    Abstract:
    Objective:Different concentration growth regulating substances of 6-BA,NAA,2,4-D and PP 333 on pollen germination and pollen tube growth were studied with pyrus pyrifoliaWhangkeumbae.Method:Pollen cultivating liquid medium were confected,with different mass concentrations of 6-BA,NAA,2,4-D and PP 333.To research the effects of different concentrations plant growth regulating substances on pollen germination and pollen tube growth.Result:Results showed that the low concentration of 6-BA and 2,4-D stimulated the pollen germination and pollen tube growth,but when the concentration up to certain value,pollen growth and pollen tube growth were inhibited.NAA and PP333 had inhibition on pollen germination and pollen tube growth.Conclusion:10 mg /L 6-BA and 5 mg /L 2,4-D stimulated the pollen germination and pollen tube growth.
    [Objective] The causes and mechanism for Davidia involucrata endangered were revealed so as to provide the reference for its pollen preservation and cross breeding.[Method]The pollen germination and pollen tube growth of Davidia involucrata were tested with one way ANOVA and multiple-comparison.[Results] The results indicated that the pollen germination and pollen tube growth would be enhanced when the concentration of sucrose,boric scid,agar in the culture medium were within a certain range.If the concentrations were too high or low,the pollen germination and pollen tube growth would be inhibited.The effect of IAA on pollen germination and pollen tube growth was in significant.The optimum temperature affecting pollen germination and pollen tube was from 20 to 25 ℃.Humidity had big effect on pollen fertility.When atmosphere was dry,its germination ability was reduced.But when atmosphere was too humid,the pollen drank too much water and broke,and its germination ability was reduced,too.[Conclusion] Davidia involucrata pollen germination and pollen tube growth varied with the change of medium component and culture conditions and its pollen germination and pollen tube growth required not only an appropriate concentration of the components,but also had the suitable culture condition.

    The results of studying the effects of several plant growth regulating substances on pollen germination and tube growth in pear (Pyrus serotina Rhed.) by cultivating pollens in liquid media showed that GA3,TRIA,IAA and 2,4D stimulated pollen germination and tube growth when pollen was cultivated in their lower concentration media.When they were in higher concentrations,they would inhibit pollen germination and tube growth.The most suitable concentrations that stimulated pollen germination and tube growth were 50~300 mg/L,3~100 mg/L,5~25 mg/L and 5~10 mg/L,respectively,for these four kinds of plant growth regulating substances.NAA inhibited pollen germination and tube growth.The inhibitory effect became stronger with its increasing concentration.PP333 and ABA had some inhibitory effect on pollen germination.Although they had some stimulating effect on pollen tube growth when they were in their lower concentrations,in higher concentrations they still inhibited pollen germination.The inhibitory effect also became stronger with the increasing concentration.The best concentrations for pollen germination were 400 mg/L and 60 mg/L for PP333 and ABA respectively.When they were above these concentrations,the stimulating effect also got worse;they even inhibit pollen tube growth.For instance,when PP333 was at 1 000 mg/L,it greatly inhibited pollen tube growth.

    The effects of five plant growth regulating substances on pollen germination and tube growth were studied by using in vitro pollen culture in Jinguang plum. The results showed that gibberellin, 6 -BA and 2,4 -D stimulated pollen germination and tube growth when they were in their lower concentrations, but they would inhibit pollen germination and tube growth when they were in their higher concentrations. The most suitable concentrations that stimulate pollen germination and tube growth were gibberellin 25 ~100 mg/L, 6-BA 12.5~25 mg/L and 2,4-D 0.5~5 mg/L. NAA and paclobutrazol inhibited pollen germination, and the inhibitory effect became stronger with the concentration increasing. But it had stimulating effect on pollen tube growth when the concentration of paclobutrazol was 0~50 mg/L.

    The effect of four types of plant growth regulating substances on pollen germination and pollen tube growth of different Catalpa bungei provenances was studied by in-vitro pollen cultivation in liquid media. The results showed that low concentration of GA3stimulated pollen germination and pollen tube growth,whereas the high concentration of GA3had inhibition effect. The most suitable concentration to stimulate the pollen germination and pollen tube growth was 300 mg / L. The effect of IAA and 2,4-D varied with the C. bungei provenances,but the general law was that the low concentrations could promote but the high concentrations would inhibit the pollen germination and pollen tube growth. The maximum pollen germination rate was obtained by 15 mg / L IAA concentration,there was significant difference in the pollen germination rate among two provenances and the control. Regarding to 2,4-D,the maximum pollen germination rate was obtained by 200 mg / L concentration. When the concentrations of both IAA and 2,4-D were over the most suitable values,these two types of plant growth regulators would inhibit the pollen germination and pollen tube growth. NAA had an inhibition effect on the pollen germination and pollen tube growth of C. bungei at different concentrations,the higher NAA concentration was,the sronger the inhibition effect would be.

    The study examined pollen germinations and pollen tube growth of Prunus mume Sieb.Let Zucc.cultured with different medium components at different pollen densities and different temperatures and different times.It was shown that in Xiyeqing,the optimal medium of pollen germination and pollen tube growth was 30 mmol/L MES buffer(pH 6.5) containing 20% sucrose,0.01% H_3BO_3,20% PEG-4000,(0.03)% Ca(NO_3)_2 ·4H_2O and 0.02% MgSO_4·7H_2O;on the medium,the germination rate reached(45.03)% and the pollen tube length reached 597.2 μm;and the optimal pollen density of pollen germination and pollen tube growth was 20~80 pollens/mL.In Prunus mume too high and too low a culture temperature were not favorable for pollen germination and pollen tube growth and optimal pollen germination and optimal pollen tube growth appeared at 25℃.The average pollen germinations of the tree varieties,Xiyeqing,Gessekai and Oushuku was 48.6% and their average pollen tube length was 762.3 μm.Cultured at different temperatures Prunus mume pollens germinated differently,and cultured at 25℃ the pollen tubes grew most quickly,mainly within 0 ~12 hours,and the average pollen tube growth rate of the tree varieties was 58.5 μm/h.

    The effects of medium components and culture conditions on pollen germination and tube growth in peach by cultivating pollens in medium were studied.Life span of pollen was also studied during the storage in different temperature ovens.It showed that:there was no difference between solid and liquid culture medium for pollen germination rate and pollen tube length;The medium with 10% sucrose was the optimum medium for pollen germination and tube growth for most peach varieties,while the boron had a positive effect on the pollen germination rate but no effect on the pollen tube length;20℃~25℃ was the optimum temperature for pollen germination and pollen tube growth;In the first 3 h stage of cultivation,the pollen germinate and growth quickly,in the second stage 3~5 h,the increase trend slowed down,and after 5 h,the increase trend to stop;The pollen activity was decreased with the store temperature and time increase.

    The effects of calcium(Ca2+),zinc(Zn2+) and molybdenum(Mo6+) on pollen germination and pollen tube growth of Paris polyphylla var.stenophylla were investigated through tissue culture and analysis of variance.The results indicated that Ca2+ inhibited the pollen germination and pollen tube growth.When the concentration of Zn2+ was high,the pollen germination and pollen tube growth would be inhibited.If the concentration of Zn2+ was low,it had no influence on the pollen germination and the growth of pollen tube.When the concentration of Zn2+ was within a certain range,the pollen tube growth would be enhanced.When the concentration of Mo6+ was high,the pollen germination and the growth of pollen tube was inhibited.If the concentration of Mo6+ was low,it had no influence on the pollen germination and the growth of pollen tube.
