Colon targeted drug delivery system (CTDDS) delivered as both local and systemic delivery of drugs. Local delivery could, for example, allow topical treatment of inflammatory bowel disease (IBD). Treatment could be enhanced when drug delivered to the target site on the colon. Systemic side effects could also be reduced. Colon specific systems is most important delivery of those drug which are normally inactivated in the upper parts of the gastrointestinal tract (GIT). Primary approaches for CTDDS (Colon targeted Drug Delivery System), which includes prodrugs, pH and time dependent systems, Bacterial enzyme dependent colonic DDS and pH and bacterial enzyme dependent colonic DDS. The novel approach of CTDDS, which includes pressure controlled colonic delivery capsules (PCDCS), CODES and osmotic controlled drug delivery are specific technique.

Oral colon targeted drug delivery system can be divided into pH dependent, time-dependent, enzymatic degradation etc. according to the ways of drug delivery. Mechanism of drug delivery, preparation method and application of oral colon targeted drug delivery system were introduced. In addition, it previewed the research direction of colon targeted drug delivery in the future, to provide a reference for the targeted new dosage form.

The last two decades of developments in drug formulations and novel drug delivery systems have been seen as the beginning of a new era leading to increased patient adherence and pharmacological response to the therapeutic regimen. One of the most difficult tasks is efficiency and target‐specific drug delivery or the extent of delivery at any given site of interest. Many currently designed drug delivery systems are precisely tailored to maximize the delivery of a particular form of drug by reducing the degradation or loss of the drug. In case of cancer treatment, the targeted drug delivery is of utmost importance as the anticancer agents are not having the ability to differentiate between healthy and tumor cells resulting in adverse effects and/or systemic toxicity. The targeted drug delivery is thus designed to focus on preventing side effects and encouraging the accumulation of the drug at the targeted site; one such promising drug delivery system is magnetosome drug delivery, i.e., drug delivery using magnetosomes (biological magnetic nanoparticles). In this article, we have summarized the system for design, development, and mode of drug delivery using magnetosomes along with the recent developments made in this field to facilitate the diagnosis and treatment of cancer.

Targeted delivery combined with controlled drug release has a pivotal function in the future of personalized medicine. Target drug delivery system is one of the most considerable novel approach towards drug delivery system. Targated drug delivery seeks to concentrate the medication in the tissues of interest while reducing the relative concentration of the medication in the ultimate tissues thus enhancing therapeutic index and bioavailability at site specific-delivery .Now a days various carrier systems which includes liposomes, niosomes, aquasomes,pharmacosomes, dendrimers, nanoparticles, microspheres, solid lipid nanoparticles, resealed erythrocytes etc. are utilized in target drug delivery system which provide site specific drug delivery. Drug targeting is the principle by which the distribution of drug in organism is maneuverer in manner such that its major fraction interacts exclusively with the target tissue at the cellular and subcellular. The present review deals with the Targeted drug delivery system its advantages, disadvantages, need of Targeted drug delivery system, Types, drug targeted to a specific organ such as Brain, kidney, heart, colon and respiratory tract and research update on Targeted drug delivery system. Various drug carriers which can be used in this advance delivery system are Niosomes, Liposomes, Nanoparticles, Monoclonal Antibodies.

In field of medicine oral route is mostly used path for the administration of drug. so, the oral route is not preferred for lower GIT disorder. So, to overcome this problem drug directly targeting to the specific site. In this review article overview on the colon targeting drug delivery system. Colon is the site where drug is targeted on the local site and used for the local colon disease. Colon targeting drug delivery system not just for the local disease but also used for the systemic delivery of the drug such as proteins, therapeutic peptides, anti-asthmatic, anti-hypertensive drug, antidiabetic drug. Now a day number of method are used to formulate CTDDS such as formation of pro drug, coating of polymers etc. colon targeting drug delivery system is a new system for targeting a drug in specific site for enhance the therapeutic effect of poorly absorbed drug. And now a day drug targeted on colon through rectal route. But this route is very painful. In this article discuss about the anatomy of the colon, factor affecting the colon targeting drug delivery system, advantages of colon targeting drug delivery system, limitation of colon targeting drug delivery system and evaluation of the colon targeting drug delivery system. The colon targeted drug delivery has a number of important implications in the field of pharmacotherapy. Oral colon targeted drug delivery systems have recently gained importance for delivering a variety of therapeutic agents for both local and systemic administration. Targeting of drugs to the colon via oral administration to protect the drug from degradation or release in the stomach and small intestine. It also ensures abrupt or controlled release of the drug in the proximal colon. Various drug delivery systems have been designed that deliver the drug quantitatively to the colon and then trigger the release of the drug. This review will cover different types of polymers which can be used in the formulation of colon targeted drug delivery systems.

In the last two decades colon targeted drug delivery has gained increased importance not just for the deliver drugs for the treatment of various colonic diseases but also for its potential for delivery of proteins and therapeutic peptides. In the past various traditional approaches used for colon targeted delivery like prodrugs, pH, time dependent, and microflora activated systems, have achieved limited success. For successful colon targeted drug delivery, the drug needs to be protected from absorption and/or the environment of the upper gastrointestinal tract and then be abruptly released into the colon. Hence continuous efforts have been made on designing colon targeted drug delivery systems with improved site specificity and versatile drug release kinetics to fulfill different therapeutic needs. In last couple of years few new systems have been developed for colon targeted drug delivery such as pressure dependent systems, CODES™ technology, micrsponges, pectin and galactomannan coating, microbially triggered osmotic systems, lectins and neoglyconjugated etc. which are reported to have better in-vivo site specificity and design rationale than the earlier approaches. This review article gives an overview of various approaches for colonic targeted drug delivery with emphasis on newer systems, their merits and demerits, in vitro/ in-vivo evaluation and market status of such delivery systems.
