Genetic transformation of Alhagi preudoalhagi with Agrobacterium rhizogenes A4

By using Agrobacterium rhizogenes A4 with Ri-plasmid, the transformation test was carried out with the cotyledons and hypocotyls of Alhagi pseudoalhadi. Transgenic plants of Alhagi pseudoalhagi were regenerated from aseptic calluses derived from hairy roots transformed by Agrobacterium rhizogenes strains A4 at or near the site of infection of cotyledon and hypocotyl segments. Both agropine and mannopine were detected not only in extract of the primary hairy roots, but also in those of the secondary hairy roots, callus derived from hairy roots and leaves of regenerated plantlets. Compared with hypocotyl segments, the time of hairy root induction from cotyledon segments was shorter and the frequency of transformation was higher.
