Study of miR - 7 reversing the malignant phenotype of U251 glioma mediated by folic acid /polyamide - amine vector

Objective To explore the gene therapy effect of miR -7 mediated by folie acid /polyamide - amine vector on human U251 glioma. Method After transfeeting FA/PAMAM/miR - 7 gene into U251 glioma cell line, the cell growth curves were drawn, the cell cycle was detected, and apoptosis was detected by apoptosis in situ assay. Then the transwell system was utilized to explore the migration ability of tumor cells, and soft agar colony assay to examine the tumorigenicity. The quantitative RT - PCR was used to detect the miR -7 level, and western blot method was used to test the protein expression of EGFR,PI3K and AKT2. Results The cell proliferation rate slowed down, as well as theirs proportion reduction in S phase cells, and the ratio of apoptosis. The cell migration and soft agar colony forming ability in U251 cells transfeeted by miR - 7 were significantly reduced than control group ( P 〈 0.05 ). With the increase of the miR - 7 level, the protein level of EGFR, PI3K and AKT2 in FA/PAMAM/miR - 7 group were much lower than the control group ( P 〈 0. 05 ). Conclusions FA/PAMAM can transfect effectively miR- 7 into glioma, play a role to inhibit glioma cells proliferation. Key words: Glioma;  microRNA ;  Genes ;  Receptor, epidermal growth factor
