    Objective To investigate the role of neuro-specific DNA repair gene Nbn in mouse hippocampus development by observing the morphological differences in Nbn-CNS-Del and Nbn-CNS-Ctr mice.Methods Serial sections and stereology were used to quantitatively analyze the development of mouse hippocampus on postnatal day 7,14 and 21.Results Compared with that of the control group,the hippocampus development of Nbn-CNS-Del mice lagged behind.In Nbn-CNS-Del mice,on day 21,the profile area of pyramidal layer of hippocampus and granular layer of dentate gyrus decreased(P0.05),and the volume densities in granular layer and polymorphic layer of dentate gyrus,and CA4 had significant differences(P0.05).Conclusion DNA repair gene Nbn,an important gene in mouse hippocampus development,affects the development and maturation of hippocampus and granular layer of DG in mice.

    AIM: To investigate the expression and distribution of p CREB in the hippocampal formation of adult rats. METHODS: SP immunohistochemical staining and immunofluorescence were performed in paraformaldehyde fixed hippocampal sections. RESULTS: p CREB was expressed throughout the hippocampal formation, with the expression in the dentate gyrus being the highest, CA3 area, the second, and CA1 area, the least. P CREB was also found to have expressed mostly in nucleus. CONCLUSION: The expressions of p CREB are different in the dentate gyrus and CA1 area, suggesting that dentate gyrus and CA1 area may play different roles in long-term memory.

    ObjectiveTo explore the possible injury mechani sm in brain of rat irradiated by EMP through observing the changes in expression of CFOS in hippocampus and dentate gyrus.MethodsThe group A, group B and group C were irradiated by high field intensity(6104,2104 a nd 6~{!A~}103 V/m respectively)electromagnetic puls es(EMP)for 5 times within 2 minutes respectively and no treatment was given to g roup D(control group).The rats were sacrificed at 1, 6, 12,24 and 48 h,sep arately. Immunohistochemical method was used to investigate the expression of C FOS and image analysis for CFOS p ositive cells was performed by CMTAS~{*2r~} image analysis system at a magnificatio n of 400. Results The rats of irradiated groups had high expres sion of CFOS in neurons of hippocampus and dentate gyrus at 6-12 h, and the nu mber of CFOS positive cells was significantly increased as compared with contr ol group ~{*)~}(P0.05)~{**~}.Conclusions The EMP could cause significant high CFOS expression in hippocampus and gyrus,which may pla y an important role in the injury of rat brain by EMP.

    To study the expression of Goalpha mRNA in hippocampus and dentate gyrus of 7-day rats with hypothyroidism.We use wistar rat model of hypothyroidism and non-radioactive in situ hybridization technique to observe the changs of Goalpha mRNA levels in hippocampus and dentate gyrus of 7-day rats with or without hypothyroidism.Perinatal hypothyroidism can increase Goalpha mRNA levels in all areas of hippocampus and dentate gyrus of 7-day rats.Thyroid hormone can down-regulate Goalpha gene transcription in hippocampus and dentate gyrus during the early development of rat brain.

    Objective To evaluate the hippocampal neurogenesis in depression.Methods A total of 20 Wistar rats were randomly divided into control and depression group(model group).Chronic unpredicted stress model was established.Body mass weigh and behavioral scores of the rats were observed.Radioimmunoassay was used to measure plasma concentrations of corticotropin releasing hormone,adrenocorticotropic hormone and cortisone,Light and electron microscope were used to detect the morphological change of hippocampal CA3 neurons.Immunohistochemistry and in situ hybridization were used to detect both protein and mRNA expressions of Nestin in hippocampal dentate gyrus.Results Compared with the control group,rats in the model group,both body mass weigh and the related increase rate were markedly decreased,and the behavioral scores were reduced significantly.In addition,neural damage was observed in hippocampal CA3 neurons,and the Nestin expressions in hippocampal dentate gyrus were evidently increased.Conclusion Hippocampal neurogenesis was increased in depression.
