The Effect of three-dimensional matrices on HLA expression by amniotic epithelial cells

Amniotic epithelial cells (AECs) with stem cell characteristics and unique immunological properties have created new hopes in regenerative medicine. Having differentiation potential into three germ layer cell type, AE cells are a precious cell source for clinical application. However, immunological changes of AE cells after culture and differentiation have limited their potential applications. The aim of this study was to determine matrix dimension effects on HLA (Human Lymphocyte Antigen) expression by these cells. Chitosan-gelatin three-dimensional (3D) (Scaffold) and two-dimensional (2D) matrices were used to evaluate MHC class I, HLA-DR and HLA-G expression in AE cells after 7 days culture. The results confirmed the effect of 3D and 2D systems on AECs immunogenicity especially on MHC class I (HLA-A, B, C) expression.
