Preparation of Chitosan Derivative Based Molecular Imprinted Polymer and Its Recognition for Protein

Chitosan derivative with aqueous solubility and containing polymerisable and crosslinkable functional groups were synthesized through chemical modification using chloroacetic acid and glycidyl methacrylate (GMA). FTIR analysis showed that GMA reacted with carboxymethyl chitosan(CMCS)by a ring opening reaction and the resultant(CMCS-GMA)could polymerize using ammonium persulphate as initiator. Thin films of CMCS-GMA polymer were formed on the surface of polystyrene microplates and were applied to imprint haemoglobin (Hb) in aqueous medium. The imprinted chitosan derivative has much higher binding capacity for Hb than the non-imprinted polymer with the same chemical composition. Protein assay of lysozyme, a structural analogue of haemoglobin, showed that the imprinted polymer also exhibits a higher selectivity for the imprinted molecule.
