Data from The ABL Switch Control Inhibitor DCC-2036 Is Active against the Chronic Myeloid Leukemia Mutant BCR-ABL<sup>T315I</sup> and Exhibits a Narrow Resistance Profile

<div>Abstract<p>Acquired point mutations within the BCR-ABL kinase domain represent a common mechanism of resistance to ABL inhibitor therapy in patients with chronic myeloid leukemia (CML). The BCR-ABL<sup>T315I</sup> mutant is highly resistant to imatinib, nilotinib, and dasatinib, and is frequently detected in relapsed patients. This critical gap in resistance coverage drove development of DCC-2036, an ABL inhibitor that binds the switch control pocket involved in conformational regulation of the kinase domain. We evaluated the efficacy of DCC-2036 against BCR-ABL<sup>T315I</sup> and other mutants in cellular and biochemical assays and conducted cell-based mutagenesis screens. DCC-2036 inhibited autophosphorylation of ABL and ABL<sup>T315I</sup> enzymes, and this activity was consistent with selective efficacy against Ba/F3 cells expressing BCR-ABL (IC<sub>50</sub>: 19 nmol/L), BCR-ABL<sup>T315I</sup> (IC<sub>50</sub>: 63 nmol/L), and most kinase domain mutants. <i>Ex vivo</i> exposure of CML cells from patients harboring BCR-ABL or BCR-ABL<sup>T315I</sup> to DCC-2036 revealed marked inhibition of colony formation and reduced phosphorylation of the direct BCR-ABL target CrkL. Cell-based mutagenesis screens identified a resistance profile for DCC-2036 centered around select P-loop mutations (G250E, Q252H, Y253H, E255K/V), although a concentration of 750 nmol/L DCC-2036 suppressed the emergence of all resistant clones. A decreased concentration of DCC-2036 (160 nmol/L) in dual combination with either nilotinib or dasatinib achieved the same zero outgrowth result. Further screens for resistance due to BCR-ABL compound mutations (two mutations in the same clone) identified BCR-ABL<sup>E255V / T315I</sup> as the most resistant mutant. In the article, the authors believe that the establishment of a healthy lifestyle for the younger generation is today considered in the practice of schools and universities as one of the priority directions of the humanization of education. A special status is given to the concept of “valeological culture”, the significance of the formation of which, as exemplified by younger schoolchildren, is due to the need to preserve and improve their physical, mental and social well-being. The authors state that health and physical capacity are a condition and basis for the disclosure of a person’s potential. The real lifestyle of a person subsequently depends on how successfully it is possible to form and consolidate the foundations of valeological culture and the skills of a healthy lifestyle at a young age. In this regard, it is believed that in universities it is necessary to pay special attention to the formation of the readiness of future teachers to organize work to develop the valeological culture of schoolchildren. The authors offer a number of important ideas in this direction, give a description of the method of work. Objective To investigate the value of diagnosis and prognostic evaluation according to detect the expression of bcr/abl fusion gene in chronic myeloid leukemia by real-time quatitative PCR.Methods To use the real-time PCR to detect the expression of bcr/abl fusion gene of 39 cases chronic myeloid leukemia and 20 cases others hematological diseases.Results Marrow and peripheral blasts showed significant corrilation with the copy of bcr/abl fusion gene.Bcr/abl fusion gene expression level of newly diagnosis chronic myeloid leukemia increased significantly.After the treatment of hydroxycarbamide±interferon,the expression level of the gene decreased markedly.The patients of recurrence or acceleration phase can be found ahead of 2-4 months By the therapy of Glevec or explantation,the most of the fusion gene turned negative.Conclusion The detection of bcr/abl fusion gene is an important way to diagnose chronic myeloid leukemia,and can be a decisive index of treatment and prognostic evaluation.
