Abstract:
Objective To observe the effect of combined bovine amnion and rb-bFGF on second-degree burn wound.Methods Forty-three patients with small-to medium-area thermal burn were included in this study.The experimental area was1%-2%.The wound of the same nature in each patient was divided into three parts which were respectively treated with perforated bovine amnion(treatment group),bovine amnion(control group 1) and vaseline gauze dressing(control group 2) by combination of rb-bFGF. Results Compared with the control 1 and 2,the wound healing duration of second-degree deep burns in treatment group was significantly shortened(F=25.36;q=7.25,3.46;P0.01).For the superficial burns,the wound healing duration in the treatment group was shorten than control group 2(F=12.29,q=5.21,P0.01),but no significant difference was noted as compared with control group 1(q=2.60,P0.05). Conclusion The combination of perforated bovine amnion and rbbFGF can obviously promote the healing of burn wounds.Keywords:

To explore the optimal dosage and method of bFGF application on surgical wound for accelerating its healing.Laceration wounds on the extremities of 73 patients were served as the target. The wounds were applied with three different doses of bFGF (75 Au/cm(2), 150 Au/cm(2), 300 Au/cm(2)) and with three application methods, i.e. once a day, three times a day and once a day with moistening. Normal saline with 0.5% metronidazol was employed as control agents. The wound healing time in every groups was recorded in days and compared with each other.When compared to each other, the wound healing time (day) was much shorter (P < 0.05) with bFGF application in dose of 75 similar 300 Au/cm(2). There were also obvious differences in wound healing time among groups treated with the same dose but with different methods (P < 0.01). In addition, three times a day and once a day with moistening of bFGF application could further shorten the wound healing time.Compared to the dosage of the agent, the frequency and method of bFGF application seemed more important for its optimal effects. In view of the convenience, feasibility and economy, once a day with moistening was recommended for clinical application.

The wound healing process of the soft tissue aims to accelerate the closing of the wound by recurring, restoring the function, and minimizing the scar tissue. This process has to occur in the surgery process to obtain better-wound healing. The aim of this study was to know the stimulation effect of the wound dressing to accelerate the wound healing process of the soft tissue using Collagen (amino acid) and Amnion (proteinase inhibitors). This true experimental study was done to 36 mice (Sprague Dawley) that were divided into three different groups, two groups were the treatment groups and one group was the control group. Each group was adapted in Pathology Laboratory of Faculty of Veterinary Medicine Bogor Agricultural University. All of the mice were injured on the skin by incising the right side of the back for 2 cm in length and 4 mm in depth and then cleaned with 0.9% NaCl solution. To the first group of treatment, the wound was applicated by Amnion, and the second group was applicated by Collagen. Each 4 mice of all group were a termination by ether and encryption on the wounded tissue was conducted on day 3rd, 7th, and 14th. Assessment for a number of fibroblast and collagen synthesis on the wound of back tissue was conducted with the histological examination by painted using hematoxylin-eosin. The results concluded that Collagen application was influencing the mean of fibroblast higher than Amnion, and the lowest was controlled with statistically significant. The mean of collagen used Collagen lower than Amnion, and the lowest was control with statistically significant. It could be concluded that collagen application was preeminent in increasing the productivity of the fibroblast, as well as in developing forms of the collagen synthesis compared with Amnion and control.

Objective To observe the curative effect of recombinant bovine basic fibroblast growth factor (bFGF) on burn wound degree II . Methods A total of 920 cases of burn wound of superficial or deep degree II were treated with bFGF at a dosage of 144U/cm, and the healing rate as well as the time for complete healing ,were observed 8(superficial degree II )and 15(deep degree II )days after treatment. Results The healing rates of superficial degree II bum wound of trial and control groups 8 days after treatment were (77.6 ± 24.4) % and (51.7±26.4)% ,and the time for complete healing of them were 9.6 ±2.4 and 12.6 ±2.9 days respectively. However,the healing rates of deep degree II bum wound of trial and control groups 15 days after treatment were( 75.2 ±26.3) % and (53.3± 25.4) % , and the time for complete healing of them were 17.5± 4.5 and 21.9±5.5 days respectively.Conclusion bFGF promotes the healing of burn wound degree H significantly.

