Serine Rejuvenated Degenerated Volvariella volvacea by Enhancing ROS Scavenging Ability and Mitochondrial Function

Serine is a functional amino acid that effectively regulates the physiological functions of an organism. This study investigates the effects of adding exogenous serine to a culture medium to explore a feasible method for the rejuvenation of V. volvacea degenerated strains. The tissue isolation subcultured strains T6, T12, and T19 of V. volvacea were used as test strains, and the commercially cultivated strain V844 (T0) was used as a control. The results revealed that the addition of serine had no significant effect on non-degenerated strains T0 and T6, but could effectively restore the production characteristics of degenerated strains T12 and T19. Serine increased the biological efficiency of T12 and even helped the severely degenerated T19 to regrow its fruiting body. Moreover, exogenous serine up-regulated the expression of some antioxidant enzyme genes, improved antioxidase activity, reduced the accumulation of reactive oxygen species (ROS), lowered malondialdehyde (MDA) content, and restored mitochondrial membrane potential (MMP) and mitochondrial morphology. Meanwhile, serine treatment increased lignocellulase and mycelial energy levels. These findings form a theoretical basis and technical support for the rejuvenation of V. volvacea degenerated strains and other edible fungi.

Resolution improvement of photothermal microscopy by the modulated difference method

Photothermal microscopy (PTM) was developed to image non-fluorescent objects. In the past two decades, PTM has reached single-particle and single-molecule sensitivity and has been used in the fields of material science and biology. However, PTM is a far-field imaging method whose resolution is restricted by the diffraction limits. This Letter reports a resolution improvement approach for photothermal microscopy called modulated difference PTM (MD-PTM), which utilizes Gaussian and doughnut formalism heating beams that are modulated at the same frequency but are of opposite phase to generate the photothermal signal. Furthermore, the opposite phase characteristics of the photothermal signals are applied to determine the objective profile from the PTM magnitude, and this helps to improve the lateral resolution of PTM. The lateral resolution is related to the difference coefficient between the Gaussian and doughnut heating beams; an increase in the difference coefficient causes a larger sidelobe of the MD-PTM amplitude, which readily forms an artifact. A pulse-coupled neural network (PCNN) is employed for phase image segmentations of MD-PTM. We experimentally study the micro-imaging of gold nanoclusters and crossed nanotubes using MD-PTM, and the results indicate that MD-PTM has merit in terms of improving the lateral resolution.

Ionospheric Pre‐Sunrise Uplift: Comparison of Sanya Incoherent Scatter Radar Observations and Numerical Simulations

Abstract Using the high spatial‐temporal resolution electron density profiles observed by the newly built Sanya Incoherent Scatter Radar at Sanya (18.3°N, 109.6°E) in its first 1‐month‐long continuous run with Barker code, we identified a substantial ionospheric pre‐sunrise uplift at low latitudes associated with the upward movement of ions. The uplift mainly occurs during local solar time 01:00–06:00 and is ∼52–240 km with significant night‐to‐night variability. With increasing geomagnetic and/or interplanetary activity, the uplift can have a larger amplitude. Further TIEGCM simulations show that the pre‐sunrise ionospheric uplift is jointly determined by the net effect of ambipolar diffusion, neutral wind, and E × B drift. The neutral wind during 00:00–05:00 usually lifts the ionosphere through equatorward wind. After 02:00, the weakening and/or even the reversal of downward E × B drift, which is not sufficient to offset the net uplift effect of wind and diffusion combination, triggers uplifting.
