Effects of Sublethal Concentrations of the Herbicide, Glyphosate, on Embryonic Development of the Indian Major Carp, Labeo rohita

Aquaculture is one of the fastest growing sectors worldwide. Currently, 50% of global fish consumption is provided by aquaculture. In India, the major cultivated fishes are Labeo catla, Labeo rohita, Cirrhinus mrigala, Hypophthalmichthys molitrix, Ctenopharyngodon idella, and Cyprinus carpio. Among these, L. rohita is a widely consumed fish. We aimed to study the effect of one of the most widely used herbicides, glyphosate, on the embryonic development of L. rohita. The 96 h LC50 of glyphosate for L. rohita embryos was found to be 20.89 mgL−1. The embryos were exposed to 1/10th, 1/5th,, and 1/3rd concentrations of LC50 for 96 h. The observed deformities included abdominal curvature, kink formation in the tail, yolk sac edema, pericardial edema, and improperly flattened swim bladder. Besides, we could observe a reduction in pigmentation at 96 h and a decrease in heart rate at 24, 48, 72, and 96 h. All these deformities led to the mortality of embryos. This study indicated that the herbicide (glyphosate) can adversely affect the natural population of the Indian major carp, L. rohita.

Endocrine disrupting chemicals (EDCs) are naturally occurring or synthetic molecules present in an ecosystem that have the potential to alter the production, release, transport, metabolism, binding, action, or elimination of natural hormones in the body.They act by mimicking endogenous hormones, antagonizing normal hormones, altering the natural pattern of hormone synthesis or metabolism, or modifying hormone receptors.The chemicals, particularly disrupting the reproductive system, can be androgenic, anti-androgenic, estrogenic or anti-estrogenic.This review provides a brief overview on spermatogenesis, disruption of spermatogenesis and effects of various EDCs on hormones involved in spermatogenesis.EDCs affect spermatogenesis in two ways, by affecting the development and function of male gonads or by disrupting the synthesis and action of hormones involved in spermatogenesis, such as, GnRH, gonadotropin, dihydroandrogen, 11-ketotestosterone, estrogen, and progesterone.

Aquatic Invasive Species: Traditional Control Options to Emerging Genetic Bio-control Strategies

Worldwide, alien species invasion already constitutes to be a major threat to the biodiversity of various ecosystems, particularly in freshwater ecosystems, and with a warming of the climate, the range of habitats suitable for its establishment may increase further. Though it is possible to manage invasive aquatic species in small invaded areas by physical removal, use of barriers or biocides, and environmental modifications, once established and widely distributed, the eradication of invasive species often becomes impractical. To date, biological control is considered the prime realistic option for controlling well established and widely distributed invasive species. But the application of classical biological control methods using unmodified living organisms (predator, parasite or pathogen) to control target populations of alien fishes is very limited to date, mainly because of the difficulties in finding suitable agents. Recent advances in genetic technologies, like chromosome set manipulations or recombinant DNA techniques, or a combination of both techniques provide opportunities that could be used for the control and eradication of individual invasive species. Approaches like sterile-male, super male, female lethal, bisex-lethal, trojan female, sex-reversed trojan female, neo female, daughterless technology, and CRISPR/CAS9 – gene editing are considered far more species-specific than mechanical or chemical methods, and also eﬃcient and cost-eﬀective. However, just a few trials of some of these methods have been undertaken in the field to date, and hence there is still a long way to go before tuning genetic biocontrol approaches into an on-field reality.

