Tracking Marrow Mesenchymal Stem Cells by Transfection with Enhanced Green Fluorescent Protein

Objective: To develop an appropriate tracing method for transplanted marrow mesenchymal stem cells(MMSCs) in vivo.Methods: MMSCs of rats were isolated from bone marrow and purified by adherent culture in vitro,and the cell membrane's marks were detected with flow cytometer.Cultured cells were transfected with expressing plamid of pEGFP-C1 mediated by lipofectamine,and transient expression was subsequently observed with fluorescent microscopy.Results: The expression of pEGFP-C1 was initially found in 24h after the transfection,reached peak in 48~72 h,and remained strong express for 12~14 more days.Conclusion: Lipofectamine mediated transfection can make pEGFP-C1 expressed in MMSCs safely and effectively,which shows that it is an ideal method for stem cell tracking.
