Expression of corticotropin-releasing hormone and corticotropin-releasing hormone type 1 receptor in rat hypothalamic slice after stimulation with corticotropin-releasing hormone stimulation in vitro

Objective To investigate the distribution and expressions of corticotropin-releasing hormone (CRH) and corticotropin-releasing hormone type 1 receptor (CRH1R) in rat hypothalamus after CRH stimulation of hypothalamic slices in rats in vitro. Methods Model of rat hypothalamic slices was employed in this study. The distribution and expressions of corticotropin-releasing hormone and corticotropin-releasing hormone type 1 receptor in rats hypothalamus after CRH stimulation of the hypothalamic slices in rats in vitro were observed by immunocytochemical method. Results An increased intracellular concentration of CRH and CRH1R in the hypothalamus was observed after CRH stimulation of the hypothalamic slices in rats. CP-154526 or H89 significantly inhibited the expressions of CRH and CRH1R. Conclusion An increased concentration of CRH conjugated with CRH1R can induce more expressions of CRH and CRH1R in rats hypothalamus after CRH stimulation of the hypothalamic slices in rats in vitro, suggesting that the expression of CRH in hypothalamic neuron may have regulatory effect on the ultrashort positive feedback through PKA pathway.
