Previous investigations have demonstrated that capsaicin-sensitive primary sensory nerves play an important role in modulation of the peripheral resistance of the circulation system. The vanilloid receptor subtype 1 (VR1) is expressed almost exclusively in the primary sensory nerves and cell bodies of these sensory neurons. Rutaecarpine (Rut) can relax vascular smooth muscle via stimulation of calcitonin gene-related peptide (CGRP) release by activation of VR1.In the present study, we examined the depressor effect of Rut and the possible mechanisms in the phenol-induced hypertensive rats, in which hypertension was induced by injecting 50 microl of 10% phenol in the lower pole of the left kidney.Acute administration of Rut (30, 100 or 300 microg/kg, i.v.) caused a depressor effect concomitantly with an increase in the plasma concentration of CGRP in a dose-dependent manner, which was blocked by capsaicin (used to deplete the CGRP from sensory nerves) or capsazepine (a competitive VR1 antagonist), causing an approximately 85% and approximately 80% change in mean arterial pressure, respectively, and by either of them, causing an approximately 90% elevation of plasma CGRP. In the chronic study, Rut at a dose of 3 or 6 mg/kg per day significantly lowered tail-cuff systolic blood pressure to 159 +/- 8 and 136 +/- 10 mmHg, respectively, compared with hypertensive rats (179 +/- 8 mmHg), and caused a sustained hypotensive effect from day 6 on. Pretreatment with capsaicin blocked the depressor effect of Rut by approximately 65%. Treatment with Rut significantly increased the synthesis and release of CGRP, as shown by the increase in the levels of CGRP mRNA and peptide in the dorsal root ganglia, the density of CGRP immunoreactive nerve fibers in the mesenteric artery, the CGRP content in the spinal cord and the plasma concentration of CGRP, which was markedly attenuated by pretreatment with capsaicin.These results suggest, for the first time, that the hypotensive effect of Rut is mediated by stimulation of CGRP synthesis and release via activation of VR1 in the phenol-induced hypertensive rat.

Effects of Selaginellin on Homocysteine-Induced Senescence in Human Umbilical Vein Endothelial Cells

Homocysteine plays a key role in endothelial cell senescence associated with atherosclerosis-based cardiovascular diseases. Selaginellin, a component extracted from Selaginella pulvinata (Hook. et Grev.) Maximo, was assessed for its ability to protect human umbilical vein endothelial cells against homocysteine-induced senescence. The endothelial cells were pretreated with various concentrations (10(-7), 3 x 10(-7), or 10(-6) M) of selaginellin for 1 hour before exposure to homocysteine. Selaginellin was shown to protect endothelial cells against homocysteine-induced senescence, as determined by senescence-associated beta-galactosidase activity, telomerase activity, and cell cycle distribution. In addition, the increase in levels of intracellular reactive oxygen species and downregulation of SIRT1 gene expression induced by homocysteine were significantly reversed by selaginellin. Our study suggests that selaginellin has a protective effect against homocysteine-induced senescence through mechanisms related to antioxidation via scavenging reactive oxygen species and upregulating the expression of SIRT1 gene.

Effect of resveratrol derivative BTM-0512 on high glucose-induced dysfunction of endothelial cells: role of SIRT1

Hyperglycemia impairs the function of endothelial cells. Sirtuin 1 (SIRT1) is involved in regulating the function of endothelial cells. Resveratrol, a polyphenol found in many plant species, exerts protective effects on endothelial cells through activation of SIRT1. The aims of this work were to explore whether BTM-0512, a novel derivative of resveratrol, is able to exert beneficial effects on high glucose-induced dysfunction of endothelial cells through regulation of SIRT1. We found that high glucose significantly impaired the function of endothelial cells as shown by reduced tube formation, cell migration, and cell adhesion concomitantly with downregulation of mRNA expression of SIRT1 and vascular endothelial growth factor as well as increased tumor necrosis factor-α release and reactive oxygen species production. These effects of high glucose were inhibited by pretreatment with BTM-0512. The beneficial effects of BTM-0512 on high glucose-induced cell dysfunction were abolished by splitomicin, a specific inhibitor of SIRT1. The regulatory effects of BTM-0512 on high glucose-induced changes in vascular endothelial growth factor mRNA expression and tumor necrosis factor-α release were also abolished by splitomicin. The results suggest that BTM-0512 exerts beneficial effects on high glucose-induced endothelial cell dysfunction through regulation of the SIRT1 - reactive oxygen species - vascular endothelial growth factor - tumor necrosis factor-α pathway.

