The present invention relates to a preparation method, a purification method and an identification method of neural stem cell-derived Exosomes, and an application of the neural stem cell-derived Exosomes in preparation of nervous system disease treatment drugs, wherein the Exosome-derived neural stem cells belong to the suspension type stem cells, and comprise mouse neural stem cells and neural stem cells induced from mesenchymal stem cells. The preparation steps comprise: under a sterile condition, collecting complete culture medium supernatants of 3-10 generations of various types of stem cells, carrying out centrifugation for 20 min at 20000 g, collecting the supernatant, adopting a 0.22 mum sterile filtration membrane to filter so as to remove impurities, carrying out centrifugation on the obtained liquid for 60 min at 120000 g, removing the supernatant, taking the precipitate to obtain the Exosomes, and adopting an immune magnetic bead method to carry out a purification treatment, wherein the Exosomes adopted as the cystic bodies secreted by the stem cells, having the complete cell membrane and carrying various types of active proteins and signal molecules in the cytoplasm and the cell membrane can partially or completely provide the effect of the neural stem cells, and can be provided for treatment of nervous system diseases. In addition, the new treatment approach and technical support is provided for nervous system disease treatment through replacement of the neural stem cells.
