Research on cell toxicity of chrysotile,nano-sil-ica,wollastonite and 3 kinds of artificial miner-al fibers

The present research has enabled us to disclose the sur- vival rate and morphological changes of the Chinese Hamster lung cell (V79),which are exposed to different mineral dusts,such as those of chrysotile,nano-silica,wollastonite and the other three kinds of synthetic mineral fibres when letting them exposed for 12 h,respec- tively.The three kinds of synthetic mineral fibres are ceramic fibre, glass fiber and rock-made wool.The cell survival rates were tested and detected by using methyl thiazolyl tetrazolium (MTI)colorimetry assay while the morphological changes of V79 cell were observed by Wright-Giemsa dyeing assay.The results of experiments enable us to find that the survival rate of V79 cell is decreased,for the prolifera- tion significantly is inhibited,but the amount of necrosis cell is in- creased,which reveals the dose effect relations.The inhibition grades of mineral dusts can be stated as follows:nanometer SiO_2 glass fiber chrysotile ceramic fibre wollastonite rock wool. According to morphological observation rule on fie inhibited cell,it was found that exposed to the dust of nano-silica,the normal shape of cell is difficult to be discovered while the morphological changes of V79 are the most serious one among the six kinds of dust.In addi- tion,it is found that exposed to the dust of rock wool,the cell mor- phological changes are by no means significant with the most cells re- maining with their normal shape.The cell morphological changes grades can be divided into six kinds of mineral dusts,that is, nanometer SiO_2 glass fiber ceramic fibre wollastonite