Chronic administration of isocarbophos induces vascular cognitive impairment in rats

Abstract Vascular dementia, being the most severe form of vascular cognitive impairment ( VCI ), is caused by cerebrovascular disease. Whether organophosphorus causes VCI remains unknown. Isocarbophos (0.5 mg/kg per 2 days) was intragastrically administrated to rats for 16 weeks. The structure and function of cerebral arteries were assayed. The learning and memory were evaluated by serial tests of step‐down, step‐through and morris water maze. Long‐term administration of isocarbophos reduced the hippocampal acetylcholinesterase ( AC hE) activity and acetylcholine ( AC h) content but did not alter the plasma AC hE activity, and significantly damaged the functions of learning and memory. Moreover, isocarbophos remarkably induced endothelial dysfunction in the middle cerebral artery and the expressions of ICAM ‐1 and VCAM ‐1 in the posterior cerebral artery. Morphological analysis by light microscopy and electron microscopy indicated disruptions of the hippocampus and vascular wall in the cerebral arteries from isocarbophos‐treated rats. Treatment of isocarbophos injured primary neuronal and astroglial cells isolated from rats. Correlation analysis demonstrated that there was a high correlation between vascular function of cerebral artery and hippocampal AC hE activity or AC h content in rats. In conclusion, chronic administration of isocarbophos induces impairments of memory and learning, which is possibly related to cerebral vascular dysfunction.

[Evaluation of asthma management from the surveys in 30 provinces of China in 2015-2016].

Objective: To evaluate the general level of asthma management in urban areas of China and further promote the national asthma management plan. Methods: A multi-center, cross-sectional survey was carried out in 30 provinces of China (except for Tibet) during Oct 2015 to May 2016. It's a questionnaire-based face-to-face survey which included asthma management using peak flow meter (PFM) and pulmonary function test, medication choice of maintenance therapy and asthma education. Results: A total of 3 875 asthmatic outpatients were recruited including 2 347(60.6%) females and 1 528(39.4%) males. The mean age was (50.7±16.7) years ranging from 14 to 99. Only 10.1%(388/3 837) patients used PFM as monitoring, whereas 62.1%(2 405/3 874) patients underwent pulmonary function test during the past year. There were 57.4%(2 226/3 875) patients treated with inhaled cortical steroid plus long-acting β(2)-agonist combinations (ICS+LABA) as daily medication. 43.3%(1 661/3 836) patients were followed up by physicians. Among this population, 1 362 asthmatic outpatients were recruited, who also took part in the asthma control survey in 2007-2008 in 10 cities. In this subgroup, 17.9%(244/1 360) were tested by PFM and 66.6%(907/1 362) by pulmonary function test during last year. As to the medication, 63.1%(860/1 362) selected ICS+LABA for daily control. There were 50.4%(685/1 359) patients in the follow-up cohort by physicians. Compared to the similar survey conducted in 2007-2008, the proportion of patients with ICS+LABA regimen and follow-up by physicians were markedly higher, while the rate of PFM use did not have significant improvement. Conclusion: Although the present level of asthma management in China is still far from ideal, asthma management has improved compared to 8 years ago. Yet the use of PFM does not significantly improve. Asthma action plan and application of PFM should be further promoted to improve the level of asthma management.目的：评估我国城区支气管哮喘（以下简称哮喘）患者哮喘管理水平，为今后开展全国哮喘管理计划提供参考依据。方法：多中心横断面研究。2015年10月—2016年5月在全国哮喘研究协作组成员所在30个城市的30家三级甲等医院呼吸内科门诊进行面对面问卷调查，调查内容包括哮喘管理（峰流速仪使用、肺功能检测）、维持治疗药物使用、哮喘教育（哮喘行动计划、医院哮喘教育），分析我国现阶段城区哮喘患者哮喘管理水平。结果：共纳入3 875例门诊哮喘患者，女2 347例（60.6%），男1 528例（39.4%），年龄14~99（50.7±16.7）岁。10.1%（388/3 837）的患者使用过峰流速仪，既往一年内62.1%（2 405/3 874）的患者做过肺功能检查，57.4%（2 226/3 875）的患者选择含吸入糖皮质激素（ICS）联合长效β(2)受体激动剂（LABA）作为每日规律使用的控制性药物，43.3%（1 661/3 836）的患者有专科医生制定哮喘治疗（随访）行动计划。其中，曾在2007—2008年哮喘控制调查的10个城市在本次调查中共纳入1 362例，17.9%（244/1 360）的患者使用过峰流速仪，既往一年内66.6%（907/1 362）的患者做过肺功能检查，63.1%（860/1 362）的患者选择ICS+LABA作为每日规律使用的控制性药物，50.4%（685/1 359）的患者有专科医生制定的哮喘治疗（随访）行动计划。与2007—2008年的调查结果相比，选用ICS+LABA作为每日规律使用的控制性药物及有专科医师制定哮喘治疗（随访）计划的比例明显升高，峰流速仪的使用率未见显著改善。结论：目前我国哮喘总体管理水平尚不理想，曾在2007—2008年哮喘调查的10城市中可以看到，哮喘管理水平在提高，而峰流速仪使用率较8年前未有显著改善。应大力推广哮喘行动计划及峰流速仪的使用，以提高哮喘管理水平。.

