Comparison of Three Detectors Used for HPLC Determination of Monoglycerides

Comparison of HPLC coupled with each of the detectors ultraviolet detector(UV),differential refractive index detector(RI) and evaporative light scattering detector(ELSD) was carried out for the determination of monoglycerides.Serious interference from impurities,low signal intensity and weak sensitivity were observed when using UV detection.However,RI detection showed good linearity over a high concentration range despite not being suitable for the detection of low-concentration samples.Similarly,ELSD detection exhibited excellent linearity over the concentration range of 0.040 to 5.000 mg/mL,with a correlation coefficient of 0.9998;the recovery of ELSD detection was 92.94%,with a relative standard deviation of 2.24% and a coefficient of variation of 2.41%.HPLC-ELSD was sensitive,accurate and repeatable and applicable for the determination of monoglycerides in emulsion.