Microplate-based incubation method for quality assessment of eggs of Indian major carp Catla catla

Incubation of fertilised eggs from the Indian major carp species, Catla catla was carriedout in tissue culture microplates to assess the quality in terms of hatching rate and posthatchsurvival rate. Eggs from five females (F1, F2, F3, F5 and F6) were strip spawned andfertilised with milt from a single male. The fertilised eggs were distributed at one egg perwell of 24 well microplates, with three plates for each female. The plates were incubated at30oC in an incubator with continuous shaking and complete hatching was observed by 36 h.The hatching rate varied within a range of 0 to 52.8% showing the maximum of 5% for F5followed by F1 (50%) which were significantly higher than the other three females (F2-18.1,F6-5.6 and F3-0%). The eggs from these two females (F1 and F5) thus could be consideredas good. The survival percentage of hatchlings recorded for a period of 8 days post-hatchingwas found to vary from 58.3 to 83.3 and there was low variability among different females onany observed day. Between these two egg quality determinants, the hatching rate was foundto be more suitable considering the wide variability shown by different females indicatingthe quality difference. The microplate method seems to be an appropriate tool to assess theegg quality in catla and can be extended to other species as well. Keywords:Catla catla, Egg quality, Hatching rate, Microplate

Climate vulnerability and induced changes in physico-chemical properties of aquatic environment can bring impairment in metabolism, physiology and reproduction in teleost. Variation in environmental stimuli mainly acts on reproduction by interfering with steroidogenesis, gametogenesis and embryogenesis. The control on reproductive function in captivity is essential for the sustainability of aquaculture production. There are more than 3,000 teleost species across the globe having commercial importance; however, adequate quality and quantity of seed production have been the biggest bottleneck. Probiotics are widely used in aquaculture as a growth promoter, stress tolerance, pathogen inhibition, nutrient digestibility and metabolism, reproductive performance and gamete quality. As the gut microbiota exerts various effects on the intestinal milieu which influences distant organs and pathways, therefore it is considered to be a full-fledged endocrine organ. Researches on Gut-Brain-Gonad axis (GBG axis) and its importance on physiology and reproduction have already been highlighted for higher mammals; however, the study on fish physiology and reproduction is limited. While looking into the paucity of information, we have attempted to review the present status of microbiome and its interaction between the brain and gut. This review will address a process of the microbiome physiological mechanism involved in fish reproduction. The gut microbiota influences the BPG axis through a wide variety of compounds, including neuropeptides, neurotransmitter homologs and transmitters. Currently, research is being conducted to determine the precise process by which gut microbial composition influences brain function in fish. The gut-brain bidirectional interaction can influence brain biochemistry such as GABA, serotonin and tryptophan metabolites which play significant roles in CNS regulation. This review summarizes the fact, how microbes from gut, skin and other parts of the body influence fish reproduction through the Gut-Brain-Gonad axis.

Biochemical and Histopathological Alteration in Subadult Labeo Rohita (Hamilton) Upon Chronic Exposure to Triflumezopyrium

Research has been done on the long-term effects of triflumezopyrium causing oxidative stress in freshwater culturable carp, Labeo rohita, after exposure to sub-lethal concentrations (2.468 ppm, 1.480 ppm, and 0.740 ppm, respectively) of the pesticide for 21 days . The liver, kidney, gills, muscle, and brain tissue of the fish were examined for histopathological and biochemical parameters such as catalase (CAT), superoxide dismutase (SOD), lactate dehydrogenase (LDH), malate dehydrogenase (MDH), alanine aminotransaminase (ALT), aspartate aminotransaminase (AST), acetylcholinessterase (AChE), and hexokinase. According to studies, increased levels of reactive oxygen species (ROS) cause oxidative damage to lipids and proteins and suppress fish's ability to produce antioxidant defenses. After 21 days of exposure, the activity of these enzymes— CAT, SOD, LDH, MDH, ALT has increased and a drop in the activity of Total Protein was found in all treatment groups as comparison to the control group. Long-term triflumezopyrium exposure increased ROS production, ultimately leading to oxidative cell damage and inhibiting the antioxidant capabilities of the fish tissues. Histopathological analysis also showed alteration in different tissues structures of pesticide treated fishes. Group of fishes exposed to highest sublethal concentration of the pesticide showed higher damage rate.