TRPC3 channel confers cerebrovascular remodelling during hypertension via transactivation of EGF receptor signalling

Ionic perturbation in vascular smooth muscle cells contributes to cerebrovascular remodelling in the setting of hypertension, but the role of transient receptor potential (TRP) channel superfamily remains unknown. The present study was conducted to define the contribution of TRP channels to cerebrovascular remodelling.By integrating quantitative PCR, western blotting, patch clamping, and Ca(2+) imaging, we identified TRP channel, subfamily canonical, member 3 (TRPC3) as the channel subtype most considerably elevated in basilar arteries of two-kidney, two-clip stroke-prone hypertensive rats. Importantly, administration of pyrazole 3 (Pyr3), a TRPC3 channel blocker, attenuated cerebrovascular remodelling. During hypertension, epidermal growth factor receptor (EGFR) was transactivated, as evidenced by marked EGFR phosphorylation, increased pro-HB-EGF shedding, and elevated activity of ADAM17 (HB-EGF sheddase). ADAM17 activity was increased owing to enhanced activation rather than elevated expression. Remarkably, Pyr3 treatment suppressed EGFR transactivation in hypertension. In proliferating basilar artery smooth muscle cells or basilar arteries of hypertensive rats, co-immunoprecipitation assay revealed an interaction between TRPC3 and ADAM17 upon Ang II stimulation.Collectively, we demonstrated that enhanced EGFR transactivation, due to increased TRPC3 expression and functional coupling of TRPC3/ADAM17, resulted in cerebrovascular remodelling. Therefore, TRPC3-induced EGFR transactivation may be therapeutically exploited to prevent hypertension-induced cerebrovascular remodelling.

[A retrospective study of the treatment of patients hospitalized for asthma exacerbation in China].

