    Abstract:
    It remains unclear whether sex differences exist during the development of visceral adipose tissue (VAT) inflammation associated with obesity. The purpose of this study was to clarify sex differences in the occurrence of senescence-related T cells (CD44high PD-1+ CD4+), which play a key role in the progression of VAT inflammation associated with high-fat diet (HFD)-induced obesity. Phase 1: C57BL/6N mice were fed either a control diet (HFC) or HFD for 5 wk. The area under the curve of the oral glucose-tolerance test (oGTT) was maximal at 15 wk in HFD-fed males and at 21 wk in females. At 17 wk, VAT weights were similar, but an increase in the number of macrophages in the VAT was observed only in HFD-fed males. In addition, the numbers of regulatory and senescence-related T cells were consistently higher in males than in females. Phases 2 and 3: 6-wk-old female mice were randomly divided into sham operation and bilateral ovariectomy (OVX) groups and fed either an HFC or HFD from 7 wk. OVX mice were subjected to 17β-estradiol releasing or placebo pellet implantation and fed an HFC. Body and VAT weights were higher in the OVX group than in the sham. The number of macrophages did not change in the OVX group with either diet. HFC-fed OVX mice exhibited high senescence-related T cells in the VAT, resembling HFC-fed male mice. This change was abolished by 17β-estradiol replacement. Thus, we demonstrated different accumulation patterns of VAT immune cells between the sexes, revealing a role for estrogen in the appearance of senescence-related T cells.NEW & NOTEWORTHY The accumulation pattern of adipose tissue differs between the sexes; however, it is unclear whether sex differences exist during the development of adipose tissue inflammation and whether estrogen plays a role. We demonstrated sex differences in immune cells' subpopulation of visceral adipose tissue. The proinflammatory environment appeared earlier in males than in females. In addition, our results suggest that estrogen plays a role in visceral adipose tissue inflammation, particularly by regulating the appearance of senescence-related T cells.

    The cellular growth of adipose tissue from Dorset Horn x Merino wethers was characterized by both hypertrophy and hyperplasia of adipose cells until the sheep were c. 11 months old, after which hypertrophy of existing adipose cells was solely responsible for increases in adipose tissue mass. Omental adipose tissue contained larger adipose cells than either the perirenal or subcutaneous sites. The weight of fat which had been deposited in the boneless carcass meat and the internal adipose tissue depots were linearly and positively correlated with the volume of the adipose cells. Decreases in the mass of adipose tissue, which accompanied nutritional restriction, were due to decreased adipose cell size, since no change was observed in the number of adipose cells per carcass after loss in weight. The cellularity characteristics of rehabilitated sheep were similar to those of sheep which had undergone continuous growth.

    Adipose tissue can be found throughout the body but is primarily distributed in several subcutaneous and visceral locations or “depots” (1). The general distribution pattern and location of adipose tissue depots and the proportion of total body fat that each depot represents varies widely between species. In humans and swine the subcutaneous depot represents a much larger proportion of total adipose tissue than in rodents. In fact, several distinct layers of subcutaneous adipose tissue are present in humans and swine. Excessive adipose tissue accumulation in humans is often associated with the adverse health consequences of the metabolic syndrome (2). An adipose tissue distribution pattern favoring visceral adipose tissue often is implicated in this syndrome.

    To identify senescence‐associated genes (SAGs) in rice leaves, senescence was induced by transferring rice seedlings into darkness. Senescence up‐regulated cDNAs were obtained by PCR‐based subtractive hybridization. Among 14 SAG clones characterized, 11 were found to be associated with both dark‐induced and natural leaf senescence. Three clones were associated only with dark‐induced leaf senescence. The possible physiological roles of these SAGs during rice leaf senescence are discussed.

    GH has profound effects on the amount and distribution of adipose tissue. GHD in both children and adults is accompanied by an increased amount of adipose tissue and by the abdominal predominance of adipose tissue. In contrast, treatment with GH reduces adipose tissue, and redistributes adipose tissue from abdominal to peripheral depots.

    SUMMARY Leaf senescence is a hiphly‐controlled sequence of events comprising the final stage of development. Cells remain viable during the process and new gene expression is required. There is some similarity between senescence in plants and programmed cell death in animals. In this review, different classes of senescence‐related genes are defined and progress towards isolating such genes is reported. A range of internal and external factors which appear to cause leaf senescence is considered and various models for the mechanism of senescence‐ initiation are described. The current understanding of senescence at the wrganelle and molecular levels is presented. Finally, same ideas are mooted as to why senescence occurs and why it should be studied further.
