Structure of C3PO and mechanism of human RISC activation

Generating a complex competent for RNA interference requires cutting and removal of the non-guide siRNA strand. Reconstitution of human RISC activity now shows that C3PO is an endonuclease critical for human Ago2-RISC activation and suggests that the C3PO consists of an octamer with active sites located on the convex interior.