Objective: To study the treatment of patients hospitalized for asthma exacerbation in China. Methods: This was a retrospective study and involved patients hospitalized for asthma exacerbation in 29 hospitals throughout China during Jan 2013 to Dec 2014. Information of the demographic features, the severity of the exacerbation, the medicine prescribed during the hospitalization and the use of mechanical ventilation were collected and analyzed. Results: During the study period, there were 3 240 patients hospitalized for asthma exacerbation, and 1 369(42.3%) of them were males 1 871(57.7%)were females. Patients of mild, moderate, severe, and life-threatening exacerbation counted for 41.7%, 37.8%, 19.2% and 1.0%, respectively of the total patients. Of all the patients, 72.6% used bronchodilators by nebulizer, 70.8% used inhaled corticosteroids by nebulizer, 60.5% used intravenous corticosteroids, 17.3% used oral corticosteroids, and 80.5% used antibiotics. The percentages of patients using systemic corticosteroids and antibiotics were higher in patients with more severe exacerbation. In patients with mild exacerbation, there were 74.9% and 52.2% who used antibiotics and systemic corticosteroids, respectively. A total of 73 patients (2.3%) used mechanical ventilation, and 62 of them used noninvasive ventilation, 16 used invasive ventilation, and 5 used both. Conclusions: Nebulizer therapy has been accepted as the main administration route of medicine in the treatment of asthma exacerbation. A large amount of patients used antibiotics and systemic corticosteroids during hospitalization, indicating there may be some overuse of these medicines.目的：调查分析我国城区支气管哮喘(简称哮喘)急性发作住院患者的治疗情况。方法：纳入我国29个省、自治区、直辖市的29个城市的29家三级甲等医院2013年1月至2014年12月因哮喘急性发作住院的患者的临床资料，对其人口学资料、急性发作严重程度、住院期间药物治疗及机械通气情况进行回顾性调查分析。结果：共纳入3 240例患者，其中男1 369例(42.3%)，女1 871例(57.7%)；轻、中、重度及危重度急性发作患者分别占41.7%、37.8%、19.2%及1.0%；72.6%的患者住院期间雾化吸入了支气管舒张剂，70.8%的患者雾化吸入了糖皮质激素(简称激素)；60.5%的患者静脉应用了激素，17.3%的患者接受了口服激素治疗；80.5%的患者住院期间应用了抗菌药物治疗。随着急性发作严重程度的增加，患者抗菌药物及全身激素的使用比例逐渐升高，即使在轻度患者中，也分别有74.9%及52.2%的患者应用了抗菌药物及全身激素。全部患者中73例(2.3%)应用了机械通气(其中应用无创通气的共62例，有创通气16例，无创＋有创机械通气5例)。结论：我国城区哮喘急性发作住院患者雾化吸入治疗已作为主要给药途径，但仍需更进一步推广；目前尚存在抗菌药物和激素不合理使用的情况。.

All‐trans retinoic acid inhibits cobalt chloride‐induced apoptosis in PC12 cells: Role of the dimethylarginine dimethylaminohydrolase/asymmetric dimethylarginine pathway

Abstract Previous studies have shown that the endogenous nitric oxide synthase inhibitor asymmetric dimethylarginine (ADMA) and its specific hydrolase dimethylarginine dimethylaminohydrolase (DDAH) are involved in the regulation of apoptosis in different cell types. In the present study, we investigated the role of the DDAH/ADMA pathway in cobalt chloride (CoCl 2 )–induced apoptosis and the antiapoptotic effect of all‐trans retinoic acid (atRA) in undifferentiated pheochromocytoma (PC12) cells. Treatment of CoCl 2 (125 μM) for 48 hr significantly induced the apoptosis of PC12 cells, concomitantly with increased intracellular reactive oxygen species (ROS) production and caspase‐3 activity. CoCl 2 treatment also decreased the activity of DDAH and the expression of DDAH2 (mRNA and protein), resulting in an increased level of ADMA. All these alterations induced by CoCl 2 were attenuated by atRA (0.1, 1, or 10 μM). Interestingly, the antiapoptotic effects of atRA were inhibited by DDAH2 small RNA interference. In contrast, DDAH2 overexpression inhibited the proapoptotic effects of CoCl 2 . We also found that treatment of exogenous ADMA (3, 10, or 30 μM) induced the apoptosis of PC12 cells in a concentration‐ and time‐dependent manner, which was inhibited by the antioxidant or the caspase‐3 inhibitor. These findings suggest that the modulation of the DDAH/ADMA/ROS pathway plays an important role in CoCl 2 ‐induced apoptosis and the antiapoptotic effects of atRA in undifferentiated PC12 cells. © 2009 Wiley‐Liss, Inc.
