Influences of mesenchyme stem cell transplantation on myocardial collagen and norepinephrine transporter in ischemia/reperfusion rats

Objective To observe the influences of myocardial transplantation of bone mesenchyme stem cells(BMSCs) on left ventricular remodeling and expression of norepinephrine transporter(NET) in rat model of 60-minute ischemia/reperfusion myocardial infarction(MI),and investigate the effective mechanism of BMSCs transplantation to cardiac sympathetic function and collagen remodeling.Methods BMSCs were separated for primary culture in Wistar rats,model of ischemia/reperfusion MI was established and randomly divided into sham-operation group,MI group and BMSCs transplantation group.The indexes of hemodynamics were observed after BMSCs transplantation.The range,area and expansion index of MI were determined after Sirius red staining.The collagen volume fraction and collagen maturity in MI area were analyzed combined polarization analysis.The expression of NET was detected by using immunohistochemistry technique.Results ① BMSCs transplantation reduced MI area in 60-minute ischemia/reperfusion rats.② BMSCs transplantation relieved the left ventricular remodeling in MI rats.③ BMSCs transplantation inhibited extracellular matrix,decreased myocardial collagen content and collagen volume fraction,and improved the expression of myocardial NET.Conclusion BMSCs transplantation can relieve post-MI ventricular remodeling,reduce myocardial collage content and improve the expression of myocardial NET.
