Abstract:
OBJECTIVE:To explore the expressions of GST-π,LRP and TopoⅡ in triple negative breast cancer,and study their clinical significance.METHODS:Totally 252 breast cancer patients without preoperative chemotherapy and endocrine therapy were divided into TNBC(triple negative breast cancer) group and the non-TNBC(non-triple-negative breast cancer) group according to the expression of ER,PR and HER-2.The expressions of GST-π,LRP and TopoⅡ were investigated using the immunohisto chemistry method in above two groups.RESULTS:Of all breast cancer patients 18.7%(47/252) were triple negative breast cancer.GST-π expression in the TNBC group was significantly higher than that in the non-TNBC group(χ2=10.466,P=0.001).No significant differences were found in LRP and TopoⅡ expression between the two groups(P=0.678,P=0.777).CONCLUSIONS:TNBC has higher drug resistance that other subtypes.High expression of GST-π may play an important role init.Keywords:

Objective To investigate the expression of p53 and Ki67 in breast cancer with negative estrogen receptor(ER),negative progesterone receptor(PR) and negative human epidermal growth factor receptor 2(HER-2)(triple negative breast cancer),and explore its clinical significance.Methods A total of 233 cases of invasive breast cancer confirmed by pathology were collected,among which 44(18.88%) were triple negative breast cancer.The expression of p53 and Ki67 in tissues of triple negative breast cancer was detected by immunohistochemical EnVision two-step methods.The tissues of 189 cases of non-triple negative breast cancer were served as controls.The correlation of expression of p53 and Ki67 in triple negative breast cancer with clinicopathological features and their relationship with prognosis were analysed.Results The positive expression rates of p53 and Ki67 in tissues of triple negative breast cancer were 45.5% and 68.2% respectively,which were significantly higher than those in tissues of non-triple negative breast cancer(20.6% and 47.1%)(P0.05).In triple negative breast cancer,the expression of p53 correlated with tumor size,histological grade,lymph node status and vessel invasion(P0.05),and the expression of Ki67 correlated with histological grade(P0.05).The expression of p53 was significantly related to that of Ki67 in all the breast cancer cases(r=0.294,P=0.000),but the expression of p53 was not significantly related to that of Ki67 in triple negative breast cancer cases(r=0.134,P0.05).In all the breast cancer cases,the overall survival of cases with positive expression of p53 was lower than that of cases with negative expression of p53(P=0.000),and the overall survival of cases with positive expression of Ki67 was lower than that of cases with negative expression of Ki67(P=0.000).In triple negative breast cancer cases,there was no significant difference in the overall survival between cases with positive expression of p53 and those with negative expression of p53(P0.05) and between cases with positive expression of Ki67 and those with negative expression of Ki67(P0.05).Conclusion The expression of p53 and Ki67 is up-regulated in tissues of triple negative cancer,while the expression of p53 is not significantly related to that of Ki67.The expression of p53 and Ki67 may be related to the prognosis of breast cancer,while may not indicate the prognosis of triple negative breast cancer.

Objective To study the clinical value of Na+/I-symporter(NIS) expression on triple-negative breast cancer and prediction of 131 I therapeutic effect. Methods 30 samples of triple-negative breast cancer(triple-negative breast cancer,TNBC)were randomly selected as the experimental group,and 30 samples of non-triple negative breast cancer(non-triple-negative breast cancer,non-TNBC)and adjacent tissue of above 30 samples of the triple negative breast cancer,randomly selected as a comparable group,NIS expression was detected by SP immunohistochemical technique and compared with their the differences. Results In triple-negative breast cancer,its adjacent tissue and non-triple negative breast cancer,the positive rates of NIS expression were 66.7 %(20/30),73.3 %(22/30) and 6.7%(2/30) respectively,NIS in Non-triple negative breast cancer group and triple-negative breast cancer group showed no statistic differences(P0.05),triple-negative breast cancer group and its adjacent tissue group showed differences significantly(P0.05),non-three-negative breast cancer group and adjacent tissue of the above triple negative breast cancer tissue group showed differences significantly(P0.05). Conclusion NIS is strongly expressed in a significant proportion of triple-negative breast cancer cells,suggesting a potential role for NIS-directed 131I-radioablative strategies in this patient population.

Objective To analyze the expression of P53 in triple negative breast cancer(TNBC) and the relationship with histological grade.Methods The expression of P53 in 35 cases of TNBC and 35 cases of non triple negative breast cancer(NTNBC) were detected by immunohistochemistry and was analyzed with histological grade.Results The expression of P53 in TNBC was more pronounced than it was in NTNBC(P 0.05);the high histological grade of TNBC was more than NTNBC(P 0.01);the expression of P53 had a positive correlation with histological grade(P 0.01).Conclusion P53 can be taken as an important factor in the differentiation grade,biological behavior and diagnosis of the breast cancer.

