    Abstract:
    Abstract Matrine, also known as oxymatrine, is an important active ingredient of traditional Chinese herb Sophora flavescens . Recent studies have found that matrine may inhibit multiple tumors through inhibiting the tumor cell proliferation, inducing cell apoptosis, blocking cell cycle, suppressing cell invasion and migration and assisting in the synergy, and attenuation of radiotherapy and chemotherapy. This study mainly investigated the role of matrine in gastric cancer and its possible mechanism. The real‐time fluorescence quantitative polymerase chain reaction technique showed that matrine inhibited the proliferation and migration of gastric tumor cells and significantly suppressed the expression of miR‐93‐5p. The dual‐luciferase reporter gene assay indicated that AHNAK was a target gene of miR‐93‐5p and regulated by miR‐93‐5p and matrine. The torsion test demonstrated that matrine exerted its role via miR‐93‐5p while miR‐93‐5p played a role by targeting AHNAK. Thus, this study found that matrine affected the progression of gastric cancer by inhibiting the function of gastric cancer cells through the possible mechanism of inhibiting miR‐93‐5p expression to increase the expression level of the downstream target gene AHNAK.

    Objective To study the method for Purification of extraction for Alkaloids from Sophora Flavescens.Methods To use Alkaloids from Sophora Flavescens、matrine、Oxymatrine as index,the macroporous resin was chosen to purify the extraction and the optimized parameters were selected.Results The AB-8 macroporous resin was chosen.Optimum process: pH = 10.0;diameter to height ratio of 1: 3;eluted by 1.5 BV water,elution flow rate was 2 BV / h to washing impurities;then eluted by 6 BV 50% ethanol,elution flow rate was 2 BV / h to elution effective component.Conclusion This optimized technology was stable,feasible.Total matrine is improved by 10.3 times,matrine and oxymatrine content increased 8 times.

    Objective:To develop a method for determining the contents of matrine and oxymatrine in total alkaloids from Sophora flavescens Ait.Methods:A Elite Hypersil NH_2 column(5 μm,250 mm×4.6 mm)was used with the mobile phase being acetonitrile-absolute alcohol-3% phosphoric acid solution(81:10:9),flow rate being 1.0 mL·min~(-1),determinating wavelength being 220 nm,the column temperature being 25℃,The injection volume was 10 μL.Results:The calibration curves of matrine,oxymatrine were in good linearity over the ranges of 5.32- 53.2 μg·mL~(-1)(r=0.9997),65.7-525.6 μg·mL~(-1)(r=0.9995),and the average recovery of matrine and oxymatrine was 99.5 %,99.7 % with RSD of 1.0% ,0.6%(n=6)respectively.Conclusion:The method is simple, rapid,reliable and can be used for the total alkaloids in Sophora flavescens Ait.control.

    [Objective] In order to right use Sophora flavescens plant resource,contents of matrine and oxymatrine of various organs of Sophora flavescens from Liaoning province were tested in the present paper.[Methods] TLC method.[Results] Matrine of seeds is the highest in content among all the tested organs,being up to 0.065% of dry weight,then stems (0.034%),roots(0.016%) and leaves(0.013%);The content of oxymatrine in roots is the highest,up to 3.847% of dry weight,and then seeds(1.982%) and stems(0.140%),no oxymatrine was detected in leaves.The alkaloid content is not significantly different between old rough roots more than 1.0cm in diameter and young thin ones of 0.5~1.0cm in diameter.[Conclusions] Contents of alkaloids in Sophora flavescens plants will keep stabilization in mature plants although the plants still grow;Oxymatrine is more than matrine in most organs of Sophora flavescens;Seeds and stems are appropriate organs for matrine extraction.

    The toxicity of three Sophora flavescens Ait alkaloids(Oxymatrine,sophoridine,matrine) to mice were systematically researched in this paper.Results are as below:the toxicity of Sophora flavescens Ait alkaloids which acted on mice were increased after mixture.In matrine and sophoridine group,c.f value was 59.1.In oxymatrine and matrine group,c.f value was 37.3,showing the synergy effects;In oxymatrine and sophoridine group,c.f value was 13.6 which indicated that had additive effect.The results of blood routine determination showed that WLT,PLT and GR were higher than those of control(P0.01).Examining the blood biochemical indicators,in the oxymatrine and sophoridine group,BUN and Cr of samples were all higher(P0.01) than those of control,In the matrine group,the quality of ALT and AST were higher than those of control(P0.01).The coefficients of kidney,heart,liver were significant higher than those of control from the statistical results of organ coefficients(P0.01,P0.05,P0.05).According to the above results,the target organs on which Sophora flavescens Ait alkaloids acted were kidney and liver.

