Pyrimidoindole derivative UM171 enhances derivation of hematopoietic progenitor cells from human pluripotent stem cells

In the field of hematopoietic regeneration, deriving hematopoietic stem cells (HSCs) from pluripotent stem cells with engraftment potential is the central mission. Unstable hematopoietic differentiation protocol due to variation factors such as serums and feeder cells, remains a major technical issue impeding the screening of key factors for the derivation of HSCs. In combination with hematopoietic cytokines, UM171 has the capacity to facilitate the maintenance and expansion of human primary HSCs in vitro . Here, using a serum-free, feeder-free, and chemically defined induction protocol, we observed that UM171 enhanced hematopoietic derivation through the entire process of hematopoietic induction in vitro . UM171 facilitated generation of robust CD34 + CD45 + derivatives that formed more and larger sized CFU-GM as well as larger sized CFU-Mix. In our protocol, the derived hematopoietic progenitors failed to engraft in NOG mice, indicating the absence of long-term HSC from these progenitors. In combination with other factors and protocols, UM171 might be broadly used for hematopoietic derivation from human pluripotent stem cells in vitro . • UM171 promotes hematopoietic derivation from human pluripotent stem cells. • UM171 enhances the derivation of hematopoietic cells at multiple developmental steps. • Hematopoietic progenitors from the UM171 group form multiple lineages in vitro .

Experimental investigation of the ease allowance of clothing and thermal-moisture comfort in exercise based on fuzzy comprehensive evaluation

The level of thermal-moisture comfort experienced by an individual is strongly influenced by the clothing they wear and also influenced by the ease allowance of that clothing. This study aimed to investigate the impact of ease allowances of clothing on thermal-moisture comfort during exercise or movement. Seventeen female subjects were asked to wear sample clothing with different sizes during a designated 5-phase exercise in a climate chamber. Throughout the exercise phases, the temperature and humidity of the microclimate under the clothing were monitored while the subjects provided subjective evaluations of their comfort levels. To account for variation in ease allowances, the 32 experimental repetitions were grouped based on the ease allowance of the clothing worn. The FCE-CRITIC method was then used to determine the variable weights of factors and to compare the thermal-moisture status to the subjective evaluation. Results revealed that humidity was more strongly weighted during RS and RC (i.e., running and recovery phase after exercise) compared to other phases, means that moisture comfort was the main factor driving the clothing comfort sensation within the two phases. The ease allowance of clothing was found to have an impact on the sensations of thermal and moisture comfort during exercise, although this effect was not significant. Furthermore, the study identified the optimum range of ease allowance for thermal-moisture comfort during exercise to be between 6 and 10 cm.

A comprehensive evaluation of a polymeric zwitterionic hydrophilic monolith for nucleotide separation

Rapid and effective separation of nucleotides (NTs) and their derivatives is crucial for studying their physiological functions. In this work, we comprehensively evaluated the separation ability of a zwitterionic hydrophilic monolith, i.e., poly(N,N-dimethyl-N-(3-methacrylamidopropyl)-N-(3-sulfopropyl)ammonium betaine-co–N,N’-methylenebisacrylamide) (poly(SPP-co-MBA)) for NTs analysis, including its selectivity, chemical stability under extremely basic condition and compatibility with hydrophilic interaction liquid chromatography (HILIC) coupled with mass spectrometry (HILIC-MS). The poly(SPP-co-MBA) monolith exhibited excellent chemical stability, as evidenced by the low relative standard deviation of retention time (0.16–1.05%) after 4000 consecutive injections over one month under strong alkaline elution condition (pH 10). After optimizing the separation conditions, including buffer pH and concentration, organic solvent content and column temperature, four nucleoside triphosphates, five nucleoside diphosphates and five nucleoside monophosphates were baseline separated within 7 min. Additionally, the mixtures containing one nucleoside and its corresponding mono-, di-, and triphosphates were baseline separated within only 3 min, respectively. It is good HILIC-MS compatibility was also confirmed by the satisfactory peak shape and high response of nine NTs. Overall, the proposed poly(SPP-co-MBA) monolith exhibited good mechanical stability and compatibility of HILIC-MS, making it a promising technique for NTs analysis.

