG in metastasis group was much higher than the negative group(P0.05).No relationship between expression of P53 and tumor typing,metastasis or differentiattion of colorectal cancer was observed(P0.05).Conclusion The expression of PTTG,is associated with the progression and metastasis of colorectal cancer,and PTTG,may be used as molecular markers for judging the metastasis potential and the prognosis of colorectal cancer.

Objective To investigate the expressions of pituitary tumor transforming gene(PTTG) and P53 in colorectal carcinoma and evaluate their clinical significance.MethodsSpecimens from colorectal carcinoma(60 cases) and normal colorectal tissues(20 cases) were analyzed by tissue microarray and immunohistochemical detection for PTTG and P53 expressions.ResultsThe positive rates of PTTG and P53 were 75.0% and 61.7%,respectively in colorectal carcinoma.The positive rates and expression grades in colorectal cancer tissues were significantly higher than that in normal colorectal tissues(P0.01).The expression of PTTG was correlated with lymph node metastasis,tumor diameter,and depth of infiltration of colorectal carcinoma(P,and P0.05).The expression grade of P53 was not correlated with differentiation,tumor diameter,and depth of infiltration of colorectal carcinoma(P0.05),however it was associated with lymph node metastasis(P0.05).The expression of PTTG was positively correlated with the expression of P53(r=0.495,P).ConclusionThe expressions of PTTG and P53 are associated with the progression and metastasis of colorectal carcinoma,and PTTG and P53 may be used as molecular markers for judging the metastasis potential and the invasion of colorectal carcinoma.

To investigate the microRNA101(miR101) expression and its clinical significance in colorectal cancer.Tissue microarrays containing 56 specimens of normal mucosa, 51 adenoma and 735 colorectal cancer were examined by locked nucleic acid in-situ hybridization(LNA-ISH) for miR101 expression. Relationship between miR101 expression and clinicopathologic parameters and prognosis of colorectal cancer patients were analyzed. Fresh frozen tissues containing 5 specimens of normal mucosa, 5 adenoma and 47 colorectal cancer were examined by RT-PCR to verify the accuracy of LNA-ISH.Expression of miR101 increased gradually from normal mucosa, adenoma to stage I colorectal cancer (all P<0.01), and decreased gradually from stage II(, stage III( to stage IIII( colorectal cancer (all P<0.01). Overexpression of miR101 was related with lower incidence of lymph node metastasis, lower metastasis rate, higher differentiation and lower recurrence rate (all P<0.01). Multivariate survival analysis demonstrated that miR101 expression was an independent prognostic factor of overall survival (HR=0.550, 95% CI: 0.351-0.863) and disease free survival(HR=0.562, 95% CI: 0.397-0.794) in colorectal cancer patients. Overexpression of miR101 predicted a better prognosis in colorectal cancer patients.Expression of miR101 is associated with the genesis and development of colorectal cancer, and may serve as an independent prognostic factor for colorectal cancer patients.
