Abstract A simple procedure was developed for rapid analysis of animal bone marrow by flow cytometry using the lipophilic cationic dye 3,3′‐dihexyloxacarbocyanine iodide (DiOC 6 (3)). The batch process allows differentiation of bone marrow cells into lymphoid, erythroid, and myeloid populations and enables classification of erythroid and myeloid cells into proliferating and maturing subpopulations. From these data, myeloid:erythroid (M:E) ratios and maturation indices for erythroid and myeloid cells (EMI and MMI, respectively) can be derived. This procedure provides the opportunity to analyze bone marrow quantitatively and offers distinct advantages to current manual methods in terms of simplicity, throughput, and reproducibility. The method has been tested successfully using marrow from Wistar rats, B 6 C 3 F 1 mice, beagle dogs, and cynomolgus monkeys. This technique facilitates the evaluation of bone marrow samples taken from preclinical safety studies or from animal colonies of large size. © 1992 Wiley‐Liss, Inc.

Objective: acute myeloid leukemia is one of the diseases with higher mortality.New technologies for the study of acute myeloid leukemia proteomic are urgently needed.The aim of this study was to screen serum biomarkers in patients with acute myeloid leukemia by using surface-enhanced laser desorption and ionization time-of-flight mass spectrometry(SELDI-TOFMS) technique.Methods: Proteomic spectra were generated by mass spectrometry in 87 cases,including 30 cases of acute myeloid leukemia that had been pathologically confirmed with 5 to 78 years,and 57 cases of healthy control aged 5 to 75 years.87 spectra obtained were used to train and develop a decision tree classification algorithm.Results:A total of 8 distinguished proteomic peaks were detected,7 peaks were used to build a proteomic pattern.The results yielded a sensitivity of 86.7%(26/30)、specificity of 96.5%(55/57).Conclusion:SELDI-TOF-MS offers a unique platform for the proteomic detection of acute myeloid leukemia.It also offers a noninvasive method to further study the proteomic changes in the development and progression of leukemia.

Both acute myeloid leukemia 1 and c‐Fos are regulatory factors of hematopoietic cell differentiation. We identified that the c‐fos promoter contains an acute myeloid leukemia 1 binding site at nucleotide positions −6–+14. c‐fos promoter activity was induced by transient overexpression of acute myeloid leukemia 1 in Jurkat T‐cells, but not by that of the short form of acute myeloid leukemia 1‐MTG8, a chimeric acute myeloid leukemia 1 protein. In 32Dcl3 myeloid cells, stable overexpression of acute myeloid leukemia 1‐MTG8 blocked the c‐fos gene transcription and cell differentiation, but that of acute myeloid leukemia did not. These data suggest that acute myeloid leukemia 1 and acute myeloid leukemia 1‐MTG8 reciprocally regulate the myeloid cell differentiation, possibly by the way of regulating c‐fos gene transcription.

The study by Callera et al., published in this issue of the Revista Brasileira de Hematologia e Hemoterapia (RBHH), 1 reveals evidence of a continuous decrease in the mortality rate in adult patients with myeloid leukemias from 1994 to 2011 in southeast Brazil.3][4] The words myeloid retirar a palavra and leukemia refer to a very wide range of disorders with varying severity and manifestations.We address here CML, AML and APL mainly to better understand the advances in each of them.I believe that there is really a trend in Brazil to follow developed countries which have reported significant changes in prognosis.Unfortunately, we still experience many difficulties, but we have had progress as well: first, tyrosine kinase is widely distributed to patients with CML in Brazil; second, the health authorities have just opened a public consultation about AML treatment, including cytogenetics and the recommendation for the use of molecular tests in the treatment protocol; and third, the results of treatments of LPA show substantial improvement. 3volution in the treatment of leukemia has greatly improved the chances of cure and disease control.More than 7500 people develop leukemia in the country today and 9000
