Abstract:
Objective To study the method of improving the consistency of the analytic results of RBC and WBC which between different SYSMEX UF-1000i automatic urine sediment analyzers.Methods To Use fresh and pure RBC and WBC urine sample to fine-tune laser optical index(S-FSC,S-SSC) of two SYSMEX UF-l000i urinary sediment analyzers,which had already calibrated by supporting calibrator,and compared and statistical analyzed with the results of RBC and WBC checked by two UF-l000i sediment analyzers before and after that fine-tuning.Results With a analyzer for the internal standard device,Before fine-tuning,the statistical bias of RBC and WBC were 16.12%and 16.65%,after fine-tuning the statistical bias of RBC and WBC were 8.16%and 2.44%,which decreased to 49.3%and 85.3%respectively;RBC slope(a) value decreased from 1.113 to 1.057,WBC decreased from 1.177 to 1.046 by correlation analysis of two UF-l000i urine sediment analyzers;and fine-tuning made no statistical difference between RBC and WBC internal quality control.Conclusion Calibrated by supporting calibrator, The resell of fresh urine RBC and WBC checked by different UF-1000 urine sediment analyzers may differ;By adjusting the SYSMEX UF-l000i laser optical index(S-FSC,S-SSC) can effectively improve the consistency of the results of RBC and WBC checked between different SYSMEX UF-l000i automatic urine sediment analyzers.Keywords:

Objective To evaluate the clinical application value of measurement parameters from Diana 5 fully automated blood cell analyzer.Methods According to criterion of preliminary evaluation of quantitative clinical laboratory methods(EP10 T2) of National Committee for Clinical Laboratory Standards(NCCLS), the complete blood count (CBC) and 5 part difference were measured by Diana 5 fully automated blood cell analyzer. Then the results were statistically analyzed using Microsoft Excel 2000 and SAS software. Results The within run CV was less than 2% for WBC, RBC and HGB in samples of high and middle value.They were less than 3% in low value,and the between run CV was 3% within them.The within run and between run CV were 2% for MCV, and those of PLT were 4.5% and 5.2%.The toal CV was 2.5% for WBC, RBC, HGB and MCV, and that of PLT was 5%.The linearity correlation coefficients of WBC, RBC, HGB, HCT and PLT were 0.999 , 0.999, 0.999, 0.998 and 0.995,respectively.The bias of PLT was 3%,but the detection values of WBC, RBC, HGB and HCT were near to true value of quality control samples.The carryover of WBC was 1%, and those of RBC and PLT were 1%, and that of HGB and MCV was 0. Compared with by those microscope examination,correlation coefficients of Neu, Lym, Mon, Eos and Bas were 0.982, 0.956 , 0.842, 0.536 and 0.328, respectively.Conclusion The results of CBC by Diana 5 blood cell analyzer possess good linearity,and high preciseness and accuracy,and the carryover is lower.The results of 5 part difference are precise and accurate, suitable for screening abnormal blood cell.

[Objective]To valucate the performance of the method used in detecting red blood cells(RBC) in urine by Sysmex UF-100 automatic flow urine Analysis(UF-100).[Methods]Collecting 750 cases of clinical urine samples under the requirement of NCCLS Literature GP16-A,analysis by above method.[Results]It has a favorable relativity in detecting of urine RBC by UF-100 and Neubauer’s Counting Board.RBC γ=0.973,t examination(P0.05);Good linearity in range of 5 000 RBC/μl and high degree.CV6% when the number of RBC is samller than 40 RBC/μl.The bacteria and candida、have different effects in RBC detection in urine.[Conclusions]UF-100 is a fast and easy testing methond to perform urine sediments quantitative erinalysis.We sugest to choose the Sysmex UF-100 as sieving rule testing RBCs methond.

Objective The aim of this study was to evaluate the consistency and accuracy of all the parameters of the urine samples detected by two automated urine sediment analyzers from Sysmex Corporation. Methods Two automated analyzers and manual microscopy examined 1,059 urine samples. The sensitivity, specificity, positive predictive value, and negative predictive value were evaluated. The consistency of all the parameters was tested. The influencing factors of false positive and false negative samples were analyzed and compared. Results All the parameters had good specificity, negative predictive value, and coincidence rate (83.95%-99.61%). The RBC, WBC, and X'TAL analyzed by UF-5000 and UF-1000i exhibited good agreement (Kappa=0.597-0.784) with those by manual microscopy. The overall concordance rates of RBC and WBC were good (RBC: r=0.9842, CCC=0.9693; WBC: r=0.9955, CCC=0.9711). Among the influencing factors, mucus filament accounted for a large proportion, which mainly affected the detection of CAST. Concurrently, the false-positive factors of EC detection were reduced, and CAST did not affect the detection of EC. Conclusion The parameters of the two instruments tested have shown high accuracy, consistency, coincidence rate, and low negative predictive value for RBC and WBC, which has ensured that UF-5000 and UF-1000i meet the clinical requirements for urine tests for disease screening. For the samples with poor consistency and false-positive factors, a conventional microscopic examination should be applied to verify the accuracy of the instrument detection.

Objective To study the accuracy in detecting white blood cell (WBC) by using Diasys R/S2003 urinary sediment workstation and UF-100 urinary sediment automatic analyzer. Methods WBC weretested in 1080 urinary samples by using Diasys R/S2003 urinary sediment workstation and UF-100 urinarysediment automatic analyzer separately. Data were analyzed to get positive rate and it's influencefactors. Results The positive rates of WBC detected by two methods were 37.6% and 45.9% separately.There was significant difference between them (P0.05). The false positive rate was 8.3% by using UF-100 urinary sediment automatic analyzer. Conclusions The accuracy of detecting WBC in urine couldincrease by applying two combined methods or using automatic analyzer to assist microscope examination.

