    Effect of Different Processing on Transgenic Ingredients and Regulatory Elements of Soybean

    Abstract:
    Based on generally modified soybeans as materials,used the qualitative research methods of PCR to study the effects to Lectin gene and exogenous gene of Cp4 epsps and promoter and terminator by crushing and sticky-heating and extrusion processing.Researches showed that endogenous gene degraded below to 407 bp through processing at 100℃ sticky-heating for 15 min.Exogenous gene degraded below to 408 bp after 100℃ sticky-heating for 15 min.Promoter and terminator turn to degrade by 100℃ sticky-heating for 3 min.The results also indicated that gene sections had no changes through mechanical disintegration and liquid nitrogen processings.While promoter and terminator showd no degrades through extrusion processing,on the contrary,endogenous gene degraded below to 836 bp and exogenous gene degraded below to 1 512 bp.

    In order to assess the effect of food processing on the degradation of endogenous gene in transgenic rice,fermented glutinous rice and rice cracker were made by using transgenic rice with Bar and Bt gene as main material. The degradation of endogenous gene(SPS gene) was detected by using different fragments-size PCR primers. It was found that the stability of gene was different under different processing conditions. The fragment of 168 bp is characteristic of good stability.

    In this study,the Lox-3 promoter was cloned from the rice cultivar Koshihikari,and the length of the promoter is 1 343 bp,which absolutely matches the corresponding sequence of Nipponbare genome.Bioinformatics analysis indicated the promoter sequence contained a common signal-response cis-element(G-box),elements responsive to temperature(CCGAAA)and to moisture stress(CACATG).Furthermore,transgenic plants driven by this promoter was obtained.Reporter gene expression was detected in roots,stems,leaves,anthers,seed capsules and embryos of transgenic plants,as well as in the embryos and the plumule rather than in endosperm,of germinating transgenic seeds.A quantitative analysis revealed that the reporter gene was induced by exogenous application with ABA,NaCl and artificial aging treatments,and these effects were supposed to be mediated by the cis-elements in the corresponding promoter.