Objective To establish an experimental model of full thickness skin defect in minipigs and to investigate the effects of basic fibroblast growth factor (bFGF) on wound healing.Motheds After the skin on back of the minipigs was shaved, the cut skin in each side of spinal column was removed with a specific traumatoqenic apparatus (18?mm in diameter) to achieve a full thickness skin defect wound. The wounds were randomly divided into two groups (n=16): the wounds treated with bFGF served as the experimental group, while those not treated with bFGF as the control group. At the 3rd, 7th and 14th day after injury, the area of the wounds and wound volume were measured and the histological examination was performed to evaluate the wounds healing.Results Three and 7 days after injury, in the bFGF treated group, the area of wounds was decreased to (1.81±0.15)?cm 2 and (0.51±0.10)?cm 2 respectively, significantly less than in the blank control group (2.00±0.21)?cm 2 and (0.61±0.11)?cm 2 respectively) (P0.05);The wound volume in the bFGF treated group was significantly decreased to (0.49±0.07)?ml and (0.02±0.04)?ml respectively, significantly smaller than in the non treated group (0.61±0.17)?ml and (0.14±0.16)?ml respectively) (P0.05). Histological examination revealed that most of the wounds in the experimental group were re epithelialization at the 14th day, but in control group most of the wounds were not healed on day 14 after operation.Conclusion The wound model of full thickness skin defect in minipig holds good to study the effects of medicine for external use on wound healing. bFGF can promote the wound healing.

Objective To discuss the effect of cover allogeneic skin on severe damaged wound, accelerate wound bed preparation and wound’s healing. Methods Eighty-four emergency patients were selected who had severe damaged wound in skin and soft tissues during June 2005 to June 2011. According to the different treatments, the patients were divided into two groups. The former had 41 cases using allogeneic skin cover as treatment group and the latter had 43 cases using first stage skin grafting as control group. The success rate and healing rate of this two treatments were observed. Results After two regular debridement, group A were covered with acellular dermis matrix (ADM), group B were covered with the Saline gauze and the Vaseline oil sands. Every 4 cm × 4 cm size of the wound of each patient was selected as a viewing area. The growth of granulation tissue and clinical treatment time of two groups were compared after treated 1 d, 3 d, 7 d, 12 d. Results After treatment of 7 d, granulation wounds of group A (24.87±0.24) were significantly different compared with group B (18.66±0.45). The time of wound healing in group A was (16.17 ± 1.87) d, which was significantly shorter than group B (32.71±3.95) d, and the difference was highly statistically significant (P 0.01). Conclusion As a wound cover allogeneic skin, it can effectively protect the wound and promote wound vascularization and shorten the clinical treatment cycle.

Aim: To observe the effects of recombinant bovine basic fibroblast growth factor (rbFGF) on acute and chronic wounds healing. Methods: 1 282 cases with acute wounds such as burn, donor site or operative wounds and chronic wounds were treated with bFGF. Another 439 cases with the similar wounds were used as control. The complete healing time, healing rates and adverse reaction of wounds healing were observed. Results: In superficial Ⅱ degree burn wounds, the healing rate for 8 days post-burn and complete healing time was 51.7%±26.4% and (12.6±2.9)d (P0.001) respectively in control group and was 70.6%±25.0% and (10.5±2.4)d (P0.001) respectively in trial group. In deep Ⅱ degree burn wounds, the healing rate for 15 days post-burn and complete healing time was 53.3%±25.4% and (21.9±5.5)d (P0.001) respectively in control group and was 69.7%±27.0% and (18.5±4.4)d (P0.001) respectively in trial group. In operative wounds, the healing rate for 8 days and complete healing time was 57.6%±29.4% and (11.4±2.7)d (P0.001) respectively in control group, and was 72.1%±28.0% and (9.5±2.4)d (P0.001) respectively in trial group. In chronic wounds, the complete healing time was (28.0±18)d in control group and (22.1±16.0)d in trial group (P=0.068). Conclusion: Recombinant bovine basic fibroblast growth factor (rbFGF) can improve the quality and the velocity of wound healing significantly.

