PENGARUH PENAMBAHAN GLUTARALDEHID SEBAGAI CROSSLINKAGENT TERHADAP PELEPASAN NATRIUM DIKLOFENAK DARI SCAFFOLD KITOSANGELATIN(dibuat dengan metode pre-freezing)

Scaffold used as replacement of defective cartilage and a biodegradable drug delivery system (DDS) for the administration of diclofenac sodium in treatment of Osteoarthritis (OA). The materials, are exclusively composed of diclofenac sodium, matrix chitosan gelatin PEG- 400. In order for scaffold to release the medicine slowly and controllable, thus it should be done cross connection which is adding glutaraldehyde 0- 2.5% concentration towards matrix of (chitosan and gelatin) which is mixed with analgesic medicine (diclofenac sodium). After it is shaped scaffold is dried by freezing method as long as twenty-four hours, then it can be done release test by using UV-VIS spectrophotometer and using Phosphate Saline Buffer media pH 7.4 ± 0.05. The result showed that diclofenac sodium from chitosan-gelatin scaffold had sustained release in 542 hours and the release was following zero order kinetic. Based on the results of analysis with Kruskal Wallis statistical test, it was found that there was a significant effect on the addition of Glutaraldehyde concentration to cumulative amount (Cumulative Du) of diclofenac sodium.
