基因枪介导抗除草剂基因2mG2(上标 -)epsps转化玉米的初步研究

Herbicide resistance 2mG2(superscript -) epsps gene was introdued into immature embryo callus of the elite maize inbred line X2211 and 78599 using microprojectile bombardment. After selection on media supplemented with different concentration glyphosate, 320 regeneration plants were regenerated. 65 of them exhibited tolerationto glyphosate excellently for herbicide resistance. It was demonstrated that the 2mG2(superscript -) epsps gene had been intergraded into 7 plants genome and expressed in transgenic plants by PCR analysis. The average transformation frequency is about 2.2%. At the same time, we studied the golden powder concentration and the shoot-distance affecting transformation efficiency in microprojectile bombardment system of maize. Considering the transformation efficiency, we think it is suitable that the golden powder concentration is 100μg/P and the shoot-distance is 60mm.
