Generation of core-shell microcapsules with three-dimensional focusing device for efficient formation of cell spheroid

We present a microfluidic device generating three-dimensional (3D) coaxial flow by the addition of a simple hillock to produce an alginate core-shell microcapsule for the efficient formation of a cell spheroid. A hillock tapered at downstream of the two-dimensional focusing channel enables outside flow to enclose the core flow. The aqueous solution in the core flow was focused and surrounded by 1.8% alginate solution to be solidified as a shell. The double-layered coaxial flow (aqueous phase) was broken up into a droplet by the shear flow of oleic acid (oil phase) containing calcium chloride for the polymerization of the alginate shell. The droplet generated from the laminar coaxial flow maintained a double-layer structure and gelation of the alginate solution made a core-shell microcapsule. The shell-thickness of the microcapsule was adjusted from 8-21 μm by the variation of two aqueous flow rates. The inner shape of the shell was almost spherical when the ratio of the water-glycol mixture in the core flow exceeded 20%. The microcapsule was used to form a spheroid of embryonic carcinoma cells (embryoid body; EB) by injecting a cell suspension into the core flow. The cells inside the microcapsule aggregated into an EB within 2 days and the EB formation rate was more than 80% with strong compaction. The microcapsule formed single spherical EBs without small satellite clusters or a bumpy shape as observed in solid microbeads. The microfluidic chip for encapsulation of cells could generate a number of EBs with high rate of EB formation when compared with the conventional hanging drop method. The core-shell microcapsule generated by 3D focusing in the microchannel was effective in forming large number of spherical cell clusters and the encapsulation of cells in the microcapsule is expected to be useful in the transplantation of islet cells or cancer stem cell enrichment.

Ferroelectric electroluminescent comb copolymer for single-material self-powered displays

Ferroelectric polymers have recently been applied in human-connected electronics as pressure (touch)-sensing materials to develop high-performance electronic skin and tactile sensing memory. Here, we report an organic synthetic route for developing a polymer possessing both ferroelectric and electroluminescent properties from which a self-powered pliable display can be readily implemented. The synthetic route involves reversible addition-fragmentation transfer-mediated graft copolymerization of poly(vinylidene fluoride) (PVDF) onto a polyfluorene (PFO) backbone, which results in a comb-like copolymer architecture composed of ferroelectric side chains (PVDFs) tethered to a light-emitting main chain (PFO). The resultant thin comb copolymer film, equipped with hardly integrable three natures (i.e., ferro- and piezoelectricity, luminescence, pliability), exhibits excellent light emission under alternating current and self-powering attributes upon mechanical deformation. This multifunctional polymer, where various properties including ferroelectricity and electroluminescence are imparted in molecular-level precision, envisions its use in a wide range of fields such as emerging self-powered interactive displays.

본 연구에서는 영유아기 자녀를 둔 어머니의 감각처리민감성 총점 및 하위요인인 저감각역, 자극흥분, 심미적 민감성이 양육스트레스에 미치는 영향에서 사회적 지지의 조절효과를 검증하였다. 이를 위하여 만 1세부터 6세 이하의 자녀를 둔 어머니 254명을 대상으로 한국형 감각처리민감성 척도, 한국판 부모 양육스트레스 검사 4판 단축형(K-PSI-4-SF), 사회적 지지 척도를 실시하였으며 수집된 자료는 SPSS 26.0 통계 프로그램을 이용하였고 위계적 회귀분석을 실시하여 변인 간 관계 및 조절효과를 검증하였다. 그 결과는 다음과 같다. 첫째, 어머니의 감각처리민감성 총점과 저감각역, 자극흥분 수준이 높으면 양육스트레스가 높았으며 심미적 민감성 수준이 높으면 양육스트레스가 낮았다. 둘째, 어머니의 감각처리민감성 총점과 하위요인 중 자극흥분과 심미적 민감성이 양육스트레스에 미치는 영향에서 사회적 지지가 유의한 조절효과를 보였다. 구체적으로 어머니의 사회적 지지 수준이 높고 어머니의 감각처리민감성 총점과 자극흥분이 높은 경우 양육스트레스 수준이 높게 나타났다. 반면, 심미적 민감성이 높은 경우 양육스트레스 수준이 낮게 나타났다. 본 연구는 어머니의 감각처리민감성과 양육스트레스의 관계에서 사회적 지지의 조절효과를 구체적으로 검증하여 어머니의 감각처리민감성을 이해하고 양육스트레스 감소할 수 있는 기초자료를 마련하였다는 데 의의가 있다.

Three dimensional co-culture of neuron and astrocyte in a micro-fluidic device

In this paper, we describe a microfluidic platform that enables three dimensional cell culture within defined microenvironments towards a study of axon-glia interaction. The microdevice can offer both 3D axon isolation from cell body and 3D spatial cell separation between neuron and astrocyte. It consists of large fluidic channels for media supply and small channels for collagen filling as a three dimensional ECM (extracellular matrix). Astrocytes were seeded in collagen gel through small fluidic channels and neuron cells were aggregated at an openings of small channel to large channels. This method separated neuronal soma from axon as well as segregatedneuron body from astrocyte. The preliminary study on the effect of astrocyte to the axon growth showed that axon growth was faster and richer in co-culture condition without growth factor.This 3D co-culture chip can provide a new method to observe the effect of astrocyte on the axon growth in three dimensional culture system.

Two-Layer Nonlinear FIR Filter and Unscented Kalman Filter Fusion With Application to Mobile Robot Localization

In this paper, we propose a new state estimator called the two-layer nonlinear finite impulse response (TLNF) filter and adopt this new filter and unscented Kalman filter (UKF) as subfilters to create the fusion TLNF/UK filter. The TLNF filter is constructed with measurements that are redefined by weighting the estimated states acquired through minimizing the cost function based on the Frobenius norm. The efficient iterative form of the TLNF filter is also developed in this paper. Using the fact that the UKF and the TLNF filter each takes a different type of memory structure, the fusion TLNF/UK filter is designed as a robust nonlinear state estimator taking both advantages of each filter. To obtain the best fusion estimates, probabilistic weights are computed based on Bayes' rule and the likelihood of each filter. Both simulation and experimental results for mobile robot indoor localization have shown that the fusion TLNF/UK filter achieves a higher level of accuracy and robustness under practical situations.
