    Abstract:
    Previously, Rhodobacter sphaeroides strain HJ was isolated to obtain a purple nonsulfur bacterium with the ability to produce high levels of hydrogen from acetate. However, the genome of this strain has not been previously sequenced. Therefore, the complete genome sequence of R. sphaeroides strain HJ is presented in this report.

    嫌気・光照射下で運転する紅色非硫黄細菌を用いた廃水処理・有価物生産システムは, その光合成従属栄養的増殖により, より多くの有価物 (菌体) を生産すると同時に温暖化ガスの排出を抑制するシステムになると考えられる. 本論文においては, 紅色非硫黄細菌 Rhodobacter sphaeroides の脂肪酸基質での無機化率を定量し, 酪酸より炭素鎖の長い基質では二酸化炭素の発生がなく逆に摂取することを示した. また, 菌体内PHB (poly-3-hydroxybutyrate) が他の菌体内成分に変換される際の二酸化炭素の発生, 摂取がほとんどないことを明らかにした. さらに, 菌体転換率と比増殖速度から, 菌体生産, 廃水処理に適した廃水原水の前処理について検討した.

    Purification procedures to obtain light-harvesting complexes were performed on two photosynthetic bacteria, i.e. Rhodospseudomonas (Rps.) palustris and Rhodobacter (Rb.) sphaeroides. In this study, purification was initially carried out using series of centrifugation towards the bacteria cell to acquire chromatophore of the bacteria. Application of detergent (lauryldimethylamine oxide, or LDAO) to the chromatophore enabled the extraction of membrane containing photosynthetic apparatus. Further purification involved the application of sucrose density gradient centrifugation and ion exchange chromatography. The purity of collected LH2 complexes can be determined by calculating the ratio of AB850 : Aprotein. The resulting LH2 complexes from both bacteria showed relatively high ratio suggesting the purity of the complexes.

    For the first time, properties and structural peculiarities of exopolysaccharides produced by phototrophic bacteria Rhodobacter sphaeroides isolated from hydrocarbonate mineral waters in Armenia with a degree of mineralization in limits of 3.5-5.0g/l,pH 7.2 have been studied.It is noteworthy that despite membership of the isolated strains in one and the same genus, these strains are significantly different by the nature of the produced exooligosaccharide.The studies implemented show that phototrophic bacteria are able to synthesize exopolysaccharides with different compositions and structures: cyclic and linear, depending on the cultivation conditions.
