FORMULATION AND EVALUATION OF BIOADHESIVE PATCHES OF DEXTROMETHORPHAN

2012
The buccal route as an alternative to other traditional method of systematic drug administration is a subject of growing interest because of numerous advantages. Oral mucosa provide a direct entry of the drug into the systemic circulation, there avoiding the first pass hepatic metabolism and gastrointestinal drug degradation which is associated with oral administration. Drug like Dextromethorphan has been selected as model drug because the drug has all the pharmacokinetics and physicochemical properties required for controlled release. To formulate buccal patches of dextromethorphan using various polymers like HPMC 4000, HPMC K 15M, PVA, Carbopol – 934, Chitosan and PVP. Prepared by solvent casting method and evaluated the patches for their physical parameters like appearance, thickness of 0.147 ± 0.009mm - 0.247 ± 0.009mm, weight uniformity, surface texture, folding endurance, drug content, surface pH6.5 ± 0.08 - 6.6 ± 0.08, swelling index, tensile strength, bioadhesive strength 3.75 ± 0.019 Kg/Cm 2 - 7.60 ± 0.018