Objective To investigate the expression of MDR1,GST-π and Topo-Ⅱ in primary breast cancer and to reveal the relationship with clinical pathological features.Methods 115 cases of breast cancer were examined to observe the expression of MDR1,GST-πand Topo-Ⅱ with immunohistochemical staining method(S-P method).Results The positive expressions of MDR1 and GST-π in breast cancer were 37.4%(43/115),27.8%(32/115) respectively.The positive rate was significantly higher in clinic stages Ⅲ than in clinic stages Ⅰ and Ⅱ(P0.01).The positive expression of Topo?Ⅱ was 48.7%(56/115),and the positive rate was significantly higher in the non?infiltrating carcinoma than in the infitrating carcinoma(P0.01).Conclusion Analysis of the expression of MDR1,GST-π and Topo-Ⅱ in breast cancer may contribute to the selection of drugs in chemotherapy and prognosis evalution of breast cancer.

Objective To determine the expression of integrin-linked-kinase in the triple negative breast cancer.Methods Immunohistochemistry SP method was used to evaluate the expression of ILK in 60 cases of triple negative breast cancer,36 cases of non-triple negative breast cancer,and the prognostic status was evaluated in combination with the follow-up data of those breast cancer patients.Results The expression level of ILK was significantly higher in the triple negative breast cancers than that of non-triple negative breast cancer.In the triple negative breast cancer,ILK expression was associated with tumor size(P0.05),axillary lymph node metastasis(P0.05),and TNM stage(P0.01),but it was not associated with patients'age and menopausal status.Kaplan-Meier survival curve showed that the ILK-rich tumor group had a higher relapse rate and mortality rate compared with the negative breast cnacer.Conclusion ILK status is one of the significant prognostic indicators of triple negative breast cancer.

Objective:To investigate the expression of Ki67 in triple negative breast cancer and the correlation between the expression of Ki67 and C-erbB-2.Methods: The expression of Ki67 and C-erbB-2 in all 320 breast cancer samples,273 were non-triple negative and 47 were triple negative breast cancer,were analyzed by immunohistochemistry.Then the relevance between Ki67 and C-erbB-2 expression levels were analyzed by statistical methods.Results: The Ki67 was over expressed in triple negative breast cancer rather than non-triple negative breast cancer(P0.01).And there was a positive correlation between Ki67 and C-erbB-2 expression levels in breast cancer(P0.01).Conclusion: Ki67 is overexpressed in triple negative breast cancer patients.

Objective To investigate the expression of p57 and ki67 in human triple-negative breast cancer(TNBC) and discuss the clinical pathological significance.Methods Using SP immunohistochemistry method,the expression of p57 and ki67 in 56 TNBC cases and 30 breast tissue adjacent to cancer cases were examined.Results The expression of and P27 in human breast cancer was located within the nucleus.①The positive expression rates of p57 and ki67 were 32.14% and 71.43% in TNBC,moreover lower and higher than the adjacent normal tissue,respectively(P0.05).②In the 56 cases of TNBC,the positive expression of p57 and ki67 in histological grade G3 was significantly higher than that in G1 and G2(P0.05).In addition,the positive expression of p57 in stage Ⅲ tumors was also higher than that in stage I-II tumors(P0.05).Furthermore,P27 expression was associated with axillary lymph node metastasis(P0.05),and was not related with histopathological type(P0.05).③Correlation analysis showed a negative correlation between p57 and ki67(P0.05).Conclusions p57 and ki67 were highly expressed in TNBC and their expressions were closely related with the occurrence,development and metastasis of TNBC.

Objective To study the expressions of P-glycoprotein(P-gp),glutathione-S-transferases π(GST-π) and topoisomerase Ⅱ(TopoⅡ) in breast carcinoma tissues and their significance.Methods Seventy-eight breast carcinoma tissues obtained from the patients without chemotherapy before operation were detected for the expressions of P-gp,GST-π and TopoⅡ by immunohistochemical method.Results In breast carcinoma tissues,the positive expression rate of P-gp,GST-π and TopoⅡ was 38.5%,46.2% and 73.1%,respectively.The difference of the expressions of 3 genes in the patients' age,size of tumor,expression of ER/PR and HER_2 was not significant(P0.05).There was significant relationship among the P-gp,GST-π positivity and lymph node metastasis(χ~2=5.175,χ~2=4.614,P0.05) as well as between the P-gp and TNM stage(χ~2=5.848,P0.05). Conclusion The expressions of P-gp,GST-π and TopoⅡ are related to the primary multidrug resistance in breast carcinoma tissues.Detecting the expressions of the three genes before chemotherapy will provide evidence for the selection of drugs and for the judgement of the prognosis.

Objective To observe the expression and significance of C-erbB-2 and GST-πin breast cancer.Methods The expression of C-erbB-2 and GST-πwere examined by using Elivision immunohistochemical method in 128 specimens of breast cancer.Results In 128 cases of the breast cancer,the positive expression rate of C-erbB-2 and GST-πwas respectively 53.13%(68/60) and 35.94%(46/82);there was significantly difference among the expression of C-erbB-2 and GST-π and lymphatic metastasis,differentiation,status of ER and PR of breast cancer(P0.01);the expression of C-erbB-2 increased with tumor diameter,but no statistical difference between the expression of GST-π and tumor size(P0.05);C-erbB-2 and GST-πhad positive correlation with each other(P0.01).Conclusion C-erbB-2 and GST-π has cooperatively in lymphatic metastasis and the increase of malignant degree of breast cancer.