Transcription factor Hoxb5 reveals the unidirectional hierarchy of hematopoietic stem cell pool

Hoxb5 exhibits preferential expression in hematopoietic stem cells (HSCs) and uniquely marks the long-term HSCs (LT-HSCs). Previous studies have demonstrated the remarkable capability of Hoxb5 to alter cell fates when enforced expression in blood progenitors, such as B cell progenitors and multipotent progenitors. Additionally, Hoxb5 deficiency does not hinder the generation of LT-HSCs. However, the specific impact of Hoxb5 deletion on LT-HSCs has remained unexplored. To address this, we developed a conditional Hoxb5 knockout-reporter mouse model, wherein Hoxb5 was knock out by the Vav-cre recombinase, and the endogenous Hoxb5 promoter drove the expression of the blue fluorescent protein (BFP). Our findings revealed that the primary recipients, who transplanted with HSCs indicating Hoxb5 deficiency by the presence of BFP (BFP-positive HSCs), exhibited comparable levels of donor chimerism and lineage chimerism to recipients transplanted with HSCs that spontaneously did not express Hoxb5 and thus lacked BFP expression (BFP-negative HSCs). However, during the secondary transplantation, recipients receiving total bone marrow (BM) from the primary recipients with BFP-positive HSCs showed significantly higher levels of donor chimerism and more robust multi-lineage chimerism compared to those receiving total BM from the primary recipients with BFP-negative HSCs. Our results indicate that deleting Hoxb5 in LT-HSCs transiently influences their lineage differentiation bias without compromising their long-term self-renewal capacity. These findings highlight the primary role of Hoxb5 in regulating lineage commitment decisions in LT-HSCs, while emphasizing that its presence is not indispensable for the maintenance of long-term self-renewal capacity.

BCL9 provides multi-cellular communication properties in colorectal cancer by interacting with paraspeckle proteins

Abstract Colorectal cancer (CRC) is the third most commonly diagnosed cancer, which despite recent advances in treatment, remains incurable due to molecular heterogeneity of tumor cells. The B-cell lymphoma 9 (BCL9) oncogene functions as a transcriptional co-activator of the Wnt/β-catenin pathway, which plays critical roles in CRC pathogenesis. Here we have identified a β-catenin-independent function of BCL9 in a poor-prognosis subtype of CRC tumors characterized by expression of stromal and neural associated genes. In response to spontaneous calcium transients or cellular stress, BCL9 is recruited adjacent to the interchromosomal regions, where it stabilizes the mRNA of calcium signaling and neural associated genes by interacting with paraspeckle proteins. BCL9 subsequently promotes tumor progression and remodeling of the tumor microenvironment (TME) by sustaining the calcium transients and neurotransmitter-dependent communication among CRC cells. These data provide additional insights into the role of BCL9 in tumor pathogenesis and point towards additional avenues for therapeutic intervention.

Effect of submerged entry nozzle wall surface morphologies on boundary layer structure and alumina inclusions transport

During the Al-killed steel continuous casting process, the molten steel corrosion and the accumulation of alumina inclusion deposits affect the submerged entry nozzle (SEN) wall surface, including the surface morphologies of the smooth wall, porous refractory wall, and clogged wall. The SEN wall surface morphology affects the boundary layer structure and alumina inclusions transport. In this study, a physical modeling method was adopted, and the surface morphologies simulation was realized by filling up the natural porous refractory material and inserting the real clog material in the polymethyl methacrylate SEN model. The velocity in the boundary layer was measured using the particle image velocimetry (PIV) technology, and the alumina inclusions transport path in the boundary layer was calculated by MATLAB software. The MATLAB codes combined the velocity data from the PIV measurement results and the inclusion transport equation. The four-quadrant analysis showed that sweep and ejection events existed in the boundary layer. The fluctuations of the velocity and the turbulent kinetic energy in the normal direction were increased in the porous refractory and the clogged wall boundary layer when the sweep and ejection events existed. The transport of the alumina inclusions with a diameter of 1–15 μm was affected by the ejection and the sweep events. The alumina inclusions moved toward the boundary in the sweep event. During the sweep event, the transport path of alumina inclusions with 1 μm diameter was close to the boundary; the alumina inclusions were more easily attached to the boundary. The alumina inclusions escaped from the boundary in the ejection event. In the porous refractory and the clogged walls, the alumina inclusion transport path in the normal direction was increased. When the SEN wall’s morphologies changed from smooth wall to porous refractory wall and clogged wall, the sweep event area proportion increased from 10.17% to 39.77%, and the ejection event area proportion decreased from 32.96% to 9.24%. Moreover, the sweep event’s probability increased from 25.83% to 28.24% when the morphologies of the SEN wall changed from smooth wall to porous refractory wall and clogged wall, which will increase the alumina inclusion deposition rate in the porous refractory wall and the clogged wall boundary.

