BACKGROUNDPolygonum multiflorum is one of the leading causes of herb-induced liver injury in China.HLA-B*35:01 is reported to be a potential biomarker of Polygonum multiflorum-induced liver injury (PM-DILI).However, little is known about the relationship between single-nucleotide polymorphisms (SNPs) and PM-DILI. Objective To evaluate the effect of microfiltration on the extract of polygonum multiflorum by inorganic ceramic membrane.Methods The extract of polygonum multiflorum was processed by inorganic ceramic membrane.The character,whole solid,content of stilbene glycoside and the flux of membrane were examined.Results The extract of polygonum multiflorum became clear after microfiltration.The decreasing rate of the whole solid was 67.9%,and the content of stilbene glycoside was 2.04 times that of the extract of polygonum multiflorum.The decay of flux of membrane had a certain regularity.Conclusion The microfiltration of inorganic ceramic membrane has good effect on the extract of polygonum multiflorum.

The effects of different ratio of polygonum hydropiper,mulberry leaf and polygonum multiflorum thunb leaf,culture time and culture tempreture on microbial species and quantity,saccharifying power,liquefying power,and ferementing rate of Xiaoqu were studied.The optimum technical conditions were determined by orthogonal test as follows: the ratio of polygonum hydropiper,mulberry leaf and polygonum multiflorum thunb leaf was 1∶1∶1 or 2∶1∶1 or 1∶2∶1,the optimum culture time was 5 d,and the optimum culture temperature was at 25 ℃.Under the optimum technical conditions,the saccharifying power,liquefying power and fermenting rate of Xiaoqu reached 480 mg/g·h,8.05g/g·h and 68.9 % respectively.

Objective:To observe the change of total anthraquinones content in Polygonum multiflorum Thunb.before and after processing.Method:The anthraquinones content was determined by a direct UV sprctrophotometry.Result: The standards and samples all had the biggest absorbance in range of(222±3)nm.The anthraquinones content of processed Polygonum multiflorum Thunb.was lower than that of Polygonum multiflorum Thunb.Conclusion:The direct UV sprctrophotometry can be used to determine the anthraquinones content from The Polygonum multiflorum Thunb.and the study provides a new experimental basis for clinical use and quality control of Polygonum multiflorum Thunb.

A high-performance liquid chromatographic (HPLC) method with ultraviolet absorption detection was developed to determine the presence of anthraquinones, polydatin, and resveratrol in Polygonum multiflorum Thunb. as well as other medicinal Polygonum species, viz., P. cuspidatum, P. oriental, P. aviculare, and P. vulgare, as well as commercial products that claim to contain P. multiflorum. The best results were obtained with a Phenomenex Gemini C18 column using gradient mobile phase composed of water (0.1% acetic acid) and acetonitrile (0.1% acetic acid). Elution was at a flow rate of 1.0 mL/min. The detection wavelength was 280 nm for anthraquinones and 320 nm for polydatin and resveratrol. The main anthraquinones identified were emodin and physcion. The HPLC pattern of P. multiflorum was also compared with 5 other species of Polygonum. The method was validated for precision, repeatability, and accuracy. The relative standard deviation was between 0.9 and 1.6%. The method was sensitive, quick, and accurate for determination of anthraquinones, polydatin, and resveratrol in 6 different species of Polygonum and can be used for quality control of P. multiflorum. The commercial samples and the 6 Polygonum species were compared microscopically, and a detailed description is provided for P. multiflorum.

The effect of Astragalus mongholicus, Polygonum multiflorum and the combination of Astragalus mongholicus and Polygonum multiflorum on thymocytes apoptosis induced by cyclophosphamide (Cy) was studied. The results were as follows: in the group C (the mice were treated with Cy, Cy mice, the control), their thymocytes revealed obvious apoptosis change, as compared with group C, in group AM, PM and AMPM (the Cy mice were treated for 14 days with Astragalus mongholicus , Polygonum multiflorum and the combination of Astragalus mongholicus and Polygonum multiflorum respectively),using flow cytometry,the sup G1 peaks, labelled ‘Ap’—the hallmark of apoptotic cells appeared, the height of Ap peaks was sequentially in group AMPM AM PM C , and Ap peak did not appear in group O. The ultrastructure showed that apoptotic thymocytes density and degree of morphologic change were sequentially in group AMPM AM PM C. The results suggested that Astragalus mongholicus and Polygonum multiflorum could play an antagonistic role on thymocytes apoptosis induced by cyclophosphamide, and the combination of Astragalus mongholicus and Polygonum multiflorum could give rise to the highest effect.

[Objective] To research the influence of polygonum multi.florum and polygonum multiflorum preparation on immunoglobulin in SD rats. [Methods] SD rats were divided into seven groups. Each group was administrated with drug,30g/kg a day orally for 90 d. The blood samples were taken on the 90 d for determination of serum IgG,IgM levels. [Results] Comparing control group and experimental groups after 60 d administration of polygonum multiflorum,polygonum multiflorum with high-dose hoelen group,polygonum mul-tiflorum with low-dose hoelen group,preparation polygoni multiflori,preparation polygoni multiflori with low-dose hoelen group the serum IgM levels were significantly elevated(P0.05),while the level after 90 d administration of polygonum multiflorum with high-dose hoelen group,polygonum multiflorum with low-dose hoelen group,preparation polygoni multiflori with high-dose hoelen group was significantly increased (P0. 05),the level of serum IgG,however,in group of polygonum multiflorum with low-dose hoelen group only after the 90 d administration was reduced. Between these groups they were statistically significant. [Conclusion] Administration of the polygonum multiflorum,preparation of polygonum multiflorum system for 60 or 90 d can induce a significant increase of serum IgM in rats,that can cause some liver damage.

Objective To compare the antioxidant effects of Polygonum multiflorum Thunb from different provenances.Methods The acute aging mice model was established by subcutaneous injection with D-galactose continuously for 40 days,and the activities of SOD and the contents of malonaldehyde(MDA) in blood and brain after being intragastrically administered with water decoction of Polygonum multiflorum Thunb from different provenances were observed.Results Intragastric administration with water decoction of Polygonum multiflorum Thunb could increase SOD activities and decrease the contents of MDA in blood and cerebral tissue of mice;and the effects varied with different provenances of Polygonum multiflorum Thunb.Conclusion Intragastric administration with water decoction of Polygonum multiflorum Thunb could relieve lipid peroxidation,and had certain anti-oxidant effects,which varied with different provenances of Polygonum multiflorum Thunb.
