Genetic transformation of indica rice 9311 with antisense Waxy gene

The indica rice variety 9311 was used to study the inductions of different culture mediums on it,and the impact of Agrobacterium concentration,infected time and drying time on confrontational callus. Then by applying co-transformation method,plasmid p13W8 which contained antisense Waxy gene and plasmid p1300 which contained marker gene were transformated into calli induced from matural embryos by Agrobacterium tumefaciens. The results show that N6 medium is the best suitable medium for callus inducing from 9311. The suitable concentration of Agrobacterium is OD600=0.6 and the incursion time is 10 min,with 1.5~2.0 h desiccation after the incursion. And PCR analysis show that the antisense Waxy gene has been integrated into the genome of the regeneration plants.