Blueberry Bruise Detection using Hyperspectral Transmittance Imaging

Abstract. Blueberries are a high-value crop for the US agricultural industry. Bruising of the berries during harvesting and processing is a significant source of monetary losses for the industry. The goal of this study was to investigate the potential of hyperspectral imaging using the transmittance mode to detect and grade blueberry bruises. A total of 90 fruits were evenly divided into three groups: control, stem bruise, and equatorial bruise. Fruits in the bruised treatment groups were dropped from 30 cm onto a steel plate using a pendulum to control the location of impact, while the control group was kept intact. A near-infrared hyperspectral imaging system was implemented to acquire transmittance images at a range of 950 nm to 1400 nm. Images were acquired from three positions for fruit in the control and stem bruise groups, and from four positions in the equatorial bruise group. Color images of sliced fruit were obtained as references. By comparing against the reference color images, the profiles of spatial and spectral intensity were evaluated to determine which wavelengths and imaging positions can most effectively detect bruising. Images at 1070 nm were processed using thresholding and morphological reconstruction to calculate bruise ratios by dividing the number of bruised pixels by the total number of fruit pixels. Correlation analysis was performed between the calculated bruise ratios and human grading for three treatments. The results indicated that it is feasible to use hyperspectral transmittance images to detect and quantify blueberry bruises.

Yield estimation of high-density cotton fields using low-altitude UAV imaging and deep learning

Abstract Background China has a unique cotton planting pattern. Cotton is densely planted in alternating wide and narrow rows to increase yield in Xinjiang, China, causing the difficulty in the accurate estimation of cotton yield using remote sensing in such field with branches occluded and overlapped. Results In this study, unmanned aerial vehicle (UAV) imaging and deep convolutional neural networks (DCNN) were used to estimate densely planted cotton yield. Images of cotton fields were acquired by the UAV at an altitude of 5 m. Cotton bolls were manually harvested and weighed afterwards. Then, a modified DCNN model (CD-SegNet) was constructed for pixel-level segmentation of cotton boll images by reorganizing the encoder-decoder and adding dilated convolutions. Besides, linear regression analysis was employed to build up the relationship between cotton boll pixels ratio and cotton yield. Finally, the estimated yield for four cotton fields were verified by weighing harvested cotton. The results showed that CD-SegNet outperformed the other tested models, including SegNet, support vector machine (SVM), and random forest (RF). The average error in yield estimates of the cotton fields was as low as 6.2%. Conclusions Overall, the estimation of densely planted cotton yields based on low-altitude UAV imaging is feasible. This study provides a methodological reference for cotton yield estimation in China.

Enhancing Electrochemical Performance of Reversible Solid Oxide Cells with Double Element-Deficient Pr_0.97Ba_0.97Co_1.5Fe_0.5O_5+δ Oxygen Electrodes

Reversible solid oxide cells (RSOCs) are state-of-the-art all-solid-state energy conversion devices that operate as either solid oxide fuel cells (SOFCs) or solid oxide electrolysis cells (SOECs), with the oxygen electrode playing a critical role in both the oxygen reduction (ORR) and oxygen evolution (OER) reactions. Herein, PrBaCo1.5Fe0.5O5+δ (PBCF) is modified via double deficiency modulation of the A-site to improve its performance as an RSOC oxygen electrode. The cation-deficient variant, Pr0.97Ba0.97Co1.5Fe0.5O5+δ (PBCF97), exhibits a balanced oxygen vacancy concentration and ionic conductivity. RSOCs using PBCF97-Gd0.1Ce0.9O2(GDC) as the oxygen electrode achieve a peak power density of 1.685 W/cm2 in SOFC mode and a current density of 2.61 A/cm2 at an electrolytic voltage of 1.5 V in SOEC mode at 800 °C. In addition, the cell demonstrates stable reversible operation for 160 h, highlighting its potential for advanced RSOC applications.
