The anti-inflammatory, analgesic, antipyretic and ulcerogenic activities and acute toxicity of 2-oxo-3-[4-(1-oxo-2-isoindolinyl)-phenyl]-butanamide (GP 650) were investigated in laboratory animals and compared with those of phenylbutazone, acetylsalicylic acid (ASA), ibuprofen, tiaramide-HCl and tinoridine-HCl. The anti-inflammatory and analgesic activities of GP 650 were found to be almost equivalent to those of phenylbutazone on the basis of various anti-inflammatory and analgesic tests while its antipyretic activity was less marked, and its ulcerogenic action and acute toxicity were much lower than those of the other agents. It is interesting to note that GP 650 possesses marked anti-inflammatory activity similar to acidic anti-inflammatory agents in chronic inflammatory models along with mild antipyretic and ulcerogenic activities found in basic anti-inflammatory agents. Therefore, GP 650 appears to be a promising anti-inflammatory and analgesic agent.

The analgesic and antipyretic effects of oxaprozin were investigated in comparison with those of indomethacin, ibuprofen, phenylbutazone and aspirin. On the various writhing tests in mice, the analgesic effect of oxaprozin was about 2 to 9 times more potent than those of ibuprofen, phenylbutazone and aspirin. On the other hand, the analgesic and antipyretic effects of oxaprozin in rats were roughly equivalent to those of aspirin, but less effective than those of the other drugs tested. On the urate synovitis test in dogs, only oxaprozin showed a prophylactic effect. Therefore, The effect of oxaprozin in mice and dogs was more potent than ibuprofen, phenylbutazone and aspirin. The metabolic rate of oxaprozin in rats is 3.5 and 7.2 times more rapid than in mice and dogs, respectively, and its blood level in rats is low. Moreover, the biological half-life of oxaprozin is 39 to 43 hr and 49 to 69 hr in dogs and humans, respectively. From these results, it is suggested that oxaprozin is more potent than ibuprofen, phenylbutazone and aspirin, and in clinical use, it is a long acting anti-inflammatory drug.

The HeI photoelectron (PE) spectra of the drugs antipyrine (phenazone, 1), aminopyrine (amidopyrine, 2), phenylbutazone (3), and indomethacin (4) are reported. These drugs possess analgesic-antipyretic and anti-inflammatory activity, and have been in clinical use for many years. Their electronic structure, which is important to their biological activity, is determined by the application of composite molecule methods to an analysis of the PE spectra.

Fenclozic acid (2-(4-chlorophenyl)thiazol-4-ylacetic acid; I.C.I. 54,450; "Myalex") is one representative of a new class of compounds with antiinflammatory, analgesic and antipyretic properties as evidenced by its activity in a variety of tests in rats, mice and guinea-pigs.2. In tests of short duration the potency of fenclozic acid is similar to that of phenylbutazone.3. In tests of longer duration fenclozic acid is more potent than phenylbutazone.4. The activity of fenclozic acid is not mediated by stimulation of the adrenals and the compound is devoid of corticosteroid-like activity.

1-Benzyl-3-(2,3-dihydroxypropoxy)-indazole (benzidol) is a novel indazolane derivative with marked antiinflammatory, analgesic and antipyretic properties. Its acute toxicity is three times less than that of benzidamine and five times less than that of phenylbutazone. Rats, mice, guinea pigs and rabbits treated with benzidol during 6 months at doses of 5, 50 and 200 mg/kg did not present any pathological alteration. The anti-inflammatory activity of benzidol is similar to that of phenylbutazone and higher than that of benzidamine. The analgesic and antipyretic properties are also considerable. Benzidol is better tolerated than the mentioned drugs with similar activities.

A series of indan-1 acids were synthesized and screened for anti-inflammatory activity. All the reported compounds showed varying degrees of anti-inflammatory activity in carrageenin-induced paw oedema test. They also exhibited appreciable antipyretic and analgesic activity in various animal test models. Among these compounds 6-methoxy-indan-1-acetic acid (compound 11) and 5,6-dimethoxy-indan-1-acetic acid (compound 12) showed activity profile close to that of phenylbutazone having prolonged action and lower toxicity than the latter.
