To investigate the effect of small interfering RNA (siRNA) silencing Apollon gene combined with tetramethylpyrazine (TMP) on the proliferation and apoptosis of human chronic myeloid leukemia cell line K562.K562 cells were divided into blank control, negative control, and RNA interference (RNAi) group. For the RNAi group, the pGPHI-GFP-Neo-Apollon eukaryotic expression vector based on the best Apollon siRNA fragments screened out in previous experiments was constructed; the blank control group received no treatment, and the negative control group was transfected with negative plasmid vector. The mRNA and protein expression of Apollon was measured by RT-PCR and cell immunofluorescence, respectively. Additionally, TMP (320 μg/mL) was applied to set TMP, TMP+negative control, and TMP+RNAi groups. The cell viability and apoptosis rate were determined by MTT assay and flow cytometry, respectively.The constructed vector was stably expressed in K562 cells. The RNAi group had significantly lower mRNA and protein expression of Apollon than the blank control group and negative control (P<0.05). The RNAi group had significantly increased proliferation inhibition rate and apoptosis rate, as compared with the blank contorl group (P<0.05). The TMP+RNAi group had significantly increased proliferation inhibition rate and apoptosis rate, as compared with the RNAi, and TMP groups (P<0.05).Apollon siRNA can significantly inhibit the proliferation and promote the apoptosis of K562 cells, and the addition of TMP can further increase the proliferation inhibition rate and apoptosis rate, suggesting that siRNA technology combined with drugs has a significant potential value in the treatment of leukemia.
Objective
To assess the use of bipolar radiofrequency device Habib 4X in hepatic malignant tumor resection.
Methods
Clinical data of 75 patients with hepatic malignant tumor undergoing hepatectomy with the use of Habib 4X in Department of Gastrointestinal Oncology, Cancer Center of Guangzhou Medical University from February 2009 to June 2013 were retrospectively analyzed. There were 55 males and 20 females with a mean age of (53±12) years old. Forty-nine cases were with primary liver cancer and 26 cases were with secondary liver cancer. The informed consents of all patients were obtained and the ethics committee approval was received. Patients received endotracheal general anesthesia. Hepatectomy was performed after tumor ablation was employed with Habib 4X device 2-3 cm away from the tumor. The intraoperative bleeding, blood transfusion, operation duration, changes of postoperative alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TB), length of postoperative hospital stay and complications were observed. Patients were followed up after being discharged from hospital and tumor recurrence and mortality were observed.
Results
All the 75 patients underwent hepatectomy successfully. The mean intraoperative bleeding was (318±89)ml, the rate of blood transfusion was 7% (5/75), the median of operation duration was 104 (17-191) min, the length of postoperative hospital stay was (11±4)d. ALT, AST, TB rose to a top level 1 d after operation, then went down progressively and liver function backed to normal 9 d after operation. Two cases suffered from bile fistula and were cured after enhancing drainage. The 1-, 3-year recurrence rates were 6% (3/49), 10% (5/49) respectively in patients with primary liver cancer and were 23% (6/26), 27% (7/26) respectively in patients with secondary liver cancer. The mortality were 4% (2/49), 27% (7/26) respectively in patients with primary liver cancer and secondary liver cancer during the follow-up. Two cases with primary liver cancer died of postoperative hepatic encephalopathy and 7 cases with secondary liver cancer died of tumor widespread metastasis.
Conclusions
Habib 4X is safe and effective for hepatic malignant tumor resection. It may reduce the postoperative tumor recurrence rate.

This paper studies the correlation between biogas output and the changes of several hydrolases during the anaerobic digestion of chicken manure. The results suggest that the enzyme activities of cellulase, lipase, #alpha# amylase and #gamma# amylase change during the anaerobic digestion, which is associated with biogas output. The maximum value of hydrolase activity appears colse to the peak biogas producing period while the activity of acid protease, base protease and neutral protease keep decreasing.
The paper has been designed based on the experimental survey and questionnaires to 797 farmers, 25 technicians in 41 villages, and 8 towns of Yuxi region, Yunnan, China—to analyze the universal problems in biogas utilization process, including (a) low biogas utilization rate, critical dysfunctional phenomenon; (b) poor comprehensive utilization of biogas, bio-slurry, and residue; and (c) imperfect post-installation service system. Combined with the investigated findings, the article has analyzed the main causes and influencing factors which lead to the different problems. In the end, having based on the adequate interpretation about the studying conclusion, recommendations and countermeasures for the future sustainable development of China's rural household biogas proper measures can be put forward onto consideration table, which includes (a) encourage joint household biogas construction; (b) carry out comprehensive utilization demonstration actively; (c) establish a relatively complete post-installation service system by encouraging technicians to get involved in the maintenance and management initiations. The establishment of incentive systems and co-operative approaches can help to achieve the ultimate target of appropriately "biogas-designed" that are sustainably developed and constructed in China.

