Differential expression of cancer stem cell surface markers in human laryngocarcinoma and the cell lines

OBJECTIVE To compare the expression of CD44ABCG2 and CD133 in laryngocarcinoma tissue from patients and laryngocarcinoma cell lines Hep2、LSC-1,to analyze the variability of expression among them,thus to make a approach to the value of the above-mentioned antibodies on screening cancer stem cells in laryngocarcinoma.METHODS By using laryngocarcinoma tissue from patients and laryngocarcinoma cell lines Hep2、LSC-1 as object,then labeling cells with CD44ABCG2 and CD133,we analyzed the expression of the above-mentioned antibodies in laryngocarcinoma cells on FACSAriaTM Cell Sorter.RESULTS Distinct populations of CD44 were identified in solid tumors of laryngocarcinoma patients with strong individual deviations,constitutively expressed on the surface of laryngocarcinoma cell lines analyzed.There were significant difference between the CD44 expression in laryngocarcinoma tissue and that in the cancer cell lines.ABCG2 expressed positively in laryngocarcinoma tissue,dim positively in Hep-2,but intensive positively expressed almost in all LSC-1 cells.And there was no significant difference between the expression of ABCG2 in laryngocarcinoma tissue and that in Hep-2.CD133 expressed positively in laryngocarcinoma tissue,dim positively in both cancer cell lines.and there was no significant difference for the expression of CD133 between laryngocarcinoma tissue and the cancer cell lines.CONCLUSION The expression of CD44ABCG2 and CD133 makes a conspicuous difference between laryngocarcinoma tissue and laryngocarcinoma lines.