目的 评价UF-100尿沉渣分析仪与DiaSys(R/S2003)尿沉渣定量分析工作站两种方法检测尿液中红细胞(RBC)、白细胞(WBC)的可比性.方法 用UF-100尿沉渣分析仪与DiaSys(R/S2003)尿沉渣定量分析工作站检测本院287例住院病人晨尿标本,对红细胞和白细胞检测结果进行比较.结果 不同人员采用同一方法检测相同标本的RBC、WBC结果均有可比性,但两种方法检测RBC、WBC的阳性率差异均有显著性(均为P<0.001).结论 UF-100尿沉渣分析仪可起到筛检作用,UF-100尿沉渣分析仪与DiaSys(R/S2003)尿沉渣定量分析工作站联合使用,可使尿沉渣分析真正做到标准化、自动化。

Objective To investigate the clinical application values of Sysmex UF-1000i automated urinary sediment analyzer.Methods 2 000 urine samples were determined by UF-1000i automated urinary sediment analyzer and microscopy.The results were contrasted,and the reasons of false positive in UF-1000i analyzer were analyzed.Results When using Sysmex UF-1000i analyzer,the detecting sensitivities of white blood cell(WBC),red blood cell(RBC),cast and small round epithelial cell were 99.2%,99.3%,94.7% and 95.1%,and the false positive rates of them were 5.8%,6.1%,10.4% and 4.6%,respectively.Many factors,like viscose rayon,crystals,bacteria,yeast-like fungus,small round epithelial cells,fat droplets and lecithin body may affect the determination of urine formed elements.Conclusions It is an effectual method to detect urinary sediments by Sysmex UF-1000i analyzer.It is very important to observe sediments by microscopy when all above interferences are positive or increase.

Objective To evaluate the function of Sysmex UF-100 fully automated Urine sediment analyzer in detection of red blood cell(RBC) and leukocyte(WBC).Methods To evaluate precision,linearity,carryover rate of the UF-100.select RBC,WBC samples in the range of 2~100/μl,100~500/μl, 500/μl,35 cases in each group,the samples were tested by the Neubauer Count Board and the UF-100 count method.The results of the two methods were compared.Results The precision of the RBC and WBC was 3.8% and 5.6% respectively,linear(r) were 0.997 and 0.999 respectively,the pollution carryover rate of 0.14% and 0.17%;Compare the results of the UF-100 and the Neubauer blood cell count board : There were no significant difference in RBC 500/μl(P 0.05),There were significant difference in RBC 500/μl(P 0.05);WBC,no significant difference in the range of 2~100/μl(P0.05), 100/μl were significant difference(P0.05).Conclusions Use the UF-100 fully automated urine sediment analyzer to test RBC and WBC,its precision,linearity were well,and its carryover rate were low but it should not completely replace the microscopic examination,the UF-100t,he Dry chemistry and microscopic examination can greatly improve the efficiency of detection.

Objective:To investigate the clinical significance of RBC-Information by UF-100 urinary sediment analyzer. Methods Using UF-100 urinary sediment analyzer,DIRUIH-300 urine dipstick and the microscopic analysis of urine sedi- ment studied 880 freshly collected urine samples and compared RBC fluorescent parameters.Results Erythrocyte proportion of measured value of 56.36% and RBCInfo of 53.02% with UF-100 automated urinalysis analyzer.Normocytic rate of 28.52% with Coincidence rate of 100%;Microcytic rate of 16.35% with Coincidence rate of 97.67%;Non-classifide rate of 55. 13% with Coincidence rate of 46.21% .Conclusion Combination detection is a reliable method to identify haematuria and mi- croscopic counting with a standardized cell chamber system is useful.

Objective To compare the performances of UF-1000i urine sediment analyzer,dry chemistry method and manual microscopy in the determinations of red blood cell(RBC) and white blood cell(WBC).Methods 996 urine samples were collected.The RBC and WBC were determined by UF-1000i urine sediment analyzer,dry chemistry method and manual microscopy.The results were compared and analyzed statistically.Results With the positive cut-off value of 25 per μL for RBC and 30 per μL for WBC,the complete concordances were 78.4% for RBC and 67.2% for WBC.The concordances defined by the grade between the 2 analyzers were found to be 85.6% for RBC and 86.5% for WBC.The false positive rates of the UF-1000i were 4.62% for RBC and 1.81% for WBC.Conclusions These 3 urinalysis methods show good concordance with each other in urine sediment examination.The UF-1000i is more accuracy and anti-interferential than UF-100,but manual microscopy is still necessary.

Objective To calibrate hematology analyzers with healthy fresh blood to make their parameters comparable and traceable.Methods Hematology analyzer Sysmex XE-2100 was adopted as the reference system to measure fresh blood samples collected from healthy subjects.The detected results of white blood cell(WBC),red blood cell(RBC),hemoglobin(Hb),hematocrit(HCT) and platelet(PLT) were used to calibrate hematology analyzer Sysmex XS-1000i,and comparison studies were performed between two analyzers before and after calibration.Results Before Sysmex XS-1000i was calibrated,the deviation arising in WBC counting displayed on Sysmex XS-1000i was beyond the limits;after it was calibrated,the deviations of all the parameters shown on Sysmex XS-1000i were within the limits.Conclusions Parameters of hematology analyzers calibrated with fresh blood are comparable and traceable.