    The contents of matrine and oxymatrine in the wildness Sophora flavescens Alt and Sophora viciifoli were detected by HPLC with ZORBAX SB-C18 (150mm×4.6mm, 5μm) column at 20℃. The mobile phase consisted of 0.04 mol/L H3PO4 (pH=1.8)-CH3OH (90:10).The flow rate was 1.0 mL/min and detection wave length was at 220 nm. The results revealed that the high content of matrine in the wild Sophora flavescens Alt especially in its root was observed. The content of matrine in root reached 0.34%. On the other hand, 2.45% oxymatrine in the Sophora viciifoli seed which was higher than Sophora flavescens Alt indicated that Sophora viciifoli had a good utilization value for producing matrine.

    Objective:To establish a high performance liquid chromatography(HPLC)method to determine the contents of matrine and oxymatrine in Sophora flavescens Ait.for selecting its suitable planting areas.Methods:A YWG-NH_2 was used as the solid phase,acetonitrile-3% phosphoric acid-ethand(80∶10∶10)as the mobile phase,with a detection wavelength at 220nm.Results:The calibration curves of matrine and oxymatrine were in good linearity over the rang of 0.050~0.061μg and 0.372~4.464μg,respectively.Conclusion:Sophora flavescens growing in Licheng,Zuoquan,Linfen in Shanxi province and Taibai in Shaanxi province contained higher oxymatrine than 3%.Content differences of matrine were smaller in different growing places.

    Objective To develop a method for determinition matrine and oxymatrine of crude extract of Sophora flavescens. Methods AichromBond-AQ C18 column(4.6 mm×250 mm,5 μm)was used as the solid phase.Methanol-0.05% triethylamine solution(70:30,V/V)was taken as mobile phase.The flow rate was 1.00 ml/min.The UV detection wavelength was 215 nm. Results The calibration curves of matrine and oxymatrine were linear in the range of 0.02~160 mg/L and 0.04~100 mg/L,respectively.The average recoveries of both were 98.59% and 99.43%,respectively. Conclusion The method established method is sensitive,simple,and accurate.

    Objective: Determination of matrine and oxymatrine in Sophora flavescens from different regions by HPLC.Method:Chromatography was perfomed on a 5μm Waters Spherisorb NH_2(4.6mm×250mm)at 25℃.The mobile phase was composed of acetonitrile-ethanol-3% phosphorous acid solution(80∶10∶10)with a flow rate of 1.0ml·min~(-1).The detective wave is 220nm.Conclusion: The contents and proportion of matrine and oxymatrine in Sophora flavescens from different regions are very distinct,this can provides the foundation for the quality evaluation of Sophora flavescens from different regions.

    Objective To observe the changes in the contents of matrine and oxymatrine after decocting flavescens for different times so as to provide the reference for the application of flavescens in compound formula.Methods HypersiL NH2(250 mm × 4.6 mm,5 μm)column was adopted.Ethanol-3% phosphoric acid(85∶7.5∶7.5)was taken as a mobile phase,the flow rate was 1.0 mL·min-1,and the detecting wavelength was 210 nm.Sample volume was 10 μL.Results While flavescens was decocted for longer time,matrine content increased and oxymatrine content did not change greatly.Conclusion In flavescens extraction,the time of decoction is not the chief factor to impact the transformation of matrine from oxymatrine.

    Alkaloid from Sophora flavescens Ait was extracted with three methods: ultrasonic method, circumfluence of condensation,and dipping in normal temperature. The extractives are mostly matrine and oxymatrine.The effect of extractives on lustrating Trichodinella of fish was also studied.The results showed that the alkaloid from Sophora flavescens Ait could lustrate Trichodinella ninuta efficiently. With the different extractives through different extracting methods,the effect on lustrating Trichordinella nenuta of fish were also different. The content of matrine was highest in extractive by ultrasonic method among three methods and the effect on lustrating Trichodinella of fish was also the best,which meant that the matter lustrating Trichodinella is matrine.
