Effects of Traditional Chinese Drug Fangxiaoyin Decoction Early Intervention on Interleukin-5 and Interleukin-12 Levels of Blood Serum in Mice with Asthma

Objective To investigate the effects of traditional Chinese drug fangxiaoyin decoction on interleukin-5(IL-5),interleukin-12(IL-12) levels of blood serum and eosinophil(EOS%) in bone marrow in allergic asthmatic mice.Methods Fifty healthy mice were randomly divided into 5 groups: normal control group;asthmatic group;low dose fangxiaoyin decoction treated group;high dose of fangxiaoyin decoction treated group and budesonide(BUD) treated group.The asthmatic mouse models were stimulated with ovalumin(OVA)administrated once from the first day of the sensitization.The drug,fangxiaoyin decoction was given in a small dosage of 500 g·L-1 crude drug,and big dosages of 2 500 g·L-1 crude drug by enteroclysis,once each day,0.4 mL every time,26 days all together.In the last time the mice were slaughtered after 48 h,and double antioxidant sandwich enzyme-linked immunosorbent test was detect the serum IL-5,IL-12,and the content of bone marrow EOS% microscopically.Results Bone marrow EOS% in asthma model group[(8.55±1.55)%]increased significantly compared with that in control group[(3.90±0.94)%](P0.01),IL-5 levels [(25.69±4.75) ng·L-1]increased significantly compared with that in control group[(17.50±4.71) ng·L-1](P0.05),IL-12 content[(65.72±8.90) ng·L-1]in asthma model group compared with that in control group[(81.60±9.40) ng·L-1](P0.01).IL-12 levels improved and serum IL-5 levels and marrow EOS %(Pa0.01) reduced significantly in the high dosage fangxiaoyin decotion group and BUD group,but the difference was not statistically different in both groups by comparison(Pa0.05).Low-dose fangxiaoyin decoction treated group had no marked improvement compared with the high-dose fangxiaoyin decotion treated group.Conclusions High-dose fangxiaoyin decotion treated group accommodate the balance of Th1 and Th2 by depressing IL-5 and lifting IL-12 levels for the purpose of treating asthma.
