不同配比组方的百部–紫菀药对咳嗽变异性哮喘大鼠Th1/Th2及Th17/Treg细胞因子平衡影响 Study on the Effects of Couplet Medicine Baibu-Ziwan with Different Proportions on Cytokines Level of Type Th1/Th2 and Th17/Treg in Cough Variant Asthma Rats

目的:探讨药对百部–紫菀平喘作用机制并探索配伍比例对药对的药效作用影响。方法:建立SD大鼠咳嗽变异性哮喘(CVA)模型,灌胃百部–紫菀不同配伍比例提取物,采用Elisa方法检测支气管肺泡灌洗液(BALF)中IL-4,INF-γ,IL-17及IL-10含量,并测定BALF中的炎性细胞变化,比较组间差异。结果:药对百部-紫菀给药后,CVA大鼠Thl/Th2及Th17/Treg水平失衡均得到部分纠正,同时嗜酸性粒细胞、淋巴细胞、中性粒细胞等炎性细胞水平明显降低,统计学显示具有显著性差异。配伍比例对药效的作用具有一定程度的影响,配伍1:1时的药效作用最强。结论:药对百部–紫菀对CVA大鼠的平喘机制与纠正Thl/Th2与Th17/Treg细胞平衡,减轻炎性细胞浸润有关,配伍比例对药对的药效作用具有一定程度的影响。 Objective: To study the antiasthma mechanism of couplet medicine Baibu-Ziwan and investigate the pharmacodynamic effects on the proportion compatibility. Method: Cough variant asthma (CVA) rats were established and orally administrated by the extractions of Baibu-Ziwan with dif-ferent compatibility. The IL-4, INF-γ, IL-17 and IL-10 levels in bronchoalveolar lavage fluid (BALF) were determined by Elisa method and the numbers of inflammatory cells in BALF changes in lung were measured, respectively. The differences were tested among the groups by SPSS 19.0 software. Results: The levels of Thl/Th2 and Th17/Treg were both corrected after administration of the extractions of Baibu-Ziwan with inflammatory cells (eosinophil, lymphocytes and neutrophil) decreased in CVA group (p < 0.05). Conclusion: The antiasthma mechanism of couplet medicine Baibu-Ziwan on CVA rats may be related to the regulating imbalance of Thl/Th2 and Th17/Treg and the decreased infiltration of inflammatory cells, and the proportion compatibility had some influences on its pharmacodynamic effects.
