Effects of CKZ on Th1/Th2 cytokine repertoire of human peripheral blood mononuclear cells

Objective:To study the immune regulation mechanisms of CKZ by investigating its effects on Th1/Th2 cytokine repertoire of human peripheral blood mononuclear cells(PBMCs).Methods:The concentrations of Th1(IFN-γ,TNF-α,IL-2) and Th2(IL-4,IL-6,IL-10) cytokines secreted by PBMCs were detected by means of cytometric bead array(CBA).Results:Cytokines secreted by PBMCs from healthy persons were mainly TNF-α and IL-6 after 12 hours of culture, and both Th1 and Th2 cytokines were increased by CKZ. Secretion of both Th1 and Th2 cytokines stimulated by PDB plus ionomycin were inhibited by CKZ, and CKZ could also inhibit the abnormally increased secretion of INF-γ,TNF-α,IL-6 and IL-10 in patients with influenza.Conclusion:CKZ can up-regulate Th1 and Th2 cytokines secreted by PBMCs from healthy person, and down-regulate Th1 and Th2 cytokines secreted by abnormally activated PBMCs.
