THE EFFECTS OF GINKGOLID B OF VARIOUS CONSISTENCY ON THEDIFFERENTIATION OF NEURAL STEM CELLS

Objective To observe the effects of Ginkgolid B of various consistency on the differentiation of neural stem cells. Methods Neural stem cells(NSCs) were dissociated and cultured from the hippocampus of newborn rat.The NSCs plated into 24 well cultured plates were divided into 20mg/L,40mg/L,60mg/L Ginkgolid B groups and control group.Four groups of NSCs were cultured respectively using DMEM/F12 containing Ginkgolid B of various consistency.At 7 days and 14 days after NSCs cultured, NF, GFAP and Oligodendrocyte special protein(Oligo) expression were detected using immunocytochemical stains respectively.The numbers of NF,GFAP and Oligo positive cells were counted.The percentages of positive cells were obtained respectively. Results The percentage of NF positive neuron-like cells in the various consistency of Ginkgolid B groups was significantly higher than that in the control group,but there were no significantly differents among the various consistency of Ginkgolid B group.The effect of Ginkgolid B of various consistency on NSCs differentiation of Oligo positive cells was insignificant.The percentage of GFAP positive cells in the various consistency of Ginkgolid B groups was significantly higher than that in the control group,and the percentage was increasing with the consistencies of Ginkgolid B.Conclusion Ginkgolid B may enhance NSCs differentiate into NF positive neuron-like cells,but it is no relation to the consistency of Ginkgolid B.Ginkgolid B may not promote NSCs differentiate into oligodendrocyte-like cells.Ginkgolid B may enhance NSCs differentiate into GFAP positive astroglia-like cells and the percentage of GFAP positive cells were increased with the consistencies of Ginkgolid B.
