    細胞学的にherpessimplexvirus (HSV) 感染と診断された患者から得られた婦人科スミアに対し, 細胞学的・免疫細胞化学的観察を行った.観察した365個の細胞は形態学的にHSV感染と診断できる250個とHSV感染の疑われた115個の細胞に分けた.そして, Papanicolaou標本脱色後に抗HSV type II家兎血清を用い, 酵素抗体間接法で染色した.免疫陽性反応はHSV感染と診断された細胞では250個中223 (89.2%) 個, HSV感染が疑われた細胞では115個中93 (80.9%) 個にみられた, よって, 細胞学的にHSV感染が疑われた症例のPapanicolaou脱色標本に酵素抗体法を応用することの意義を認めた.しかし, 細胞質が変性により好酸性を示した感染細胞では, 著しい免疫反応の低下がみられた.細胞形態学的にHSV感染が疑われた細胞の核が好塩基性小顆粒を伴う空胞を含むことが重要であった.さらにPapanicolaou標本において重積性や濃染性核の存在もHSV感染の診断のために無視すべきでないと思われる.

    Herpes Simplex Virus (HSV) specified polypeptide antigens were analyzed by immunoblotting and enzyme-linked immunosorbent assay (ELISA) methods. In general, the virus specified proteins showed marked cross-reactivity. However, the HSV-1 specified glycoprotein C could be detected by HSV-1 infected rabbit antiserum only. Two low molecular weight polypeptides showed considerable reactivity with HSV-2 induced immune sera as revealed by an immunoblocking modification of the immunoblotting method. These proteins may prove useful in the serological diagnosis of HSV Type 1 and Type 2 infections.

    日本, スウェーデン, ケニアの3カ国で分離された疫学的に関連のない, それぞれ104, 68, 32株, 計204株のHSV 1分離株について4種の制限酵素 (BamH I, Kpn I, Sal I, Hind III) を用いてゲノムを比較した。4制限酵素による130の切断点のうち27の切断点において, 3カ国のうち, いずれかの2カ国間で有意差が認められた。すなわち, 日本とスウェーデン間で17, 日本とケニア間で22, スウェーデンとケニア間で17切断点に有意差が認められた。27切断点の変異による断片のうち, 特定の1カ国の分離株にのみ存在する, または, 存在しない断片が合計20認められた。21切断点の変異をパターン化し204株のHSV 1株を分類したところ92の異なる切断パターンに分類され, ケニア株1, スウェーデン株2株に共通した1パターンを除いて, それぞれの国の分離株は異なる切断パターンに分類された。以上の結果は地理的または人類学的に離れた国または人種間では, 分布しているHSV 1のゲノム構造が極めて異なることを示唆している。

    The AmpliVue HSV 1+2 assay was compared to the ELVIS HSV ID and D(3) Typing Culture System for the qualitative detection and differentiation of herpes simplex virus 1 (HSV-1) and HSV-2 DNA in 1,351 cutaneous and mucocutaneous specimens. Compared to ELVIS, AmpliVue had sensitivities of 95.7 and 97.6% for detecting HSV-1 and HSV-2, respectively. Following arbitration of discordant results by an independent molecular method, the AmpliVue assay had a resolved sensitivity and specificity of 99.2 and 99.7%, respectively, for both HSV-1 and HSV-2, whereas ELVIS had a resolved sensitivity of 87.1% for HSV-1 and 84.5% for HSV-2.

    Objective To investigate the situation of patients with genital suspected herpes simplex virus(HSV) infection in Xintang,Guangzhou.Methods To test herpes simplex virus(HSV-I/II) of 1202 cases of outpatients by fluorescence quantitative polymerase chain reaction technology(FQ-PCR).Results The positive rate of 1202 casas of outpatients were 0.67%(8/1202) with HSV-I and 23.46%(282/1202) with HSV-II.The total positive rate of HSV infection were 24.13%(290/1202).There was significant difference between the positive detection rate of HSV-I and HSV-II(P0.01).The positive rate of HSV in male and female patients were 24.62%(193/748) and 23.21%(97/418),respectively.There was no significant difference of the positive detection rate between male and female(P0.05).Conclusion ① The detection rate of HSV in the outpatients with suspected genital herpes HSV infection was high in Xintang area,which nearly a quarter.The infection rate was difference in different types of HSV,and the main type was HSV-II.The result illustrated that clinical diagnosis should pay attention to detect HSV in outpatients with suspected genital herpes virus in order to avoid misdiagnosis.② There was no difference of HSV infection between male and female.③ The features of FQ-PCR were simplicity,high sensitivity,fast in detection of HSV and it is worth for clinical application.

    While superinfection with different herpes simplex virus (HSV) types has been demonstrated in animals, the ability of the two HSV types to colonize and reactivate in the same anatomic region in humans has not been well demonstrated. In 6 patients, both HSV-1 and HSV-2 was recovered from genital lesions. In 4 of them, who initially acquired genital HSV-1 infection, subsequent HSV-2 infection presented as a prolonged episode of genital lesions and a marked increase in the frequency of genital recurrences. While most of the subsequent clinical reactivations were HSV-2, in 2 patients the recurrence rate of genital HSV-1 increased after the acquisition of HSV-2. These data demonstrate the ability of a second HSV type to infect the same anatomic region and illustrate the difference in reactivation frequency of the two types in the same person. Typing of HSV isolates may be useful in persons with recent alteration in recurrence rates of genital HSV.