The author also points out that improving the efficiency of trademark utilization can help promote economic restructure and the mode of economic growth transformation.
Objective To obtain effective siRNA fragment of HOXA10 gene and verify its function,to supply experimental evidence for tumor prevention and curation by RNAi targeting to HOXA10 gene.Methods Three pairs of small interfering RNA targeting to the different sites of HOXA10 were designed and introduced into A549.The mRNA expression of HOXA10 of A549 was detected by semi-quantitative RT-PCR,the cell proliferation was assayed by MTT,the apoptosis was measured by flow cytometry.The most effective siRNA assay was screened and was tested the relationship between it and proliferation and apoptosis.Results The mRNA of HOXA10 was inhibited by three siRNAs in A549 cells,among which siRNA1 gave the strongest inhibiting of HOXA10 by ODR was (20.190±1.698) %.The inhibitory rate of cell proliferation was (69.793±2.092) % and the apoptosis rate was (29.593±2.670) %.Conclusion siRNA1 can specifically degrade HOXA10 mRNA and inhibit the proliferation of A549 cell and promote its apoptosis.

This research is intended to explore H2 generation capacity of the fermentation using different organic wastes-dung of pig,cow,horse and chickens,and stalks of all kinds of crops from rice to bean.With the batch fermentation process in a lab scaled apparatus,in which temperature is controlled,and pH is adjusted with lactic acid,comparative experiment is conducted.Result shows pig dung tops all the animal dung while barley stalk is best among the all crops tested in H2 production.
To investigate the relationship between the long noncoding RNA (lncRNA) Prostate cancer-associated transcription factors 14 (PCAT14) and the clinical characteristics of prostate cancer and immune cell infiltration.The relationship between PCAT14 expression and the clinicopathological characteristics of prostate cancer was analyzed based on The Cancer Genome Atlas (TCGA) database. Receiver operating characteristic (ROC) curves were used to evaluate the value of PCAT14 as a diagnostic marker for prostate cancer. The relationship between PCAT14 and immune cell infiltration was analyzed to explore the effect of PCAT14 on the immune-related functions of prostate cancer.The ROC curve showed that PCAT14 had a significant diagnostic ability (area under curve = 0.818) for prostate cancer. A reduced expression of PCAT14 in prostate cancer was related to T stage, N stage, primary therapy outcome, residual tumor, Gleason score, and age. The expression of PCAT14 was independently associated with the progression-free interval in prostate cancer patients. The infiltration of immune cells in prostate cancer showed a significant negative correlation between the expression of PCAT14 and plasmacytoid dendritic cells, activated dendritic cells, regulatory T cells, and neutrophils.PCAT14 is highly expressed in prostate cancer and is expected to be a diagnostic marker. PCAT14 might promote the development of prostate cancer through chemokines, antimicrobials, and cytokines that affect the infiltration of immune cells.
Based on the early works of winery waste water treatment with new anaerobic filter(AF).The effectiveness of six different new upflow anaerobic filters for treating the pretreatment winery waste water at 25 ℃ invariable temperatures was discussed.The devices have the same types of soft and hard packing media,but different media arrangement.The results showed that when influent concentration was about 3 000 mg/L,the best HRT was 24 h,and organic loading rate(OLR)was 3.75 kg COD/(m3·d).In that situation,with the soft media on the underneath,hard media was the top of the reactors;the optimal volume ratio of soft and hard packing media was 2∶1.The density of soft and hard packing media was 13.7 kg/m3,85 kg/m3 respectively.COD removing rate was 92.12%,suspended solids(SS) was 72.65%,the biogas recovery rate was 0.133 m3/kg CODremoved in this reactor.