Objective To observe the clinical effect of collagen sponge on partial-thickness burn wound.Methods Clinical observation was performed on the same bodies,the same parts(including the antimere),the same-depth wound surfaces of inpatients with extent of burn 30% of total body surface area(TBSA),ranging in age from 16 to 64,averaging 30±12 years old,from April 2006 to October 2008.Collagen protein sponge was used on the observed wound surfaces,and 1% sulfadiazine silver gauze on control wounds.Results A total of 52 severely burned patients were observed,37 males,15 females,extent of burn from 1% to 30%,averaging(14.31±10.20)%.In superficial partial-thickness burn group,the 7-day healing rates of observed and control wounds(24 cases in each) were(76.07±9.84)%,(66.79±11.87)%,respectively,the completely healing time was(9.07±1.56) d,(11.93±1.64) d,respectively.In deep partial-thickness burn group,the 14-day healing rates were(69.44±9.06)%,(60.00±8.57)%,respectively,the completely healing rate were(18.33±1.57)d,(21.39±1.65)d,respectively.There was significant difference in above-mentioned healing indexes between observed and control wounds(P0.05),but there was not in red swelling of wound edge,discharge or bacterial culture rate.No abnormal changes were noted in blood or urine routine,liver or kidney function,nor systemic or local allergic reaction in the course of treatment.Conclusion Collagen sponge,which is of good promotion in the healing of patial-thickness burn wounds,is a safe and effective wound covering material.

OBJECTIVE: To observe the clinical effect of perforated bovine amnion combined with recombinant bovine basic fibroblast growth factor (rb-bFGF) on degree Ⅱ burn wounds.
METHODS: A total of 43 patients with small and medium-size thermal burn were collected, and the area of testing wound was 1%-2%. The wounds with the same nature were divided into three groups: perforated bovine amnion (treatment group), bovine amnion (control 1 group), and vaseline gauze dressing (control 2 group). All the three groups combined with rb-bFGF.
RESULTS: Compared with control 1 group (P<0.01) and control 2 group (P<0.05), the treatment group could obviously decrease the healing time of deep degree Ⅱ burn wounds. For superficial degree Ⅱ burn wounds, compared with the control 2 group, the treatment group could also decrease the healing time; however, there was no significant difference between treatment group and control 1 group (P>0.05). Dressing was not changed frequently, and the pain was relieved. Rash or other adverse effects were not detected in the three groups.
CONCLUSION: The combination of perforated bovine amnion and rb-bFGF can obviously promote the healing of burn wounds.

Objective: To study the curative effect and mechanism of bFGF(basic fibroblast growth factor) combined with Bashigao in treating Ⅱ° burned wound.Method: 168 cases of early stage Ⅱ° burned wound were randomly divided into cure group(bFGF combined with Bashigao) with 86 cases,and contrast group(paraffin oil) with 82 cases,according to the principle of comparability,observed the wound healing time.Result: The wound healing time of superficial Ⅱ° wound in the cure group was 8.0±0.8 days,and the contrast group was 12.5 ±1.6 days;the wound healing time of deep Ⅱ° wounds in the cure group were 16.5 ±1.75 days,and the contrast group was 25.5±1.85 days,There's difference significant statisticaly.Conclusion: bFGF combined with Bashigao can significantly shorten the healing time of Ⅱ° burn wounds.
