Anti-Atopic Effect of Scutellaria baicalensis and Raphanus sativus on Atopic Dermatitis-like Lesions in Mice by Experimental Verification and Compound-Target Prediction

Abstract:
Scutellaria baicalensis Georgi and Raphanus Sativus Linne herbal mixture (SRE) is a Chinese herbal medicine. In this study, we aimed to evaluate the therapeutic efficacy of SRE as an active ingredient for 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) and to predict the underlying therapeutic mechanisms and involved pathways using network pharmacological analysis. Treatment with SRE accelerated the development of AD-like lesions, improving thickness and edema of the epidermis. Moreover, administering the SRE to AD-like mice suppressed immunoglobulin E and interleukin-4 cytokine and reduced T lymphocyte differentiation. In silico, network analysis was used to predict the exact genes, proteins, and pathways responsible for the therapeutic effect of the SRE against DNCB-induced AD. These results indicated that the SRE exerted protective effects on the DNCB-induced AD-like model by attenuating histopathological changes and suppressing the levels of inflammatory mediators. Therefore, the SRE can potentially be a new remedy for improving AD and other inflammatory diseases and predicting the intracellular signaling pathways and target genes involved. This therapeutic effect of the SRE on AD can be used to treat DNCB-induced AD and its associated symptoms.Keywords:

Taking seeds of Raphanus sativus L.as materials,the effects of some factors including light intensity,pH value,temperature,H2O2 and metallic ions on the stability of chlorophyll in Raphanus sativus L.were studied,in order to provide useful information for developing chlorophyll in Raphanus sativus L.The results showed that chlorophyll in Raphanus sativus L.was of a poor stability of light intensity treatment,while its stability of temperature and H2O2 treatments was satisfactory.pH value had small effects on preservation of chlorophyll in Raphanus sativus L.,and treatments of Zn2+,Cu2+ could increase the stability of chlorophyll.

We reviewed the domestic studies on Scutellaria Baicalensis Georgi.Scutellaria Baiealensis Georgi has a variety of pharmacological effects,and the main effects are anti-microbial,anti-inflammatory and anti-tumor functions.Scutellaria Baicalensis Georgi has good developing prospects.Up to now,Chinese researches on Scuteilaria baicalensis Georgi have gained gratifying results,but some aspects are still existing defects such as the studies on active ingredients and formula of Scutellaria baicalensis Georgi have not been deeply studied.We propose that human genomics and proteomics researching results should be adopted in the study of Scutellaria Baicalensis Georgi,together with the advanced technologies of bio-chip and biological chromatography being used to elucidate mechanisms at molecular level.

OBJECTIVE:To determine the contents of Total flavonoids and metal elements in Scutellaria baicalensis Georgi and to study the function of the microelements in treating cerebrovascular diseases.METHODS:The contents of macroelements(Na,Ca,Mg,and K)and microelements(Fe,Mn,Cu,and Zn)were determined by atomic absorpt ion spectropho- tometry(AAS);the Total flavonoids in Scutellaria baicalensis Georgi were extracted by alcohol extraction method,and the content of Scutellaria baicalensis Georgi was determined by UV spectrophotometry.RESULTS:The results showed that metal elements and Flavonoids were rich in Scutellaria baicalensis Georgi.CONCLUSION:The determination results provide scientific basis for the reasonable use of Scutellaria baicalensis Georgi to eliminate free radicals,prevent and treat cerebrovas- cular diseases as well as data for the wide exploitation and use of Scutellaria baicalensis Georgi.
